革兰氏染色结果的影响因素及原因分析

摘    要:研究了菌龄、涂片厚度、媒染时间、乙醇脱色程度等对革兰氏染色结果的影响。结果表明,在进行革兰氏染色时,大肠杆菌在营养琼脂培养基上的培养时间不宜超过25小时,涂片时应为均匀薄片,媒染时间以30~60s为宜。乙醇脱色程度是革兰氏染色的关键,脱色时间应严格控制在20—30s。而枯草芽孢杆菌的培养时间不宜超过16小时,媒染时间以30~90s为宜,脱色时间应严格控制在30s以内。

Abstract:The influences of bacterium age, the even thickness of smear, mordant agent time, the degree of ethanol decolouration and other related factors on the result of Gram's dye of bacterium are stud- ied. The result shows that E. coli in the nutrient agar culture time should not more than 25 hours in Gram's dyeing. Smear should be even and thin. The mordant dyeing time takes 30 -60s as suitable. The degree of ethanol decolouration is key to experiment and the time of decolouration is strictly controlled between 20-30s. While the cultivation time of Bacillus should not more than 16 hours, the mordant dyeing time takes 60 - 90s as suitable. The dec olorization time should be strictly controlled in 30s.
