摘    要:
综述了现代二核磁共振技术测定DNA结构的进展、方法及其原理和一般步骤,从DNA的基本结构出发,应用二维NOE谱及其它NMR方法,获得大量的质子间距离、键长和键角数据,再经距离几何学、分子动力学等数据处理手段、可获得知DNA在溶液中的三维结构。

Abstract:
Three dimensional structure of DNA in solution has an important significance for understanding the functions of DNA and the misteries of living science.In this paper the methodlogy,strategy and basic principles of structure determination of nuclei acids by 2D NMR techniques have been discussed.From the basic structure of DNA,2D NOE spectroscopy and other NMR methods are applied to the determination of data about interproton distances,bond length and bond angles.Then three dimensional structure of DNA in solution can be obtained by data processing techniques,such as Distance Geometry and Molecular Dynamics.
