用昆虫谷氨酸电位检测磷酰胺酯化合物的活性

摘    要:采用电生理细胞内微电极记录果蝇3龄幼虫神经-肌肉接点的兴奋性接点电位(excitatory junctional poten-tials,EJP),观察加药前后EJP幅值的 变化以及电位是否被阻断及阻断时间,实验结果初步表明:18种磷酰胺酯BPON系列均可阻断EJP,但它们的阻断时间相关很大,从7.25min到43.00min。这些化合物对谷氨酸电位的作用可分为三种:1.电位逐渐被阻断;2.电位先

Abstract:Most of insecticides are neurotoxins. They can suppress sodium channels of nerve membrane or acetylcholine receptor channels of synapses. When one type of insecticides are applied too long, the insects will produce insensibility to it. So, it is important to seek other insecticides for delaying insensibility of the isnects. As it is known, the glutamate receptor channels exist in the neuromuscular junction and the muscle cell of the insects. In early days, our research work showed that the phosphorodiamidoates compounds could take effect on glutamate receptor. The work reported in this paper investigates the effect of the 18 kinds new phosphorodiamidoates compounds (BPONs) on the glutamate potential. The experiments mere done on third instar larvae of the fruit fly Canton S strain. The excitatory junctional potentials (EJP) of the instar neuromuscular junction was induced by a glass microelectrode. The experiments showed that the 18 kinds of BPONs all could block EJP of glutamate. The blocking durations of each of the BPONs were different from 7.25 to 43.00min . So, the blocking action of BPONs compounds could be divided into three types:1.EJP was blocked progressively; 2. EJP was blocked after a short exciting period; 3. EJP was blocked rapidly. The effect of overall toxicity showed that the death rate of fruit fly in the second type is the highest.
