Small molecule activators of SIRT1 as therapeutics for the treatment of type 2 diabetes

Calorie restriction extends lifespan and produces a metabolic profile desirable for treating diseases of ageing such as type 2 diabetes. SIRT1, an NAD+-dependent deacetylase, is a principal modulator of pathways downstream of calorie restriction that produce beneficial effects on glucose homeostasis and insulin sensitivity. Resveratrol, a polyphenolic SIRT1 activator, mimics the anti-ageing effects of calorie restriction in lower organisms and in mice fed a high-fat diet ameliorates insulin resistance, increases mitochondrial content, and prolongs survival. Here we describe the identification and characterization of small molecule activators of SIRT1 that are structurally unrelated to, and 1,000-fold more potent than, resveratrol. These compounds bind to the SIRT1 enzyme-peptide substrate complex at an allosteric site amino-terminal to the catalytic domain and lower the Michaelis constant for acetylated substrates. In diet-induced obese and genetically obese mice, these compounds improve insulin sensitivity, lower plasma glucose, and increase mitochondrial capacity. In Zucker fa/fa rats, hyperinsulinaemic-euglycaemic clamp studies demonstrate that SIRT1 activators improve whole-body glucose homeostasis and insulin sensitivity in adipose tissue, skeletal muscle and liver. Thus, SIRT1 activation is a promising new therapeutic approach for treating diseases of ageing such as type 2 diabetes.

Action Learning, Action Research, and Process Management Association (ALARPM) is an organization of volunteers dedicated to the international expansion of action learning, action research, and process management, through world congresses. It has existed for over a dozen years now, despite significant stresses and strains, and has successfully conducted five world congresses with a sixth one in 2003. This history of ALARPM shows that a small group can set out to be international and inclusive from the beginning, so long as it also develops processes to sustain itself internally.     

The influence of cobalt on the endoplasmatic reticulum of the horse bean (Vicia faba L.)

Summary The exogeneous application of cobalt induces the differentiation of membrane complexes of the endoplasmic reticulum. After longer acting of cobalt these membranes are dilated and later destroyed. This fact can manifest itself also in some disturbances of the cell division.     

Aquatic parameters and life history observations of the Great Basin spadefoot toad in Utah

The distribution and breeding habitats of the Great Basin spadefoot toad ( Scaphiopus intermontanus ) were investigated in the Bonneville Basin of western Utah. The permanent springs and man-made reservoirs used for breeding were largely found below the 1600 m elevation. The pH's ranged between 7.2 and 10.4 and the total dissolved solids between 170 and 4800 mg/l. The springs were less alkaline than the rain-filled reservoirs. The lack of aquatic vegetation was a common feature of the reservoirs and most of the springs. Observations of breeding without rain are noted as well as the lack of breeding with rain. The snout – vent lengths of adult spadefoots are greater in the Bonneville Basin than in other parts of the Great Basin. Utilization of permanent water sources and stimuli for emergence and breeding, as well as the larger adult size of S. intermontanus in the Bonneville Basin, are discussed in relation to the diverse precipitation patterns, the sparseness of the water sources, and the Holocene history of the Great Basin.

Summary The ontogeny ofa-foetoprotein (AFP) has been studied in the chicken using polyacrylamide gel gradient electrophoresis and electroimmunoassay from 7 days of incubation until after hatching. The results are discussed in the light of previous on the ontogeny of AFP in mammals.     

Microwave devices: carbon nanotubes as cold cathodes

To communicate, spacecraft and satellites rely on microwave devices, which at present are based on relatively inefficient thermionic electron sources that require heating and cannot be switched on instantaneously. Here we describe a microwave diode that uses a cold-cathode electron source consisting of carbon nanotubes and that operates at high frequency and at high current densities. Because it weighs little, responds instantaneously and has no need of heating, this miniaturized electron source should prove valuable for microwave devices used in telecommunications.
