Distribution and first reports of Branchiobdellida (Annelida: Clitellata) on crayfish in the prairie provinces of Canada

Orconectes virilis (northern crayfish) were collected from 67 sites in Alberta, Saskatchewan, Manitoba, and far-western Ontario, Canada, and yielded 2 species of branchiobdellidans, Cambarincola vitreus and Cambarincola chirocephalus. This is the first report of branchiobdellidans in the Prairie Provinces. Cambarincola vitreus was distributed across the study area, but C. chirocephalus appeared to be restricted to southeastern Saskatchewan, southern Manitoba, and the site in western Ontario. Neither branchiobdellidan species was observed on crayfish in the Beaver River or the South Saskatchewan River and associated tributaries upstream (west) of Saskatoon, Saskatchewan, despite multiple sampling at these locations.     

Systematic pathway analysis using high-resolution fitness profiling of combinatorial gene deletions

Systematic genetic interaction studies have illuminated many cellular processes. Here we quantitatively examine genetic interactions among 26 Saccharomyces cerevisiae genes conferring resistance to the DNA-damaging agent methyl methanesulfonate (MMS), as determined by chemogenomic fitness profiling of pooled deletion strains. We constructed 650 double-deletion strains, corresponding to all pairings of these 26 deletions. The fitness of single- and double-deletion strains were measured in the presence and absence of MMS. Genetic interactions were defined by combining principles from both statistical and classical genetics. The resulting network predicts that the Mph1 helicase has a role in resolving homologous recombination-derived DNA intermediates that is similar to (but distinct from) that of the Sgs1 helicase. Our results emphasize the utility of small molecules and multifactorial deletion mutants in uncovering functional relationships and pathway order.     

A simple procedure for removal of intra- and extra-tumoral macrophages from primary tumor cultures

Summary Improved growth of tumor cells in primary cell suspensions isolated from solid tumors can be achieved by addition of an antifungal agent, mycostatin, to the culture medium. Mycostatin inhibits the cytotoxicity of macrophages and also quickly detaches phagocytes without any apparent retardation of the growth of tumor cells. Anti-endotoxin (anti-lipid-A) antibodies

Summary Anti-lipid-A, anti-endotoxin, antibodies have been measured by a passive haemolysis test using antigen from anE. coli Re mutant. Titres in the normal population are low but do rise in situations in which there has been gram-negative sepsis. Distribution of Entocytheridae (Crustacea: Ostracoda) in the northern prairies of North America and reports of opportunistic clitellate annelids on crayfish hosts

Northern crayfish, Orconectes virilis (Hagen, 1870), were collected from 89 sites across Alberta, Saskatchewan, Manitoba, Montana, North Dakota, and Minnesota. The entocytherid ostracod Thermastrocythere riojai (Hoff, 1943) was found on O. virilis at 45 of the 89 sites, distributed primarily in the eastern and southern portion of the study area. These observations of T. riojai greatly extend the known range of the species. The widespread distribution of T. riojai suggests that the dearth of entocytherid records from other parts of Canada is a result of nontargeted sampling rather than true absence. In addition, we report on observations of 3 noteworthy associations of oligochaetes with their crayfish hosts. Encontramos el ostracódo entocitérido Thermastrocythere riojai (Hoff, 1943) en O. virilis en 45 de los 89 sitios, distribuidos principalmente en la parte sur y la parte este del área de estudio. Estas observaciones de T. riojai amplían considerablemente el área de distribución conocida de esta especie. La distribución extensa de T. riojai sugiere que la escasez de registros de entocitéridos en otras partes de Canadá es el resultado del muestreo inespecífico y no de una ausencia verdadera. Además, reportamos observaciones de 3 asociaciones sobresalientes entre los oligoquetos y los acociles huéspedes.     

Loss of integrin alpha(v)beta8 on dendritic cells causes autoimmunity and colitis in mice

The cytokine transforming growth factor-beta (TGF-beta) is an important negative regulator of adaptive immunity. TGF-beta is secreted by cells as an inactive precursor that must be activated to exert biological effects, but the mechanisms that regulate TGF-beta activation and function in the immune system are poorly understood. Here we show that conditional loss of the TGF-beta-activating integrin alpha(v)beta8 on leukocytes causes severe inflammatory bowel disease and age-related autoimmunity in mice. This autoimmune phenotype is largely due to lack of alpha(v)beta8 on dendritic cells, as mice lacking alpha(v)beta8 principally on dendritic cells develop identical immunological abnormalities as mice lacking alpha(v)beta8 on all leukocytes, whereas mice lacking alpha(v)beta8 on T cells alone are phenotypically normal. We further show that dendritic cells lacking alpha(v)beta8 fail to induce regulatory T cells (T(R) cells) in vitro, an effect that depends on TGF-beta activity. Furthermore, mice lacking alpha(v)beta8 on dendritic cells have reduced proportions of T(R) cells in colonic tissue. These results suggest that alpha(v)beta8-mediated TGF-beta activation by dendritic cells is essential for preventing immune dysfunction that results in inflammatory bowel disease and autoimmunity, effects that are due, at least in part, to the ability of alpha(v)beta8 on dendritic cells to induce and/or maintain tissue T(R) cells.
