摘    要:以体积分数50%的甲醇溶液为溶剂,用超声提取法从开口箭药材中提取绿原酸,用高效液相色谱法检测提取液中绿原酸含量。提取条件为:超声功率240 W,超声频率40 kHz,提取温度60℃,超声提取30 min。色谱条件为:色谱柱为Inertsil ODS-3C18(4.6 mm×150 mm,5μm)柱,以乙腈-0.4%磷酸水溶液(体积比9∶91)为流动相,流速1.0 mL/min,检测波长327 nm,柱温30℃。绿原酸在0.2553.06μg/mL范围内与峰面积呈现良好的线性关系,y=31 388x+152.43(R2=0.999 9),平均加样回收率99.4%,RSD为2.12%(n=3)。检测结果表明,不同产地开口箭药材中绿原酸的含量有一定的差异,其变化范围在32.843.06μg/mL范围内与峰面积呈现良好的线性关系,y=31 388x+152.43(R2=0.999 9),平均加样回收率99.4%,RSD为2.12%(n=3)。检测结果表明,不同产地开口箭药材中绿原酸的含量有一定的差异,其变化范围在32.846.62μg/g。该方法简便、准确、重复性好,可用于开口箭药材的质量控制。

Abstract:The chlorogenic acid is extracted from Tupistra chinensis using 50%methanol , and the conditions of the ultrasonic wave are as follows: extraction ultrasonic power of 240 W, ultrasonic frequency of 40 kHz,temperature of 60 ℃ and ultrasonic treatment time of 30 min.Samples are analyzed on an Inertsil ODS-3C18 column(4.6 mm ×150 mm, 5 μm) using acetonitrile-0.4%phosphoric acid as mobile phase , and speed of flow is 1.0 mL/min, the examination wave length is 327 nm and the column temperature is 30 ℃. The chlorogenic acid shows a good linearity in the range of 0.255-3.06 μg/mL, egression equation being y =31 388x +152.43(R^2 =0.999 9) and the average recovery being 99.4%(RSD=2.12%).The result shows that there are some differences in the content of chlorogenic acid in Tupistra chinensis among different origins .The established method is simple , credible , accurate , repeatable and can be used for the quality control of Tupistra chinensis.
