Dystrophic mutation (dy2J) affecting regulation of lactate dehydrogenase (LDH) and pyruvate kinase (PK) in C57BL/6J mice

Summary The genotype difference (dystrophic vs nondystrophic) in the LDH isozymes is observed in kidney. These differences are evident only at birth and at early developmental stages (before the expression of dystrophic symptoms). The tissue specific genotype differences for PK are limited to the thigh muscle (M form) and heart (L form), after the onset of the condition. These differences may reflect the pleiotropic effect of the dy^2J locus during the temporal regulation of these and other enzymes implicated in muscular dystrophy (MD).This research was financed by a Natural Sciences and Engineering Research Council Canada grant to S.M.S.     

The antennae and mating behaviour ofDrosophila females

Summary The females ofDrosophila bipectinata andD. malerkotliana are able to discriminate between their own and alien males in the absence of antennae. Effect of hormones on adenyl cyclase activity of rabbit mammary gland in vitro

Ovarian hormones namely beta-estradiol and progesterone were observed to stimulate the activity of adenyl cyclase of the mammary gland from pregnant rabbit in vitro, unlike the lactating tissue where it was inhibited. On the other hand, non-ovarian hormones like hydrocortisone, prolactin and insulin did not have a similar effect on this enzyme.

A complementary transposon tool kit for Drosophila melanogaster using P and piggyBac

With the availability of complete genome sequence for Drosophila melanogaster, one of the next strategic goals for fly researchers is a complete gene knockout collection. The P-element transposon, the workhorse of D. melanogaster molecular genetics, has a pronounced nonrandom insertion spectrum. It has been estimated that 87% saturation of the approximately 13,500-gene complement of D. melanogaster might require generating and analyzing up to 150,000 insertions. We describe specific improvements to the lepidopteran transposon piggyBac and the P element that enabled us to tag and disrupt genes in D. melanogaster more efficiently. We generated over 29,000 inserts resulting in 53% gene saturation and a more diverse collection of phenotypically stronger insertional alleles. We found that piggyBac has distinct global and local gene-tagging behavior from that of P elements. Notably, piggyBac excisions from the germ line are nearly always precise, piggyBac does not share chromosomal hotspots associated with P and piggyBac is more effective at gene disruption because it lacks the P bias for insertion in 5' regulatory sequences.     

Coordination of the random asynchronous replication of autosomal loci

Random monoallelic expression and asynchronous replication define an unusual class of autosomal mammalian genes. We show that every cell has randomly chosen either the maternal or paternal copy of each given autosome pair, such that alleles of these genes scattered across the chosen chromosome replicate earlier than the alleles on the homologous chromosome. Thus, chromosome-pair non-equivalence, rather than being limited to X-chromosome inactivation, is a fundamental property of mouse chromosomes.     

Using Action Research to Improve Communication and Facilitate Group Work in a Multicultural Classroom: A South African Case Study

This study started with problems that my colleagues (and I) were experiencing with interaction and intercultural communication among students in our diverse classrooms. Educators were also experiencing difficulties in motivating students to work effectively in groups. The purpose of this paper was to seek solutions to these problems by exploring variations of the group oral assessment structure in a multilingual and multicultural context. Four phases of assessments were conducted at two tertiary institutions in South Africa using a combination of action research and a participatory approach. Not only did this study succeed in addressing the problems but it also revealed the added benefits of using an action research methodology.
