白芍总苷对NAFLD大鼠肝脏NF—kB/p65蛋白表达及羟脯氨酸含量的影响

摘    要:目的:观察白芍总苷(TGP)对高脂饮食诱导的非酒精性脂肪肝NAFLD大鼠肝组织核转录因子-κB(NF-κB)和羟脯氨酸(Hyp)及血清血清谷草转氨酶(AsT)、谷丙转氨酶(ALT)、还原型谷胱甘肽(GSH)和白介素1β(IL-1β)、肿瘤坏死因子α(TNF—α)和白介素-6(IL-6)水平的影响.方法:结果:TGP200mg·kg-1组能明显降低NAFLD模型组大鼠血清ALT和AST水平和提高GSH含量(P〈0.01)、能明显抑制肝组织中k的阳性表达及Hyp的生成(P〈0.01)、使血清IL-1B、TNF-a和IL-6水平明显降低(P〈0.01)、使肝脏的病理学改变明显减轻.TGP100mg·kg-1对上述指标无明显改变.结论:TGP能明显改善NAFLD大鼠的肝功能及病理性变化,其机制可能是通过下调NF-κB/p65的表达、降低血清IL-1β、TNF-n和IL-6水平、抑制肝脏Hyp的产生,从而对NAFLD大鼠发挥治疗作用.

Abstract:Objective:To investigate the influence of total glucosides of paeony on NF - kB/p65 pro- tein expression , the content of hydroxyproine (Typ).serum alanine aminotransferase(ALT), aspartate aminotransferase (AST) , glutathione (GSH) and Interleukin 1β(IL-1β), tumor necrosis factor alpha (TNF- α) and interleukin -6 (IL-6) in nonalcoholic fatty liver disease rat induced by high fat diet. Meth- ods. the NAFLD rat model was established by high fat diet. All rats were randomly divided into 4 groups, normal group, model group, TGP (100 mg · kg-1 .200 mg · kg-1)group, at the end of four weeks, the rats were killed by 3% barbiturates, taking blood from heart, the level of serum AST and ALT was determinated by Henry mothed, the content of GSH was determinated by colorimetric meth- od,the level of IL-1β.TNF-αand IL- 6 was tested by ELISA, extracting liver for HE staining to observe hepatocyte pathological changes, NF-kB/p65 protein expression in the liver tissue was detected by im- munohistochemistry, the content of Hyp was detected by chloroform -T method. Results: TGP(200 nag · kg-1)could significantly reduce the level of serum AST and ALT , and improve the level of GSH in NAFLD rats(P〈0.01), TGP(200 mg · kg-1)could significantly inhibt the expression of NF-kB/p65 and the formation of Hyp in liver(P〈0.01), and reduc the contents of serum IL-1β.TNF-α and IL-6(P d0.01), TGP(200 mg · kg-1)could significantly improve the histopathological changes of liver. TGP (100 mg · kg-1) had no significant effect on the above indicators. Conclusion. TGP(200 mg · kg-1) could significantly improve the liver function and histopathological changes in NAFLD rats, the mecha- nism might be related to down regulating the expression of NF-kB/p65,reducing the level of serum IL- 1β.TNF-α and IL-6,inhinting the formation of Hyp in liver, and to play a role in the treatment of NAFLD rats.
