Summary A biphasic dose-response pattern is generated by the isoquinoline, 3-carboxysalsolinol, in analgesia tests conducted in mice. Carbidopa pretreatment enhances this effect, as well as the morphine-induced analgesic increase by 3-carboxysalsolinol. Naloxone blockade of all of these responses suggests an interaction of the alcohol-based isoquinoline with central opiate receptors.We gratefully acknowledge the receipts of drugs from Merck and Co., West Point, Pennsylvania, and Endo Laboratories, Garden City, New York. The branchial gland of the dibranchiate cephalopods is described as the site of haemocyanin synthesis. Because there is no equivalent to this organ in tetrabranchiate cephalopods the localization of haemocyanin synthesis remained unknown for a long time. In this study we could confirm the conclusions from prelimnary investigations concerning the copper content of the midgut gland ofNautilus, which gave the first indications for a possible localization of haemocyanin synthesis in this organ. We developed a polyclonal antibody againstNautilus haemocyanin, tested its specificity, and used it on ultra-thin sections of the tissue of the midgut gland. It could be shown that there is a clear imunogold precipitation only on the triangular basal cells in the terminal alveoli. All the other types of cell in this organ were free of any immunoreactivity. It can be supposed that the triangular basal cells in the terminal alveoli of the midgut gland are the sites of haemocyanin synthesis inNautilus.  相似文献

These experiments systematically investigated ethanol preference in both the C57Bl/6N and C57Bl/6J mice utilizing three-choice 2-bottle preference test. In addition, these sublines were evaluated for whole brain methionine-enkephalin levels, which were significantly lower in C57Bl/6J mice (alcohol preferring) compared to C57Bl/6N mice (alcohol non-preferring). This finding supports the involvement of the peptidyl opiates in ethanol seeking behavior.  相似文献

The thymus is organized into medullary and cortical zones that support distinct stages of T-cell development. The formation of medulla and cortex compartments is thought to occur through invagination of an endodermal epithelial sheet into an ectodermal one at the third pharyngeal pouch and cleft, respectively. Epithelial stem/progenitor cells have been proposed to be involved in thymus development, but evidence for their existence has been elusive. We have constructed chimaeric mice by injecting embryonic stem (ES) cells into blastocysts using ES cells and blastocysts differing in their major histocompatibility complex (MHC) type. Here we show that the MHC class-II-positive medullary epithelium in these chimaeras is composed of cell clusters, most of which derive from either embryonic stem cell or blastocyst, but not mixed, origin. Thus, the medulla comprises individual epithelial 'islets' each arising from a single progenitor. One thymic lobe has about 300 medullary areas that originate from as few as 900 progenitors. Islet formation can be recapitulated after implantation of 'reaggregated fetal thymic organs' into mice, which shows that medullary 'stem' cells retain their potential until at least day 16.5 in fetal development. Thus, medulla-cortex compartmentalization is established by formation of medullary islets from single progenitors.  相似文献
