Nanoporous molecular frameworks are important in applications such as separation, storage and catalysis. Empirical rules exist for their assembly but it is still challenging to place and segregate functionality in three-dimensional porous solids in a predictable way. Indeed, recent studies of mixed crystalline frameworks suggest a preference for the statistical distribution of functionalities throughout the pores rather than, for example, the functional group localization found in the reactive sites of enzymes. This is a potential limitation for 'one-pot' chemical syntheses of porous frameworks from simple starting materials. An alternative strategy is to prepare porous solids from synthetically preorganized molecular pores. In principle, functional organic pore modules could be covalently prefabricated and then assembled to produce materials with specific properties. However, this vision of mix-and-match assembly is far from being realized, not least because of the challenge in reliably predicting three-dimensional structures for molecular crystals, which lack the strong directional bonding found in networks. Here we show that highly porous crystalline solids can be produced by mixing different organic cage modules that self-assemble by means of chiral recognition. The structures of the resulting materials can be predicted computationally, allowing in silico materials design strategies. The constituent pore modules are synthesized in high yields on gram scales in a one-step reaction. Assembly of the porous co-crystals is as simple as combining the modules in solution and removing the solvent. In some cases, the chiral recognition between modules can be exploited to produce porous organic nanoparticles. We show that the method is valid for four different cage modules and can in principle be generalized in a computationally predictable manner based on a lock-and-key assembly between modules.  相似文献

The unique and evolutionary highly conserved major vault protein (MVP) is the main component of ubiquitous, large cellular ribonucleoparticles termed vaults. The 100 kDa MVP represents more than 70% of the vault mass which contains two additional proteins, the vault poly (ADP-ribose) polymerase (vPARP) and the telomerase-associated protein 1 (TEP1), as well as several short untranslated RNAs (vRNA). Vaults are almost ubiquitously expressed and, besides chemotherapy resistance, have been implicated in the regulation of several cellular processes including transport mechanisms, signal transmissions and immune responses. Despite a growing amount of data from diverse species and systems, the definition of precise vault functions is still highly complex and challenging. Here we review the current knowledge on MVP and vaults with focus on regulatory functions in intracellular signal transduction and immune defence. Zusammenfassung Mit Hilfe der Mikroelektrophorese wurde die Wirkung von Dexamethasonphosphat, Dopamin und ACTH lokal an Einzelneuronen des Hypothalamus der Ratte geprüft. Dexamethason-phosphat (ein synthetisches Corticosteroid) hemmte mehrheitlich die Aktivität der steroidempfindlichen Zellen im Hypothalamus, einige wenige dieser Zellen wurden aktiviert. Dopamin hemmte die Aktivität dieser steroidempfindlichen Neuronen sehr stark. ACTH dagegen aktivierte diese Zellen. Diese Resultate werden in Zusammenhang mit einem negativen und einem positiven «Feedback»-Mechanismus, der die ACTH-Bildung steuert, diskutiert.  相似文献

The filamins are cytoplasmic proteins that regulate the structure and activity of the cytoskeleton by cross-linking actin into three-dimensional networks, linking the cell membrane to the cytoskeleton and serving as scaffolds on which intracellular signaling and protein trafficking pathways are organized (reviewed in refs. 1,2). We identified mutations in the gene encoding filamin B in four human skeletal disorders. We found homozygosity or compound heterozygosity with respect to stop-codon mutations in autosomal recessive spondylocarpotarsal syndrome (SCT, OMIM 272460) and missense mutations in individuals with autosomal dominant Larsen syndrome (OMIM 150250) and the perinatal lethal atelosteogenesis I and III phenotypes (AOI, OMIM 108720; AOIII, OMIM 108721). We found that filamin B is expressed in human growth plate chondrocytes and in the developing vertebral bodies in the mouse. These data indicate an unexpected role in vertebral segmentation, joint formation and endochondral ossification for this ubiquitously expressed cytoskeletal protein.  相似文献

Summary In adrenalectomized rats, the injection of a single high dose of 5-hydroxytryptamin results, as in intact animals previously reported, in a strong increase of the number of platelets in the peripheral blood. Résumé Des agents blastomitogéniques furent décelés non seulement dans les extraits de 4 genres propres à la famille des Légumineuses, mais aussi dans les graines d'espèces déterminées appartenant aux familles des Solanacées, Composées, Ephédracées et Clusiacées. Ces 4 dernières sources, non mentionnées au préalable, témoignent de la répartition variée de ces agents dans la nature et donnent lieu à des conjectures aux points de vue de leur affinité chimique, du mécanisme de leur action blastomitogénique, de leur rôle dans la physiologie végétale et de leur importance dans l'évolution.  相似文献

This work presents the first list of Coleoptera species found in Hohenbergia augusta (Vell.) E. Morren and Vriesea friburgensis Mez. (Bromeliaceae), in Santa Catarina state, Brazil, most of them identified to the species level. Beetle species richness and composition were compared between the bromeliad species and between Atlantic forest and restinga habitats. Biological information on some beetle species is reported for the first time. We also discuss the aquatic habit of the hydrophilid Lachnodacnum luederwaldti Orchymont, 1937 (Sphaeridinae) inside bromeliad phytotelms.  相似文献
