HSP70与ACP在小鼠前胃癌及大鼠胃癌癌变过程中表达的比较研究

摘    要:目的:本实验分别用NSEE(N-Nitrososarcosineethylester)和MNNG(N-Methyl-N-nitro-ni-trosoguanidine)作为诱变剂建立NIH小鼠前胃癌动物模型和Wistar大鼠胃癌早期动物模型,探讨HSP70和酸性磷酸酶在小鼠前胃癌及大鼠胃癌癌变过程中的表达和意义.方法:在癌变过程中的不同时期处死实验动物,分别取小鼠前胃及大鼠的胃,并将两种材料各分为2部分,一部分作病理切片和免疫组化;一部分作电泳和免疫印迹.对小鼠前胃癌及大鼠胃癌癌变过程中细胞形态的改变,HSP70和ACP的表达进行跟踪研究.结果:(1)在小鼠前胃癌癌变过程中,HSP70的表达在给药后14 d,28 d增强,42 d虽然有所下降,但仍然高于对照组,此后HSP70表达依次增强,且总变化趋势为增强;ACP活性随癌变进程也呈递增趋势.(2)在大鼠胃癌癌变过程中,HSP70在21 d表达较高,然后恢复至正常,在105 d以后逐渐增高;ACP的表达与HSP70一致.细胞形态的变化与HSP70、ACP的表达呈现出一定的对应关系.结论:在诱发NIH小鼠前胃癌及Wistar大鼠胃癌的过程中,HSP70、ACP的表达均呈升高趋势,同时与组织细胞形态的改变相对应.

Abstract:Objective: Using NSEE and MNNG as induced reagent,the animal models of NIH mouse's pregastric carcinoma and Wistar rat's gastric carcinoma were made,both of which were used to explore the meaning and expressions of heat shock proteins 70 and acid phophotase.Methods: The experimental animals were killed in different periods of inducing cancer,the pregastric of every mouse and the gastric of every rat were taken.The material of every mouse or rat was divided into two parts.One of the parts was used for histopathologic slices and immunohistochemestry,and the other was used for electrophoresis and Western blotting.Then,the dynamic changes of cell shapes,HSP70 and ACP were studied. Results:(1) During the development of NIH mouse's pregastric carcinoma,on the 14th day and 28th day the expressions of HSP70 increased,on the 42nd day,the expression of HSP70 was between the 14th day and 28th day,then the expression of HSP70 increased obviously again,so the trend of HSP70 expression was still increasing.The trend of ACP activity was increasing.(2) During the development of Wistar rat's gastric carcinoma,the change of the HSP70 is that the high expressions by 21st day,then weaken and recover to the general,and improve the expression step by step after 105th day.The change of ACP is as same as HSP70.The change of cell shapes is corresponds with the expression of HSP70 and ACP.Conclusion: During the carcinogenesis of NIH mouse's pregastric carcinoma and Wistar rat's gastric carcinoma,HSP70 and ACP appeared regular changes and they were all corresponding to the changes of cell shapes.
