地黄脱毒技术研究与应用

摘    要:建立了地黄的脱病毒技术与工厂化生产技术,研究表明:地黄茎段培养适宜的消毒剂为0.05%HgCl2水溶液,消毒时间为8min~10min;诱导茎尖适宜的激素种类为6-BA,用量0.25mg/L;地黄试管苗工厂化生产的培养基配方为MS+1/4大量元素+1/2微量元素+1/2白砂糖,可使成本降低48.7%.田间应用结果表明:脱毒地黄的光合速率、蒸腾速率及块茎膨大速度较对照明显增加,表现趋势为试管苗〉原原种苗〉原种苗〉对照苗。

Abstract:The technique of Rehmannia virus-free and its application were studied.The results showed that the proper sterilizing agent was 0.05 % HgCl_2 for 810 min and the phytohormone added in medium was 0.25mg/L 6-BA for stem tip culture of Rehmannia.The MS medium contained with 1/4 macro element,(1/2) micro element and (1/2) sugar was the best in mass production of test-tube plantlet of Rehmannia which could reduce the cost by 48.7 %.The photosynthesis rate ,transpiration ratio and root expanding speed of virus-free Rehmannia increased greatly in contrast with the CK in field experiment,and the results was test-tube plantlet>pre-original seedlings>original seedlings>seedlings.
