We carried out a genome-wide association study of IgA nephropathy, a major cause of kidney failure worldwide. We studied 1,194 cases and 902 controls of Chinese Han ancestry, with targeted follow up in Chinese and European cohorts comprising 1,950 cases and 1,920 controls. We identified three independent loci in the major histocompatibility complex, as well as a common deletion of CFHR1 and CFHR3 at chromosome 1q32 and a locus at chromosome 22q12 that each surpassed genome-wide significance (P values for association between 1.59 × 10?2? and 4.84 × 10?? and minor allele odds ratios of 0.63-0.80). These five loci explain 4-7% of the disease variance and up to a tenfold variation in interindividual risk. Many of the alleles that protect against IgA nephropathy impart increased risk for other autoimmune or infectious diseases, and IgA nephropathy risk allele frequencies closely parallel the variation in disease prevalence among Asian, European and African populations, suggesting complex selective pressures.  相似文献

Estimation of time since death is one of the challenges in forensic science. There are many approaches to estimate the postmortem interval, including both physical and thanatochemistry methods. Decomposition is triggered by a process called autolysis, which induces destructive changes in the cell leading to cell death. Based on the process of cell death signaling, this study analyzed the early postmortem interval (2–8 h since death) using the study of the mRNA expression of Fas Ligand (FasL) and phosphatase and tensin homologue deleted on chromosome 10 (PTEN) by Quantitative-PCR. Results of the study indicate a time-dependent increase in the mRNA levels of both proteins up until 6 h after death. Using a regression analysis in these first 6 h, a positive linear correlation was found between the mRNA expression of these proteins and the time since death. Since PTEN and FasL are implicated in signaling pathways, both proteins are potential candidates to analyze the time since death in time intervals of 6 h or less. Further research is needed to find additional cell death markers and expand the time period for time since death estimation.  相似文献

The protocadherins (Pcdhs), which make up the most diverse group within the cadherin superfamily, were first discovered in the early 1990s. Data implicating the Pcdhs, including ~60 proteins encoded by the tandem Pcdha, Pcdhb, and Pcdhg gene clusters and another ~10 non-clustered Pcdhs, in the regulation of neural development have continually accumulated, with a significant expansion of the field over the past decade. Here, we review the many roles played by clustered and non-clustered Pcdhs in multiple steps important for the formation and function of neural circuits, including dendrite arborization, axon outgrowth and targeting, synaptogenesis, and synapse elimination. We further discuss studies implicating mutation or epigenetic dysregulation of Pcdh genes in a variety of human neurodevelopmental and neurological disorders. With recent structural modeling of Pcdh proteins, the prospects for uncovering molecular mechanisms of Pcdh extracellular and intracellular interactions, and their role in normal and disrupted neural circuit formation, are bright.  相似文献

本文探讨了醋制泥鳅对小鼠运动耐力的影响机制.将24只健康雄性ICR小鼠分为安静对照组(A组)、运动训练组(T组)、泥鳅运动组(V组).T组与V组小鼠游泳训练三周后进行力竭游泳,24 h后取材计算心系数,HE染色光镜下观察心肌组织,RT-PCR法检测心肌肝细胞生长因子(HGF)和肌球蛋白重链(MHC)基因的表达.结果表明:V组小鼠的力竭游泳时间非常显著地长于T组(P0.01);T组的心系数显著高于A组和V组(P0.05),并可见心肌细胞界限模糊、心肌纤维断裂和间质水肿,而A组与V组无明显病理表现;与A组比较,T组的HGF、β-MHC mRNA表达显著增高(P0.05)和α/β-MHC mRNA比值显著降低(P0.05),V组的HGF、MHC mRNA和α/β-MHC mRNA值差异不显著(P0.05);V组的α/β-MHC mRNA值显著高于T组(P0.05).总之,醋制泥鳅能明显提高负荷游泳训练小鼠的运动耐力,与减轻心肌损害和促进心肌MHC mRNA表达比值的稳定关系密切.  相似文献

本实验以两株酵母菌为研究对象,通过正交设计,验证了原生质体制备过程中各因素对酵母细胞活力的影响.研究发现,在原生质体制备过程中渗透压稳定剂的种类、浓度对于原生质体的形成率和活力有着重要的影响.离子型的渗透压稳定剂如KCl可以提高细胞的形成率,而醇类稳定剂如山梨醇对于酶解后原生质体活力的保持有着较好的效果.  相似文献
