细胞ATP检测试剂的优化及其灵敏度的测定

为配制细胞ATP检测试剂并检测其灵敏度,按照常规报告基因分析试剂配制方法,将其中主要成分Luciferase(荧光素酶)和Luciferin(荧光素)及ATP浓度进行倍比稀释后找到最适合反应的最佳浓度值;按照最佳浓度配制反应液,将ATP浓度倍比稀释测定报告基因数值并绘制标准曲线,分析检测灵敏度;另外选取肿瘤细胞株H1299,倍比稀释后裂解细胞应用配制好的反应液进行ATP检测,分析最低检测细胞数。结果:Luciferase和Luciferin最佳反应浓度分别为1.25μg/mL和87.5μmol;按照最佳浓度配制反应液后,绘制ATP浓度标准曲线,找到ATP检测最低线为10-12mol/L,可以检测到H1299细胞最低值为100个。说明本实验配制的细胞ATP检测试剂灵敏度高具有良好的应用价值。
