| 摘 要： | 常规的农杆菌质粒DNA提取常采用碱裂解和酚、氯仿提纯方法，得率较低，一般在50ng／uL以下。就经典的碱裂解法(方法一)和在碱裂解法的基础上改进的方法二(去除了酚、氯仿提取过程，减少了提取过程中对人的伤害，且缩短了提取时间)以及改进的方法三进行了比较，实验结果表明：改进的方法二更适宜于做PCR检测，而方法三则适合于重组质粒DNA的酶切、PCR鉴定等。

| Abstract: | The usual protocol to extract plasmid DNA from Agribacterium tumefaciens is called alkaline lysis. Based on this method(called Method I in this paper) ,a new protocol called Method II and MethodIII respectiveLy is modified , experimental result indicates that Method II ,which is simple, reliable , is more suitable for PCR method to assay target DNA,meanwhile Method III is reliably usable to test recombination plasmid DNA with restriction endonuclease and PCR method. |
