HMC毒素对同核异质玉米叶肉细胞原生质体的影响

摘    要:制备并纯化了玉米小斑病菌C小种致病毒素(HMC),利用根冠细胞进行毒素活性检测,结果表明其对玉米C型不育细胞质具有专化活性.同时提取了同核异质玉米C103叶肉细胞原生质体,并用纯化后不同质量浓度的HMC毒素对同核异质C103叶肉细胞原生质体进行处理,光学显微镜下观察并统计HMC毒素对同核异质玉米叶肉细胞原生质体的影响,从而对玉米小斑病菌C小种的致病性进行研究.研究结果表明,HMC毒素可以导致C103玉米C型不育细胞质叶肉细胞原生质体质膜破裂,细胞内容物泄漏.

Abstract:A toxin preparation from Bipolaris(Helminthosporium) maydis Race C(HMC-toxin) with apparently biological activities was special active against root cap cells from Charrua male-sterile(C) cytoplasm corn.Protoplasts from mesophyll cells of Charrua male-sterile(C) cytoplasm corn were extracted and treated with specific toxin so as to study the mechanism of Bipolaris(Helminthosporium) maydis Race C.The microscocpe observation showed that the protoplasts were destroyed following the cytoplasts leaking out.The statistics indicated that HMC toxin was specific to destroy the protoplasts of the Charrua male-sterile(C) cytoplasm.
