美普他酚合成工艺改进

摘    要:报道了以Br2为芳构化试剂、以六氢-3-(3-氧代-1-环己烯基)-1-甲基-2H-氮杂卓-2-酮为原料,制备六氢-3-(3-羟苯基)-1-甲基-2H-氮杂卓-2-酮,和用RED-Al为还原剂、以3-乙基-六氢-3-(3-羟苯基)-1-甲基-2H-氮杂卓-2-酮为原料制备美普他酚的新方法。该方法工艺简单,能耗少,适合大规模制备。n(六氢-3-(3-氧代-1-环己烯基)-1-甲基-2H-氮杂卓-2-酮):n(Br2)-1:1,n(3-乙基-六氢-3-(3-羟苯基)-1-甲基-2H-氮杂卓-2-酮):n(REI)-A1)-1:2,反应温度为室温,反应适宜时间分别为1h和6h,芳构化收率为90%,还原收率达70%。

Abstract:Hexahydro-3-(3-hydroxyphenyl)-1-methyl-2H-azepin-2-one was prepared by aromatization of hexahydro-1-methyl-3-(3-oxocyclohexen-1-yl)-2H-azepin-2-one using Br2 reagent in 90% yield at room temperature.Meptazinol was obtained by reduction of 3-ethylhexahydro-3-(3-hydroxyphenyl)-1-methyl-2H-azepin-2-one using RED-Al as reducing agent in 70% yield.The present approach offers several advantages such as shorter reaction time,high yields,less power consumption,and mild reaction condition,thus suitable for large-sca...
