Electrochemical behavior of gold and its associated minerals in alkaline thiourea solutions

Electrochemical measurements were conducted to study the electrochemical behavior of gold (Au) and its commonly associated minerals in alkaline thiourea solutions. The results indicated that without addition of any stabilizer, selective dissolution of Au from stibnite and pyrite was only possible at relatively low thiourea concentrations. As Na₂SiO₃ was added, pyrite started to become active and an oxidation peak appeared; the oxidation peaks of arsenopyrite and chalcocite appeared earlier than that of Au. The chalcocite peak shifted in the positive direction and the peak current increased. Stibnite did not show an oxidation peak and its current was nearly zero. Adding Na₂SiO₃ favored the selective dissolution of Au when its minerals were associated with chalcocite and stibnite. At pH 12, the Au anode dissolution peak current increased with stabilizer concentration. At 0.38 and 0.42 V and for Na₂SiO₃ concentration below 0.09 M, the current density continuously increased with Na₂SiO₃ concentration. The Na₂SiO₃ concentration had to be adequate to stabilize thiourea. When the potential was higher than 0.42 V, the surface of the Au electrode started to passivate. With an additional increase in potential, the presence of Na₂SiO₃ could not stop the inevitable decomposition of thiourea.

Sequencing of phosphopeptides sulfonated by 4-sulfophenyl isothiocyanate using post-source decay matrix-assisted laser desorption/ionization time-of-flight mass spectrometry

Phosphorylation/dephosphorylation is probably the most common and important reversible post-translational modification of proteins. Analyzing the functional effects of phosphorylation is helpful for understanding the biological functions of proteins. Identification of the phosphorylation sites of phosphorylated protein is a prerequisite for research on phosphorylation. In this work, an effective and simple method of identification of protein phosphorylation sites has been developed. Phosphopeptides were selectively enriched with immobilized metal affinity chromatography (IMAC) and subsequently chemically modified by 4-sulfophenyl isothiocyanate, and then the chemically modified phosphopeptides were sequenced with post-source decay (PSD) matrix-assisted laser desorption/ionization (MALDI) time-of-flight mass spectrometry for detecting phosphorylation sites. The charge of derivatization by 4-sulfophenyl isothiocyanate introduces a negative sulfonic acid group at the N-terminus of a peptide, and enables the selective detection of only a single series of C-terminal y-type ions. This chemically assisted method greatly simplifies the extremely complex pattern of PSD fragment ions and makes the PSD spectra more easier to be interpreted. The phosphorylation sites of a synthesized model phosphopeptide and human c-myc protein have been successfully identified by this method.

Transitional phenomenon of particle dispersion in gas-solid two-phase flows

Without using any turbulent model, direct numerical simulation of a three-dimensional gas-solid two-phase turbulent jet was performed by finite volume method. The effects on dispersion of particles with different Stokes numbers by the transitional behavior of turbulent structures were investigated. To produce high-resolution results and reduce the computation and storage, the fractional-step projection algorithm was used to solve the governing equations of gas phase fluid. The low-storage, three-order Runge-Kutta scheme was used for time integration. The governing equations of particles were solved in the Lagrangian framework. These numerical schemes were validated by the good agreement be-tween the statistical results of flow field and the related experimental data. In the study of particle dis-persion, it was found that the effects on particle dispersion by the spanwise vortex structures were prominent. The new behaviors of particle dispersion were also observed during the evolution of the flow field, i.e. the transitional phenomenon of particle dispersion occurs for the particles with small and intermediate Stokes numbers.     

Modification of the rib operon derived from Bacillus subtilis and its expression in Escherichia coli

A riboflavin operon （rib operon） derived from Bacillus subtilis 368 was modified on structure and the resulting operons were expressed in various strains of Escherichia coli. The results showed that the optimization of the rib operon and the host strain used for expression are two main factors affecting the riboflavin production. Replacing the promoterl and rfn box of the rib operon with a strong constructive promoter spol drastically increased the expression of the rib genes. When E. coli JM109 was used as the host strain, the highest riboflavin production reached 95.3μg/mL （about eight times higher than that of the unmodified r/b operon）. In addition, when tetracycline （20 μg/mL） was used as the selective pressure, compared with the ampicillin resistant transformants, a higher riboflavin yield was obtained in tetracycline resistant host strain.     

A passive optical network based on optical code division multiplexing and time division multiple access technology

A passive optical network （PON） scheme based on optical code division multiplexing （OCDM） for the downstream traffics is proposed and analyzed in detail. In the PON, the downstream traffics are broadcasted by OCDM technology to guarantee the security, while the upstream traffics pass through the same optical fiber by the common time division multiple access （TDMA） technology to decrease the cost. This schemes are denoted as OCDM/TDMA-PON, which can be applied to an optical access network （OAN） with full services on demand, such as Internet protocol, video on demand, tele-presence and high quality audio. The proposed OCDM/TDMA-PON scheme combines advantages of PON, TDMA, and OCDM technology. Simulation results indicate that the designed scheme improves the OAN performance, and enhances flexibility and scalability of the system.
