Rosette formation in white blood cell cultures from patients with acute myeloblastic leukemia

Zusammenfassung In Monozyten enthaltenden Leukozyten-Kulturen von Patienten mit akuter myeloischer Leukämie entstehen nach ungefähr 8 h Inkubationszeit Rosetten, die aus einem von Myeloblasten umgebenen Monozyten bestehen. In Analogie zu anderen in vitro-Systemen, in welchen sich eine Rosettenbildung beobachten lässt, könnte diese zelluläre Reaktion auf dem Vorhandensein von spezifischen Antikörpern im Patienten-Serum beruhen, welche gegen die leukämischen Myeloblasten gerichtet sind.     

Verbesserte Methode zur Gewinnung und Verarbeitung von freien Zellen aus kleinsten Volumina von Körperflüssigkeiten mittels Membranfiltration

Summary A cell collector for concentrating small numbers of exfoliated cells on membrane filters from minute volumes (0.05–3 ml) of body fluids and culture media is described. The apparatus incorporates a membrane pump, delivering a vacuum of 10–100 mm Hg. Filtration units (funnels and filter holders) with filtrating areas of 3, 5 and 8 mm diameter are used depending on the volume and the cell content of the fluid to be investigated. A rapid staining procedure based on the classical May-Grünwald-Giemsa technique for light microscopic observations and a modified technique for processing the cell-coated filters for electron microscopy are presented in some detail.

The assessment of tobacco smoke toxicity in organ culture. II. Ultrastructural studies on the immediate response of foetal rabbit tracheal epithelium to short-term exposures of whole smoke.

Foetal rabbit tracheal organ cultures were exposed under defined conditions to whole cigarette smoke and fixed immediately for electron microscopy. After an exposure to 4 or 8 puffs, epithelial intercellular spaces were enlarged, the apical portion of many cells bulged out into the lumen and many cilia were swollen. An exposure to 12 puffs produced a breakdown of the epithelium.     

Targeted mutation of Cyln2 in the Williams syndrome critical region links CLIP-115 haploinsufficiency to neurodevelopmental abnormalities in mice

Williams syndrome is a neurodevelopmental disorder caused by the hemizygous deletion of 1.6 Mb on human chromosome 7q11.23. This region comprises the gene CYLN2, encoding CLIP-115, a microtubule-binding protein of 115 kD. Using a gene-targeting approach, we provide evidence that mice with haploinsufficiency for Cyln2 have features reminiscent of Williams syndrome, including mild growth deficiency, brain abnormalities, hippocampal dysfunction and particular deficits in motor coordination. Absence of CLIP-115 also leads to increased levels of CLIP-170 (a closely related cytoplasmic linker protein) and dynactin at the tips of growing microtubules. This protein redistribution may affect dynein motor regulation and, together with the loss of CLIP-115-specific functions, underlie neurological alterations in Williams syndrome.     

The assessment of tobacco smoke toxicity in tissue and organ culture: An in vitro arrangement for exposure under defined conditions

Zusammenfassung Eine Apparatur zur Berauchung von Zell-und Organkulturen unter kontrollierten Bedingungen (Anzahl und Dauer der Puffs, Temperatur, Luftfeuchtigkeit) wird beschrieben. Das System basiert auf der Verwendung der Filtrona-Berauchungsmaschine (Cigarette Components Ltd.); die zusätzlichen Elemente können leicht aus laboreigenem Material zusammengestellt werden. Frischrauch und wasserdampfgesättigte Gasmischungen (z.B. Luft/CO₂) können in der Berauchungskammer in variablen Proportionen gemischt werden.
