贻贝多糖的结构修饰及其体外抗肿瘤活性测定

摘    要:采用三氧化硫吡啶法对厚壳贻贝多糖(MT1)结构进行硫酸化修饰,并对修饰后多糖进行了体外抗肿瘤活性测定。在样品:三氧化硫吡啶=1:6,温度为90℃条件下反应6 h,实验结果表明:硫酸化多糖(MTS)得率为34%,且修饰后硫酸根含量从6.51%提升到32.95%。体外抗肿瘤活性测定结果显示:浓度为8 mg/m L的MT1、MTS对前列腺癌DU-145细胞作用72 h后的抑制率分别为79.35%、85.04%,且IC50为2.04 mg/m L,1.72 mg/m L;而对人肺癌H1299细胞作用72 h后的抑制率分别为:81.28%、85.32%,以及IC50分别为2.46 mg/m L,1.71 mg/m L。结论:在该条件下修饰后的硫酸化贻贝多糖能提高抗肿瘤活性。

Structure Modification and Research on Antitumor Activity of Mussel Polysaccharide

Abstract:Mussel polysaccharide structure wasmodified bysulphur trioxide pyridine, and antitumor activity of Sulfated polysaccharides was investigated in vitro. The condition of Sulfuric acid esterification experiment was that:the quality proportion of sample and sulphur trioxide pyridine was 1:6, reaction temperature was 90℃and reaction time was 6h.It showed that:the yield of sulfated polysaccharide(MTS) was 34%, and the content of SO42-was improved from 6.51%to 32.95%. Antitumor activity test in vitro showed that:inhibitory effect of MT1, MTS on DU-145 was 79.35%,85.04%, and IC50 was 2.04mg/mL,1.72mg/mL. The inhibitory effectno H1299 was 81.28%, 85.32% and IC50 was 2.46 mg/mL, 1.71mg/mL. Conclusion: Sulfated polysaccharide can improve the antitumor activity.
