Eya1-deficient mice lack ears and kidneys and show abnormal apoptosis of organ primordia. 总被引:1,自引:0,他引:1
Haploinsufficiency for human EYA1, a homologue of the Drosophila melanogaster gene eyes absent (eya), results in the dominantly inherited disorders branchio-oto-renal (BOR) syndrome and branchio-oto (BO) syndrome, which are characterized by craniofacial abnormalities and hearing loss with (BOR) or without (BO) kidney defects. To understand the developmental pathogenesis of organs affected in these syndromes, we inactivated the gene Eya1 in mice. Eya1 heterozygotes show renal abnormalities and a conductive hearing loss similar to BOR syndrome, whereas Eya1 homozygotes lack ears and kidneys due to defective inductive tissue interactions and apoptotic regression of the organ primordia. Inner ear development in Eya1 homozygotes arrests at the otic vesicle stage and all components of the inner ear and specific cranial sensory ganglia fail to form. In the kidney, Eya1 homozygosity results in an absence of ureteric bud outgrowth and a subsequent failure of metanephric induction. Gdnf expression, which is required to direct ureteric bud outgrowth via activation of the c-ret Rtk (refs 5, 6, 7, 8), is not detected in Eya1-/- metanephric mesenchyme. In Eya1-/- ear and kidney development, Six but not Pax expression is Eya1 dependent, similar to a genetic pathway elucidated in the Drosophila eye imaginal disc. Our results indicate that Eya1 controls critical early inductive signalling events involved in ear and kidney formation and integrate Eya1 into the genetic regulatory cascade controlling kidney formation upstream of Gdnf. In addition, our results suggest that an evolutionarily conserved Pax-Eya-Six regulatory hierarchy is used in mammalian ear and kidney development. 相似文献

Summary After two-stage nephrectomy in rats the potassium concentration in the red blood corpuscles (RBC) decreases from 10.6 ± 0.3 to 5.5 ± 0.3 mEq per 100 ml of RBC within 48 h. The decrease is accompanied by a much smaller increase in the plasma potassium concentration; the hyperkaliemia in nephrectomized rats is less pronounced than in nephrectomized dogs or anuric humans. Na+ in RBC increases by about 44% after nephrectomy; while there is only a very slight decrease of Na+ in plasma. Plasma chlorides drop from 10.92 ± 0.08 mEq/100 ml of plasma to 6.00 ± 0.81 mEq % within 48 h after nephrectomy. RBC chlorides tend to increase again after an initial drop from 5.22 ± 0.07 to 3.82 ± 0.90 mEq% within the first 8 h. 相似文献

Chromatin packaging in mammalian spermatozoa requires an ordered replacement of the somatic histones by two classes of spermatid-specific basic proteins, the transition proteins and the protamines. Temporal expression of transition proteins and protamines during spermatid differentiation is under translational control, and premature translation of protamine 1 leads to precocious nuclear condensation and sterility. We have previously suggested that the double-stranded (ds) RNA binding protein Prbp (encoded by the gene Tarbp2) functions as a translational regulator during mouse spermatogenesis. Here we show that Prbp is required for proper translational activation of the mRNAs encoding the protamines. We generated mice that carry a targeted disruption of Tarbp2 and determined that they were sterile and severely oligospermic. Using immunohistological analysis, we determined that the endogenous Prm2 mRNA and a reporter mRNA carrying protamine 1 translational-control elements were translated in a mosaic pattern. We showed that failure to synthesize the protamines resulted in delayed replacement of the transition proteins and subsequent failure of spermiation. The timing of Prbp expression suggests that it may function as a chaperone in the assembly of specific translationally regulated ribonucleoprotein particles. 相似文献

Neonatal heart cells populated collagen gels show rhythmic contractions which can be recorded optically and electrically. Optical recordings revealed two populations of rhythmically contracting gels: 1) highly coherent contracting collagen gels with normally distributed contraction interval times and contraction amplitudes, and 2) irregularly contracting gels with a multi-modal distribution of contraction interval times and amplitudes. The irregularly contracting gels were shown to be semi regular, which means that a short contraction interval was preferentially followed by another short interval. The volume of the collagen gel during the contraction decreased, and our calculations indicate that the myocytes expel 3–10 times their own volume from the gel. Changes in electrical potential were observed depending on the location of the electrodes. These electrical, ECG-like changes in potential were maximal when one electrode was placed in the centre and the other at the edge of the gel. The results of this study indicate that myocyte-populated collagen gels are a very promising system for studies of electrophysiology and coherent contractions. 相似文献
