Karyotypes of the long-tailed vole ( Microtus longicaudus ) in isolated mountain ranges of the American Southwest

The long-tailed vole ( Microtus longicaudus ) exhibits karyotypic variation in western North America. In the American Southwest, the species has a disjunct, relict distribution. However, karyotypes obtained from 20 specimens in 5 isolated mountain ranges (White Mountains, Arizona; Mount Taylor and the Sangre de Cristo, Jemez, and Mogollon mountains, New Mexico) exhibited no variation. These and previously reported karyotypes from the region had a diploid number (2n) of 56 and fundamental number (FN) of 84, which is the most commonly reported karyotype for the species.     

Low density lipoprotein receptor-related protein 1 couples β1 integrin activation to degradation

Low density lipoprotein receptor-related protein (LRP) 1 modulates cell adhesion and motility under normal and pathological conditions. Previous studies documented that LRP1 binds several integrin receptors and mediates their trafficking to the cell surface and endocytosis. However, the mechanism by which LRP1 may regulate integrin activation remains unknown. Here we report that LRP1 promotes the activation and subsequent degradation of β1 integrin and thus supports cell adhesion, spreading, migration and integrin signaling on fibronectin. LRP1 interacts with surface β1 integrin, binds the integrin activator kindlin2 and stimulates β1 integrin–kindlin2 complex formation. Specifically, serine 76 in the LRP1 cytoplasmic tail is crucial for the interaction with kindlin2, β1 integrin activation and cell adhesion. Interestingly, a loss of LRP1 induces the accumulation of several integrin receptors on the cell surface. Following internalization, intracellular trafficking of integrins is driven by LRP1 in a protein kinase C- and class II myosin-dependent manner. Ultimately, LRP1 dictates the fate of endocytosed β1 integrin by directing it down the pathway of lysosomal and proteasomal degradation. We propose that LRP1 mediates cell adhesion by orchestrating a multi-protein pathway to activate, traffic and degrade integrins. Thus, LRP1 may serve as a focal point in the integrin quality control system to ensure a firm connection to the extracellular matrix.

Lead isotopes reveal bilateral asymmetry and vertical continuity in the Hawaiian mantle plume

The two parallel chains of Hawaiian volcanoes ('Loa' and 'Kea') are known to have statistically different but overlapping radiogenic isotope characteristics. This has been explained by a model of a concentrically zoned mantle plume, where the Kea chain preferentially samples a more peripheral portion of the plume. Using high-precision lead isotope data for both centrally and peripherally located volcanoes, we show here that the two trends have very little compositional overlap and instead reveal bilateral, non-concentric plume zones, probably derived from the plume source in the mantle. On a smaller scale, along the Kea chain, there are isotopic differences between the youngest lavas from the Mauna Kea and Kilauea volcanoes, but the 550-thousand-year-old Mauna Kea lavas are isotopically identical to Kilauea lavas, consistent with Mauna Kea's position relative to the plume, which was then similar to that of present-day Kilauea. We therefore conclude that narrow (less than 50 kilometres wide) compositional streaks, as well as the larger-scale bilateral zonation, are vertically continuous over tens to hundreds of kilometres within the plume.     

Place de la prolyl hydroxylase dans la synthèse du collagène de cellules KB

Summary KB cells, which synthetized collagen at a low rate, shown a prolyl hydroxylase activity at the same rate that fibroblast. The relationship between collagen synthesis and prolyl hydroxylase activity in these cells was discussed.

Genetics of allopatric populations of the montane vole ( Microtus montanus ) and Mogollon vole ( Microtus mogollonensis ) in the American Southwest

Variation at 21 allozyme loci and 10 restriction enzymes within the mitochondrial DNA control region was assessed among 3 allopatric populations of montane vole ( Microtus montanus ) in the American Southwest. Among populations of M. montanus , the population from the White Mountains, Arizona, was most genetically divergent. Genetic and biogeographic evidence supported recognition of the White Mountains population as a distinct subspecies, M. m. arizonensis . Results also supported recognition of the Mogollon vole ( Microtus mogollonensis ) as distinct from the Mexican vole ( Microtus mexicanus ).
