西北寒燥证证候动物模型的生物学基础研究

摘    要:为揭示西北干燥寒冷环境对机体产生影响的生物学基础,选用ICR小鼠,根据中医病因学说理论,用人工气候箱模拟新疆这种特有的干燥寒冷环境,并作用于小鼠,建立寒燥证证候模型,以正常喂养为空白对照组.试验处理第21天后,用摘眼球取血法和解剖法分别取出小鼠的血液和各种脏器,在HE染色光镜下观察其组织病理学改变.用透射电镜观察其细胞超微结构的改变,并用高效液相色谱-电化学检测法测定小鼠脑组织和外周血中单胺类神经递质的变化.和空白对照组相比,组织形态学方面的改变主要是中心血管的轻度扩张和淋巴细胞增多;超微结构方面主要以水肿表现为主;单胺类神经递质方面,外周血中的去甲肾上腺素(NE)、多巴胺(DA)明显低于对照组(P<0.01),5-羟色胺(5-HT)高于对照组(P<0.05),而脑中去甲肾上腺素、多巴胺和5-羟色胺含量均显著高于对照组(P<0.01).寒燥环境可导致小鼠体内"寒燥之气"的产生,由此推论寒燥环境是西北寒燥证形成的主要原因;且西北寒燥证不是机体某一脏器的改变,而是整体性、全身性、系统性的改变.
