多能硫杆菌RubisCO基因的亚克隆及各种启动子对其表达的影响

摘    要:将多能硫杆菌(Thiobacilusversutus)RubisCO基因从两个方向亚克隆到pBR322和pKK223-3载体上,通过酶切的方法对各种质粒的插入方向进行了确证.并进一步对这两种插入方向的质粒在大肠杆菌(Escherichiacoli)中的表达情况进行了酶活性的测定,该基因片段在pKK223-3载体的tac启动子的启动下,表达活性比lac启动子以及pBR322载体上的P1和P2启动子高.

Abstract:The RubisCO gene of Thiobacillus versutus was subcloned to the vectors of pBR322 and pKK223 3 from two directions.The inserting direction of these plasmids had been determined by the way of restriction enzyme.The enzyme activity was expressed by the recombinant plasmids in E.coli had been assayed.The expression activity of the RubisCO gene is higher at the promotion of tac promoter (pKK223 3) than that of the lac, P 1 and P 2 promoters.
