华根霉固态发酵产纤溶酶的工艺优化和初步提纯

摘    要:采用华根霉TK317(Rhizopus chinensis)进行固态放大发酵实验,发酵培养基初始含水量为0.75mL/g,固体发酵罐中湿度控制在70%,发酵前期30h通风量为640L/min,后30h通风量变为560L/min,酶活力最高,达到706.3U/g干基。研究并确定了中空纤维超滤浓缩,硫酸铵盐析的分离提取工艺,经过纯化,酶的比活力达23.32U/mg,比浸提液提高11.05倍,酶活力回收率为70.2%。

Abstract:The research results on solid fermentation technology of Rhizopus chinensis indicated: the appropriate conditions were beginning moisture content 0. 75 mL/g dry substance, fermentation humidity 70%, beginning ventilation 640 L/min,560 L/min after 30 h. Productivity of the fibrinolytic enzyme went up to peak value by above conditions. The crude enzyme was purified with ultrafiltra-tion concentration and ammonium sulfate fractionation. The specific activity was 23.32 U/mg, 70. 2% of the fibrinolytic enzyme was recovered with 11.05-fold purification.
