摘    要:研究了在Cu(Ⅱ)的存在下柔红霉素与DNA或RNA的荧光猝灭反应,发现单、双链DNA及RNA均能大幅猝灭柔红霉素-Cu(Ⅱ)二元体系的荧光,最佳PH值范围为5.4-7.1,最大激发和发射波长分别在505和555nm。在最佳条件下,测定双链DNA线性范围为0-0.6μg/mL,检测限为0.008μg/mL,相对标准偏差在3.0%以内。温度的影响和吸收光谱证实了DNA对此二元体系的荧光猝灭机理为静态猝灭机理。

Spectrofluorometric Determination of Nucl eic Acids with Daunorubicin in the Presence of Cu(Ⅱ)
Abstract:The fluorescence quenching reaction of daunorubicin with DNA or RN A in the presence of Cu(Ⅱ) was studied. Studies involving natural calf thymus DNA, denatured calf thymus DNA, and yeast RNA revealed that double-stranded, si n gle-stranded DNA and RNA are capable of quenching the fluorescence of the Cu 2+ a nd daunorubicin binary complex, but single-stranded DNA showed little quenching effect. Maximum fluorescence was produced in the pH range 5.4~7.1, with maximum excitation and emission wavelengths at 505 nm and 555 nm, respectiv el y. Under optimal conditions, the calibration graphs were linear between 0 and 0. 6  μg*mL-1for double-standed calf thymus DNA. The corresponding de tection limit was 0.008 μg*mL-1. The relative standard deviation (six replicate s) was within 3 .0% in the linear range. The effect of temperature on the fluorescence intensity and the absorption spectra of Cu2+-daunorubicin complex in the presence of vari ous concentrations of CT DNA show that the quenching process follows the static mechanism.
