Potato (Solanum tuberosum L.) is the world's most important non-grain food crop and is central to global food security. It is clonally propagated, highly heterozygous, autotetraploid, and suffers acute inbreeding depression. Here we use a homozygous doubled-monoploid potato clone to sequence and assemble 86% of the 844-megabase genome. We predict 39,031 protein-coding genes and present evidence for at least two genome duplication events indicative of a palaeopolyploid origin. As the first genome sequence of an asterid, the potato genome reveals 2,642 genes specific to this large angiosperm clade. We also sequenced a heterozygous diploid clone and show that gene presence/absence variants and other potentially deleterious mutations occur frequently and are a likely cause of inbreeding depression. Gene family expansion, tissue-specific expression and recruitment of genes to new pathways contributed to the evolution of tuber development. The potato genome sequence provides a platform for genetic improvement of this vital crop. 相似文献

The M-type kinesin isoform, Kif9, has recently been implicated in maintaining a physical connection between the centrosome and nucleus in Dictyostelium discoideum. However, the mechanism by which Kif9 functions to link these two organelles remains obscure. Here we demonstrate that the Kif9 protein is localized to the nuclear envelope and is concentrated in the region underlying the centrosome point of attachment. Nuclear anchorage appears mediated through a specialized transmembrane domain located in the carboxyl terminus. Kif9 interacts with microtubules in in vitro binding assays and effects an endwise depolymerization of the polymer. These results suggest a model whereby Kif9 is anchored to the nucleus and generates a pulling force that reels the centrosome up against the nucleus. This is a novel activity for a kinesin motor, one important for progression of cells into mitosis and to ensure centrosome-nuclear parity in a multinuclear environment. 相似文献

We present the results of twisted plasma waves driven by twisted ponderomotive force. With the beating of two, co-propagating, Laguerre-Gaussian (LG) orbital angular momentum (OAM) laser pulses with different frequencies and also different twist indices, we can obtain the twisted ponderomotive force. Three-dimensional particle-in-cell simulations are used to demonstrate the twisted plasma waves driven by lasers. The twisted plasma waves have an electron density perturbation with a helical rotating structure. Different from the predictions of the linear fluid theory, the simulation results show a nonlinear rotating current and a static axial magnetic field. Along with the rotating current is the axial OAM carried by particles in the twisted plasma waves. A detailed theoretical analysis of twisted plasma waves is also given. 相似文献

Multiple-rock magnetic investigations conducted on the loess-paleosol sequences at Kurtak in Southwestern Siberia reveal that
the mass-normalized low-field magnetic susceptibility profiles reflect changes in lithology between relatively unweathered
primary loess of glacial periods and the interglacial paleosols. Maxima in susceptibility values correspond with the least-weathered
loess horizons, and minima with the humic horizons of soils. Frequency-dependent susceptibility of the loess-paleosol sequences
at Kurtak is very low and practically uniform, indicating the dominance of non-SP ferrimagnetic minerals and negligible pedogenesis.
The history of temperature-dependence of susceptibility (TDS) and stepwise acquisition of the isothermal remanent magnetization
(SIRM) have confirmed that magnetite is predominant magnetic mineral, and only few maghemite and probably hematite are present
within the studied section. Anisotropy of the magnetic susceptibility (AMS) can be used to monitor tilt and disturbance of

Large igneous provinces (LIPs) are known for their rapid production of enormous volumes of magma (up to several million cubic kilometres in less than a million years), for marked thinning of the lithosphere, often ending with a continental break-up, and for their links to global environmental catastrophes. Despite the importance of LIPs, controversy surrounds even the basic idea that they form through melting in the heads of thermal mantle plumes. The Permo-Triassic Siberian Traps--the type example and the largest continental LIP--is located on thick cratonic lithosphere and was synchronous with the largest known mass-extinction event. However, there is no evidence of pre-magmatic uplift or of a large lithospheric stretching, as predicted above a plume head. Moreover, estimates of magmatic CO(2) degassing from the Siberian Traps are considered insufficient to trigger climatic crises, leading to the hypothesis that the release of thermogenic gases from the sediment pile caused the mass extinction. Here we present petrological evidence for a large amount (15?wt%) of dense recycled oceanic crust in the head of the plume and develop a thermomechanical model that predicts no pre-magmatic uplift and requires no lithospheric extension. The model implies extensive plume melting and heterogeneous erosion of the thick cratonic lithosphere over the course of a few hundred thousand years. The model suggests that massive degassing of CO(2) and HCl, mostly from the recycled crust in the plume head, could alone trigger a mass extinction and predicts it happening before the main volcanic phase, in agreement with stratigraphic and geochronological data for the Siberian Traps and other LIPs. 相似文献
