响应面法优化洋金花中东莨菪碱的提取

摘    要:运用响应面法进行实验设计,对洋金花中东莨菪碱在超声提取过程中的各因素进行考察优化.采用HPLC法测定东莨菪碱的质量浓度,以盐酸浓度、提取时间、液料比为考察因素,东莨菪碱提取率为响应值,用Box-Behnken响应面法设计提取工艺实验,建立预测方程,优化提取工艺.结合实际可操作性,确定最佳提取条件为:盐酸浓度3 mol·L~(-1),提取时间43 min,液料比25 mL·g~(-1).实验结果显示,实际值与预测值的偏差为2.76%,相关性良好,说明对洋金花中东莨菪碱的提取进行响应面法优化合理可行.

Abstract:In this study, the extraction method of scopolamine from Datura metel L. was the ultrasonic extraction, while its content was detected by HPLC. Factors in the extraction process were investigated in a series of experiments designed by response surface methodology. Combined with the actual situation and the operable principles, the optimized conditions were as follows: the volume of hydrochloric acid was 3 mol?L-1, extraction time was 43 min, and the ratio of liquid to solid was 25 mL?g-1. The results displayed that the difference between the real value and the predicted value was 2.76%, showed good correlation, which indicating that the optimization on the extraction of scopolamine from the flower of Datura metel L. by response surface methodology was reasonable and practicable.
