Heat shock protein 90 (Hsp90) is a molecular chaperone that plays a key role in the conformational maturation of oncogenic signalling proteins, including HER-2/ErbB2, Akt, Raf-1, Bcr-Abl and mutated p53. Hsp90 inhibitors bind to Hsp90, and induce the proteasomal degradation of Hsp90 client proteins. Although Hsp90 is highly expressed in most cells, Hsp90 inhibitors selectively kill cancer cells compared to normal cells, and the Hsp90 inhibitor 17-allylaminogeldanamycin (17-AAG) is currently in phase I clinical trials. However, the molecular basis of the tumour selectivity of Hsp90 inhibitors is unknown. Here we report that Hsp90 derived from tumour cells has a 100-fold higher binding affinity for 17-AAG than does Hsp90 from normal cells. Tumour Hsp90 is present entirely in multi-chaperone complexes with high ATPase activity, whereas Hsp90 from normal tissues is in a latent, uncomplexed state. In vitro reconstitution of chaperone complexes with Hsp90 resulted in increased binding affinity to 17-AAG, and increased ATPase activity. These results suggest that tumour cells contain Hsp90 complexes in an activated, high-affinity conformation that facilitates malignant progression, and that may represent a unique target for cancer therapeutics.  相似文献

Despite the growth of the IT industry and the high demand for IT skills university departments seem unable to attract enough student to cater for the needs of the profession.There seems to be a misconception about the difference between IT user skills and the discipline of Computing.This article attempts to do three things:1)reinforce the view that a crisis of IT skills shortage is looming;2)reiterate the fact that computing science is an exciting discipline that is central to the knowledge economy and that career prospects in IT-related jobs are very good despite the fears surrounding outsourcing and 3)suggest that HE computing departments must be flexible enough to embrace a two-facetted challenge:a)the changing landscape of IT Skills and b)the high hopes and aspirations of the first generation of our digital-native student community.  相似文献

Here we present a finished sequence of human chromosome 15, together with a high-quality gene catalogue. As chromosome 15 is one of seven human chromosomes with a high rate of segmental duplication, we have carried out a detailed analysis of the duplication structure of the chromosome. Segmental duplications in chromosome 15 are largely clustered in two regions, on proximal and distal 15q; the proximal region is notable because recombination among the segmental duplications can result in deletions causing Prader-Willi and Angelman syndromes. Sequence analysis shows that the proximal and distal regions of 15q share extensive ancient similarity. Using a simple approach, we have been able to reconstruct many of the events by which the current duplication structure arose. We find that most of the intrachromosomal duplications seem to share a common ancestry. Finally, we demonstrate that some remaining gaps in the genome sequence are probably due to structural polymorphisms between haplotypes; this may explain a significant fraction of the gaps remaining in the human genome.  相似文献

Community structure is one of the most best-known properties of complex networks. Finding communities help us analyze networks from a mesoscopic viewpoints instead of microscopic or macroscopic one. It helps to understand behavior grouping. Various community detection algorithms have been proposed with some shortcomings in time and space complexity, accuracy, or stability. Label Propagation Algorithm(LPA) is a popular method used for finding communities in an almost-linear time-consuming process. However, its performance is not satisfactory in some metrics such as accuracy and stability. In this paper, a new modified version of LPA is proposed to improve the stability and accuracy of the LPA by defining two concepts-nodes and link strength based on semi-local similarity-,while preserving its simplicity. In the proposed method a new initial node selection strategy, namely the tiebreak strategy, updating order and rule update are presented to solve the random behavior problem of original LPA. The proposed algorithm is evaluated on artificial and real networks. The experiments show that the proposed algorithm is close to linear time complexity with better accuracy than the original LPA and other compared methods. Furthermore, the proposed algorithm has the robustness and stability advantages while the original LPA does not have these features.  相似文献

Loss of tight association between epidermis and dermis underlies several blistering disorders and is frequently caused by impaired function of extracellular matrix (ECM) proteins. Here we describe a new protein in mouse, Fras1, that is specifically detected in a linear fashion underlying the epidermis and the basal surface of other epithelia in embryos. Loss of Fras1 function results in the formation of subepidermal hemorrhagic blisters as well as unilateral or bilateral renal agenesis during mouse embryogenesis. Postnatally, homozygous Fras1 mutants have fusion of the eyelids and digits and unilateral renal agenesis or dysplasia. The defects observed in Fras1-/- mice phenocopy those of the existing bl (blebbed) mouse mutants, which have been considered a model for the human genetic disorder Fraser syndrome. We show that bl/bl homozygous embryos are devoid of Fras1 protein, consistent with the finding that Fras1 is mutated in these mice. In sum, our data suggest that perturbations in the composition of the extracellular space underlying epithelia could account for the onset of the blebbed phenotype in mouse and Fraser syndrome manifestation in human.  相似文献

We report a high-quality draft of the genome sequence of the grey, short-tailed opossum (Monodelphis domestica). As the first metatherian ('marsupial') species to be sequenced, the opossum provides a unique perspective on the organization and evolution of mammalian genomes. Distinctive features of the opossum chromosomes provide support for recent theories about genome evolution and function, including a strong influence of biased gene conversion on nucleotide sequence composition, and a relationship between chromosomal characteristics and X chromosome inactivation. Comparison of opossum and eutherian genomes also reveals a sharp difference in evolutionary innovation between protein-coding and non-coding functional elements. True innovation in protein-coding genes seems to be relatively rare, with lineage-specific differences being largely due to diversification and rapid turnover in gene families involved in environmental interactions. In contrast, about 20% of eutherian conserved non-coding elements (CNEs) are recent inventions that postdate the divergence of Eutheria and Metatheria. A substantial proportion of these eutherian-specific CNEs arose from sequence inserted by transposable elements, pointing to transposons as a major creative force in the evolution of mammalian gene regulation.  相似文献

Structural, thermal, and mechanical properties of pure blend and nanocomposites based on polyurethane (PU) and polyvinyl chloride (PVC) doped with low different content of single walled-carbon nanotubes (SWCNTs) were studied. The nanocomposites at different concentration were prepared via casting technique. The interaction between PU/PVC and CNTs were examined via FT-IR studies. The changes in the structures of the nanocomposites were examined using X- Ray Diffraction (XRD), and the results indicated that the amorphous domains of nanocomposites increased with increasing SWCNTs content. Transmission electron microscope (TEM) observation indicated that SWCNTs surface was wrapped with the polymer with the thermal properties of nanocomposites improved. The mechanical behavior of the nanocomposites was evaluated as a function of SWCNTs content. The main enhancement in tensile properties was observed, e.g., the tensile strength and elastic modulus increased compared with the pure blend, which may be attributed to the interaction and adhesion between CNTs and the polymer matrices due to the hydrogen bonding between carbonyl groups (C=O) of polymer blend chains and carboxylic acid (COOH) groups of CNTs.  相似文献

Since the discovery of the first extrasolar giant planets around Sun-like stars, evolving observational capabilities have brought us closer to the detection of true Earth analogues. The size of an exoplanet can be determined when it periodically passes in front of (transits) its parent star, causing a decrease in starlight proportional to its radius. The smallest exoplanet hitherto discovered has a radius 1.42 times that of the Earth's radius (R(⊕)), and hence has 2.9 times its volume. Here we report the discovery of two planets, one Earth-sized (1.03R(⊕)) and the other smaller than the Earth (0.87R(⊕)), orbiting the star Kepler-20, which is already known to host three other, larger, transiting planets. The gravitational pull of the new planets on the parent star is too small to measure with current instrumentation. We apply a statistical method to show that the likelihood of the planetary interpretation of the transit signals is more than three orders of magnitude larger than that of the alternative hypothesis that the signals result from an eclipsing binary star. Theoretical considerations imply that these planets are rocky, with a composition of iron and silicate. The outer planet could have developed a thick water vapour atmosphere.  相似文献
