泥鳅胚胎冷冻前后生物学性状变化的研究

摘    要:运用石蜡切片和电镜技术,研究了泥鳅胚胎经液氮冷冻前后外部形态及显微和亚显微结构的变化。形态变化:受精孵冷冻后卵膜起皱,表面六边形结构被破坏。体色素期胚冷冻后表面破裂、胚体收缩。

Abstract:This paper presents a study on changes of the shape, microstructure and ultrasturcture of Misgurnus anguillicaudatus embryos after frozen with liquid nitroen by means of light and. electron microscopy. Results are summarized as follows. Change of shape: Chorion of fertilized egg becomes wrinkled and its hexagon frames are disrupted after having been frozen. After body-pigmented staged embryo is frozen, its skin also becomes broken, its body was shortened. Change of structure: Yolk globules break and stick to isotropic blocks after being forzen. After embryos frozen, there are meshy ice holes in the body. Cell of ectoderm are damaged but most of entoderm cell remain normal. The spearlike ice grow along with the major axis of muscular fibre. Under higher magnification, we can see that muscular fibrelles are illegible and some mitochondria is disintegrated. There are both intracellular and extracellular ice occuring in damaged cells after being frozen. The study also sugguest that DMSO and freezing slowly can reduce the freezing damage.
