摘    要:基因簇的异源表达在放线菌次级代谢产物的发现、产量优化、生物合成途径的解析以及非天然代谢产物合成途径的构建中得到了广泛的应用,成为主要策略之一。近年来,随着高通量测序技术以及基因编辑技术的快速发展,大量的新颖次级代谢产物合成基因簇被发现,异源表达在研究放线菌次级代谢产物中起到越来越关键的作用。基于此,回顾了异源表达技术在次级代谢产物研究中的关键作用,并对其潜在的应用前景进行了展望,旨在为基因簇异源表达技术更为广泛的应用提供参考。

Advances in Research on Heterologous Expression System of Secondary Metabolite Synthesis Gene Clusters of Actinomycetes

Abstract:Heterologous expression of gene clusters has been widely used in the discovery of secondary metabolites of actinomycetes, the yield optimization, the analysis of biosynthetic pathways, and the construction of non-natural metabolite synthesis pathways.It has become one of the main strategies. In recent years, with the rapid development of high-throughput sequencing technology and gene editing technology, a large number of novel secondary metabolite synthesis gene clusters have been discovered.The heterologous expression plays an increasingly important role in studying secondary metabolites of actinomycetes. Based on this, the key role of heterologous expression technology in the study of secondary metabolites was reviewed, and its potential application prospects were prospected. The aim was to provide reference for the wider application of gene cluster heterologous expression technology.
