摘    要:为探索提取产黄青霉基因组DNA的简便方法,采用起始菌体量0.2 g,10 g/L十六烷基三甲基溴化铵裂解液,通过液氮/65℃反复冻融对菌种破壁,代替了液氮研磨,且不需要玻璃珠,操作简便,重复性好,提取的DNA质量高,极大地方便了后续基因操作.

Abstract:A method for extraction of genomic DNA from filamentous fungus Penicillium chrysogenum were developed.Mycelia were lysed by 10 g/L CTAB(Hexadecyltrimethylammomum bromide) and a liquid nitrogen/65 ℃ freeze-thaw procedure.The quality of DNA was demonstrated by PCR(polymerase chain reaction).This rapid extraction procedure is a good repeatability method and is suitable for treating many samples at one time,and it provides a method of high throughput strategy.
