| 过氧化氢诱导前列腺癌PC3细胞HSP60表达和Akt蛋白磷酸化的研究 |

| 摘 要： | 以不同浓度H_2O_2刺激PC3细胞24h,应用MTT法分析细胞活力变化,并采用蛋白印迹方法检测HSP60蛋白、Akt蛋白(总Akt蛋白)和磷酸化Akt蛋白的表达水平.结果表明,15.625μmol/L的H2O2可诱导PC3细胞内HSP60表达显著升高(p0.05),7.312μmol/L和15.625μmol/L的H_2O_2均会导致磷酸化Akt蛋白的量显著下降(p0.01),但该两种浓度的刺激对细胞内总Akt蛋白量并无显著影响(p0.05).

Research of Hydrogen Peroxide Induced Up-regulation of HSP60 and Down-regulation of Phosphorylated Akt in PC3 Cells |

| Abstract: | PC3 cells were stimulated with H_2O_2 in different concentrations for 24 hours,the viability of PC3 cells were measured by MTT assay.Western blotting was used to detect HSP60 protein levels,p-Akt and total Akt protein levels as well.The results of the study confirmed that higher concentrations of H_2O_2(15.625μmol/L)could significantly induce up-regulation of the HSP60e xpression(p<0.05),it also results down-regulation of the p-Akt protein expressing at both 7.312μmol/L and 15.625μmol/L concentration of H_2O_2(p<0.01),but the stimulation of the two concentrations of the H_2O_2 could not change the total amount of Akt protein levels(p>0.05). |
