摘    要:目的:首次从秋葵荚中分离制备异槲皮苷和槲皮素-龙胆二糖苷对照品并应用高效液相色谱法建立秋葵荚中两种黄酮苷含量的测定方法.方法:色谱柱:Welch Materials XB-C18(4.6 mm×250 mm,5μm);流动相∶甲醇-水(42∶58);检测波长:256 nm;流速:1 mL.min-1;柱温:30℃.结果:异槲皮苷和槲皮素-龙胆二糖苷进样量分别在0.165μg~0.825μg和0.155μg~0.775μg范围内呈良好的线性关系(r=0.9999,r=0.9995),平均回收率(n=6)分别为99.2%、100.3%.结论:该方法简便、准确,重复性好,可用于测定秋葵中异槲皮苷和槲皮素-龙胆二糖苷的含量.

Abstract:To firstly report the isolation of quercetin 3-0-glucopyranoside and quercetin3-0-gentiobioside from the crude extract of okra,and establish an HPLC method for determination of the two flavonoid glycosides.Methods: Welch Materials XB-C18(4.6 mm×250 mm,5 μm) column was adopted;The mobile phase was methanol-water(42:58) at the flow rate of 1 mL·min-1;The detecting wavelength was 256 nm,and the column temperature was 30 ℃.Results: The method had a good linearity in the range of 0.165 μg~ 0.825 μg(r=0.9999) for quercetin 3-0-glucopyranoside and 0.155 μg~ 0.775 μg(r=0.9995) for quercetin3-0-gentiobioside.The recoveries were 92% and 100.3%.Conclusion: The method is simple,accurate and reproducible.It can be used for determination of flavonoid glycosides in okra.
