新型蛋白酶抑制剂基因hwtx-11在大肠杆菌中的克隆表达及杀虫功能研究

将化学合成的蜘蛛毒素蛋白酶抑制剂基因hwtx-11克隆至载体pGEX4T-1上,在大肠杆菌BL21(DE3)中进行表达.重组菌株在IPTG诱导下,经SDS-PAGE和Western blot分析表明,表达的GST-HWTX-11融合蛋白的相对分子量约为33 kD.对重组菌株诱导表达后的产物进行生物活性测定,GST-HWTX-11融合蛋白对甜菜夜蛾(Spodoptera exiguaHubner)和棉铃虫(Helicoverpa armigera)在3d时的LC50分别为86.6μg/mL、119.4μg/mL;其与Cry1Ac对甜菜夜蛾和棉铃虫幼虫毒力也有明显的协同作用.     

J2EE平台双因素认证的设计与实现

网络信息安全技术的发展,使得单一的用户口令(PIN)认证方式认证强度已经不能满足用户的安全要求.本文分析了单一认证的弱安全性,结合PKCS 11标准和J2EE平台的EJB组件技术,设计并实现了基于用户口令(PIN)认证和硬件USBKey数字证书身份认证的双重因素认证系统.在所建系统模型基础上,重点构建了双因素认证系统的安全认证协议和签名、认证算法.最后的测试结果显示,在与单一认证系统性能相近的基础上,安全性较高.     

微生物生态学研究进展——第十一届国际微生物生态学大会综述

第11届国际微生物生态学会于2006年8月在奥地利维也纳召开。此次会议以“隐藏的力量——行动中的微生物群落”为主题,特邀了当今世界在微生物生态学领域最为著名的科学家作专题报告,内容广博,反映了当今微生物生态学研究的发展动向。本文简要介绍了第十一届国际微生物生态学大会的概况和部分演讲摘要。

拟南芥钙依赖蛋白激酶CPK11与ABA响应元件结合因子ABF4相互作用的研究

为研究钙依赖蛋白激酶CPK11参与ABA信号通路的方式,本文利用体外酵母双杂实验(Y2H)以及体内双分子荧光互补实验(BiFC)分析CPK11与ABA响应元件结合因子(ABA-responsive element binding factors, ABFs)ABF4之间的关系.酵母双杂交实验表明CPK11与ABF4存在体外相互作用,双分子荧光互补实验表明CPK11与ABF4存在体内相互作用.以上实验共同证明CPK11与ABF4存在直接相互作用.作为CPK11的同源蛋白CPK4与转录因子ABF4在植物体内也存在相互作用.以上实验表明CPK11及其同源蛋白CPK4可能通过与转录因子ABF4相互作用从而参与钙离子介导的ABA信号通路.     

A nonparametric method for asymmetrically extending signal extraction filters

Two important problems in the X‐11 seasonal adjustment methodology are the construction of standard errors and the handling of the boundaries. We adapt the ‘implied model approach’ of Kaiser and Maravall to achieve both objectives in a nonparametric fashion. The frequency response function of an X‐11 linear filter is used, together with the periodogram of the differenced data, to define spectral density estimates for signal and noise. These spectra are then used to define a matrix smoother, which in turn generates an estimate of the signal that is linear in the data. Estimates of the signal are provided at all time points in the sample, and the associated time‐varying signal extraction mean squared errors are a by‐product of the matrix smoother theory. After explaining our method, it is applied to popular nonparametric filters such as the Hodrick–Prescott (HP), the Henderson trend, and ideal low‐pass and band‐pass filters, as well as X‐11 seasonal adjustment, trend, and irregular filters. Finally, we illustrate the method on several time series and provide comparisons with X‐12‐ARIMA seasonal adjustments. Copyright © 2010 John Wiley & Sons, Ltd.     

The molecular basis and clinical aspects of Peutz-Jeghers syndrome

Peutz-Jeghers syndrome (PJS) is a classic, but not widely known hereditary trait. Its clinical hallmarks are intestinal hamartomatous polyposis and melanin pigmentation of the skin and mucous membranes. In addition, PJS predisposes to cancer . The most common malignancies are small intestinal, colorectal, stomach and pancreatic adenocarcinomas. Other cancer types that probably occur in excess in PJS families include breast and uterine cervical cancer, as well as testicular and ovarian sex cord tumors. The relative risk of cancer may be as high as 18 times that of the general population, and the cancer patients' prognosis is reduced. Recently, the predisposing locus was mapped to 19p13.3 using a novel method. Subsequently, the causative gene was shown to be LKB1 (a.k.a. STK11), a serine/threonine kinase of unknown function. Although preliminary reports seem to suggest a minor role for LKB1 in sporadic tumorigenesis, further investigations are needed.
