国产无机三氧化物聚合物的免疫相容性研究

摘    要:目的 以细胞因子(肿瘤坏死因子α和白细胞介素1β)为研究对象,对国产无机三氧化物聚合物(MTA)介导的炎症反应进行相关的免疫学评价.方法 运用双抗夹心ELISA检测新材料国产MTA引起体外小鼠的单核巨噬细胞系RAW264.7细胞TNF-a和IL-1β的变化.结果 未经脂多糖诱导的RAW264.7细胞与阳性材料接触后,TNF-α和IL-1β的表达均高于阴性材料,差异均有显著性意义(P〈0.05);国产MTA组和进口MTA组TNF-α和lL-1β的表达与阴性材料比较差异无显著性意义(P〉0.05);经脂多糖诱导的RAW264.7细胞与阳性材料、国产MTA、进口MTA接触后,TNF-a和IL-1β的表达均高于阴性材料,差异均有显著性差异(P〈0.01).结论 国产MTA具有良好的生物相容性.

Abstract:Objective To conduct an immunological evaluation on the inflammatory reaction mediated by the domestic mineral trioxide aggregate (MTA) ,with tumor necrosis factor α (TNF-α) and interleukin 1β(IL-1β) as indices. Methods The sandwich ELISA is used to determinate the TNF-α and IL-1β in the macrophages reacted with the domestic MTA. Results Macrophages stimulated with positive materials produce extremely significant more TNF-α and IL-1β compared to negative materials ( P 〈 0. 05 ). There are no significant differences in TNF-α and IL-1β between the domestic MTA, imported MTA and negative materials ( P 〉 0. 05 ) ; Stimulated with positive materials, the domestic and imported MTA the lipopolysaccharide?induced macrophages produce more TNF-α and IL-1β compared to negative materials, with extremely significant differences (P 〈 0. 01 ). Conclusion Domestic MTA shows good biocompatibility.
