雪灵芝对大鼠脑缺血再灌注损伤的保护作用

摘    要:观察雪灵芝对大鼠脑缺血再灌注损伤的保护作用.将Wister大鼠30只分为5组,喂养1周,其中3组每天定时分别灌喂1 mL高、中、低不同浓度的雪灵芝溶液,对照组每天定时灌喂1 mL生理盐水.第8天给大鼠做颈总动脉结扎手术.夹闭颈总动脉前分别灌喂不同浓度的雪灵芝溶液.1 h后麻醉大鼠,夹闭大鼠颈总动脉30 min,再灌注120 min,造成大鼠脑缺血再灌注损伤模型,断头取脑并测定大鼠脑组织中的相关生化指标.结果显示:脑组织中,丙二醛(MDA)、乳酸脱氢酶(LDH)、一氧化氮(NO)较对照组明显增高,而超氧化物歧化酶(SOD)较对照组降低.雪灵芝能降低脑组织中MDA、LDH、NO含量.获得了雪灵芝可能通过抗自由基和减轻NO介导的神经毒性来发挥对脑缺血再灌注损伤的保护作用的结论.

Abstract:The protective effect of Xue lingzhi(herb arenariae) on brain ischemia-reperfusion(IR) in rats is studied in this paper.Thirty wister rats were dirided into five groups and fed for one week. Different concentrations of herb arenariae liquor(250,500,and 1 000 mg/kg) were fed to three groups of rats,respeclively,for one week.Same quantity of physiolgical saline was fed to the reference group of rat also for one week.Operations were done on the rats on the eighth day.Those carotid arteries of rats were closed for 30 min and followed by reperfusion for 120 min.The levels of malondialdehyde(MDA),superoxide(SOD),lactate dehydrogenase(LDH) and nitric oxide(NO) in brain bemispheres were detereminded after IR,and the herb arenariae on them were evaluated.After 30 min ischemia and 120 min reperfusion,the contents of MDA and NO increased,and LDH activity increased,but SOD activity decreased in the brain tissues.Herb arenariae could decrease the contents of MDA and NO,the activity of LDH.The results indicate that Xue lingzhi(herb arenariae) provides neuroprotection by antagonizing free radicals and alleviating NO-induced neurotoxic effect on cerebral ischemia-reperfusion in rats.
