| 摘 要： | 探讨血清饥饿法对人表皮细胞HacaT细胞周期的影响．采用无血清和低血清浓度的DMEM培养基饥饿HaCaT细胞，分别培养6h，12h，22h时，用倒置显微镜观察细胞形态，并且收集各组细胞，用流式细胞仪分析细胞周期各个时相细胞百分比．结果发现HaCaT细胞系在含0．2％FBS的DMEM培养基中培养12h，细胞仍然贴壁生长，并且可以使G0／G1期细胞百分率达到66．5％．因此，血清饥饿法可以应用于人表皮细胞HaCaT同步于G0／G1期，从而为后期药物筛选打下基础．

| Abstract: | We have determined the effect of cell cycle in human normal keratinocyte line HaCaT by serum starvation in the study.By using serum-free and low concentration serum DMEM culture medium,cell line HaCaT was cultured;and the cells were collected in 6h,12h,22h,respectively,then cell form was observed by inverted microscope and the results were analysed by flow cytometry.After HaCaT cells in DMEM culture medium containing 0.2%FBS was cultured for 12 h,the cells were adherent growth and 66.5% at G0/G1 phase.The h... |
