Pancreatic islet cell suspensions from newborn rats; different preparation procedures,viability and (pro)insulin biosynthesis

Summary Islet cell suspensions were prepared from neonatal rat pancreatic islets. While mechanical disintegration results in a higher yield, cells prepared by trypsin treatment appear to be better preserved. Trypsin treatment of pancreatic islets during the cell preparation procedure does not influence the stimulation by glucose of (pro)insulin biosynthesis in freshly isolated cells.Investigations were carried out as a part of the HFR Diabetes mellitus and Fettstoffwechselstörungen supported by the Ministry of Health of the GDR.     

On SETAR non‐linearity and forecasting

We compare linear autoregressive (AR) models and self‐exciting threshold autoregressive (SETAR) models in terms of their point forecast performance, and their ability to characterize the uncertainty surrounding those forecasts, i.e. interval or density forecasts. A two‐regime SETAR process is used as the data‐generating process in an extensive set of Monte Carlo simulations, and we consider the discriminatory power of recently developed methods of forecast evaluation for different degrees of non‐linearity. We find that the interval and density evaluation methods are unlikely to show the linear model to be deficient on samples of the size typical for macroeconomic data. 在南苏鲁超高压变质带发现了原生高密度流体包裹体. 这种包裹体在超高压榴辉岩的石榴石中呈孤立和小群分布, 冷冻法确定包裹体的低共熔温度≤–52℃, 在重复冷冻-升温的特定阶段, 液相包裹体中出现气泡, 测得液-气均一温度≤–12.5℃, 由此得到流体成分属CaCl₂-NaCl-H₂O体系, 流体密度为1.27 g/cm³, 在相当于榴辉岩相峰期温度下的压力约为2.4 GPa, 接近超高压变质条件. 这类包裹体在榴辉岩的折返过程中与寄主矿物发生反应, 形成含水硅酸盐矿物, 并使其密度降低, 转变成气液多相包裹体.     

Crystal structure of a cholera toxin-related heat-labile enterotoxin from E. coli

Examination of the structure of Escherichia coli heat-labile enterotoxin in the AB5 complex at a resolution of 2.3A reveals that the doughnut-shaped B pentamer binds the enzymatic A subunit using a hairpin of the A2 fragment, through a highly charged central pore. Putative ganglioside GM1-binding sites on the B subunits are more than 20A removed from the membrane-crossing A1 subunit. This ADP-ribosylating (A1) fragment of the toxin has structural homology with the catalytic region of exotoxin A and hence also to diphtheria toxin.     

Requirement for a functional Rb-1 gene in murine development.

Human retinoblastomas can occur both as hereditary and as sporadic cases. Knudson's proposal that they result from two mutational events, of which one is present in the germ line in hereditary cases, has been confirmed by more recent molecular analysis, which has shown both events to involve loss or mutational inactivation of the same gene, RB-1 (ref. 2). RB-1 heterozygosity also predisposes to osteosarcoma, and RB-1 allele losses are seen in sporadic lung, breast, prostate and bladder carcinomas. RB-1 is expressed in most, if not all, tissues and codes for a nuclear phosphoprotein which becomes hypophosphorylated in the G0 growth arrest state and in the G1 phase of the cell cycle. To gain a further insight into the role of RB-1 we and other groups have generated mice carrying an inactivated allele of the homologous gene, Rb-1 (ref. 10), by gene targeting. We report here that young heterozygous mice do not appear abnormal and do not develop retinoblastoma at a detectable frequency. However, homozygous mutant embryos fail to reach term and show a number of abnormalities in neural and haematopoietic development. Broadly similar results are reported by the other groups.     

Participatory Model Construction and Model Use in Natural Resource Management: a Framework for Reflection

In this article we propose a framework which can assist analysts in their reflection on the requirements for a participatory modelling exercise in natural resource management. Firstly, we distinguish different types of formal models which may be developed, ranging from models that focus on (bio)physical mechanisms to models which also include the actors involved in the utilisation of the resource and the social mechanisms that co-determine actor behaviour. Secondly, we consider what different modes of stakeholder participation entail for model construction and use. Finally, we propose six different purposes for a modelling exercise (clarify arguments and values, research and analyse, design and recommend, provide strategic advice, mediate, and democratise), and highlight conditions that affect the appropriateness of stakeholder participation for each purpose. The framework does not provide a straightforward recipe for the selection of participatory modelling methods, but we expect that the systematic reflection it affords will help analysts to make appropriate choices while designing a modelling exercise.

Summary A survey of recent work on the theories of the peculiar mechanical behaviour of substances with rubber-like properties is given. All the theories are based on the assumption that these substances (including rubber itself) are composed of very long, flexible chainmolecules. Any segment of a chain is linked to two neighbouring segments by bonds similar to those which are present in typical solids and to all other neighbours by linkages like the bonds prevailing between the particles of liquids. The chain molecules are intermingled so as to form a kind of a felt.This state of aggregation is intermediate between the solid and the liquid; it is called the rubber-like state (état gommoïdal). Elasticity, viscosity and plastic flow as well as the influence of crystallization, vulcanization and degradation are discussed in terms of this theory.     

Structural and Synthetic Proposals for Lippialactone

Lippia javanica, a shrub of up to two meters in height, is used in South Africa against influenza, measles, rashes, stomach problems and headaches, depending on the traditional healer, and is therefore known as “fever tea” or “musudzungwane” in Tshivenda. Its essential oil （containing up to 75% piperitenone） has good insect repellent activity. In Zimbabwe and Malawi it is used mainly as a nerve tonic.
