摘    要:目的 观察五味子粗多糖与泽泻提取物联合应用对酒精诱导小鼠急性肝损伤的保护作用.方法 50只小鼠随机分成5组,分别为空白对照组(CON)、酒精性肝损伤模型组(MOD)、五味子泽泻2:1组、五味子泽泻1:1组和五味子泽泻1:2组.各药物处理组小鼠每日灌胃五味子泽泻提取物100mg/kg,空白对照组和酒精性肝损伤模型组给予相同体积溶媒,连续30d.酒精性肝损伤模型组和五味子泽泻各给药组的小鼠末次给药1h后均灌胃给予50%乙醇溶液12mL/kg.16h后取小鼠的血和肝脏,计算各组小鼠的肝指数,检测血清中谷草转氨酶(AST)和谷丙转氨酶(ALT)的水平,检测肝组织中微量还原型谷胱甘肽(GSH)和甘油三酯(TG)的含量,应用苏木精-伊红染色(HE)法观察肝脏病理学变化.结果 与酒精性肝损伤模型组比较,五味子泽泻1:1组和1:2组均可降低小鼠肝脏指数(P<0.05),降低血清中ALT和AST水平(P<0.05),升高肝组织中GSH含量(P<0.01),且降低TG含量(P<0.05或P<0.01),改善肝脏病理学变化.结论 五味子多糖与泽泻提取物联合应用对小鼠急性酒精性肝损伤具有一定的保护作用.

Abstract:Objective To observe the protective effects of Schisandra polysaccharides and Alisma orientalis extract on the acute alcohol-induced liver injury in mice.Method 50 Mice were randomly divided into 5 groups,control group,alcoholic liver injury model group,Schisandra and Alisma (2:1) treatment group,Schis-andra and Alisma (1:1) treatment group,and Schisandra and Alisma (1:2) treatment group.The mice in Schisandra and Alisma treated groups were intragastrically administered with Schisandra polysaccharides and Alisma orientalis extract (100 mg/kg),and those in the control and model group were given the same volume solvent for 30 days in the same way.1 Hour after the last administration,50% ethanol (12 mL/kg) was administered to the mice in model group and Schisandra and Alisma treated groups.16 Hours later,blood and liver samples of mice were collected for the calculation of liver indexes of mice in each group and the detection of aspartate aminotransferase (AST) and alanine aminotransferase (ALT) levels in serum as well as the contents of reduced glutathione (GSH) and triglyceride (TG) in liver tissues.Pathological changes of the mice liver were observed by hematoxylin-eosin staining (HE).Results Compared with the alcohol-induced liver injury model,Schisandra and Alisma extract could significantly reduce the liver index in mice (P<0.05) and the levels of ALT and AST in serum (P<0.05),significantly elevate the content of GSH (P<0.01),significantly reduce the content of TG in liver tissues (P<0.05),and significantly improve the pathological changes of the liver tissues.Conclusion Application of Schisandra polysaccharides and Alisma orientalis extract should have a protective effect on the acute alcohol-induced liver injury in mice.
