针对Exendin-4的螺旋结构特点, 采用去掉螺旋序列、 用3个连续Ala替换螺旋以及Gln13替换为Tyr13的方式对N端α-螺旋及C端α-螺旋进行改造, 设计出4个结构模拟物. 通过圆二光谱和荧光光谱进行结构分析, 结合模拟物生物活性实验, 分析Exendin-4螺旋结构与生物活性的关系. 结果表明:  Exendin-4的N端螺旋比C端螺旋对生物活性影响更大; 在抗二肽基肽酶(DPPⅣ酶)的稳定性实验中, C端螺旋比N端螺旋具有更好维系Exendin-4稳定性的作用; 螺旋结构中Tyr13可作为提高Exendin-4生物活性的重要优化位点.  相似文献

马蹄寺石窟群地处河西走廊中部,气候条件十分恶劣。由于常年受到温度、湿度、酸碱盐等作用的影响,石窟岩体性能出现了不同程度的劣化现象。通过开展室内模拟试验,研究以上多种恶劣环境的循环作用对于岩体抗压强度、波速、质量的影响规律,同时使用X射线衍射仪(X-ray diffraction,XRD)、X射线荧光光谱(X-ray fluorescence,XRF)和扫描电子显微镜(scanning electron microscope,SEM)对循环作用过程中样品的矿物组分、元素含量和微观形貌进行分析研究。研究结果表明,循环作用使样品中的长石类矿物和方解石含量降低,黏土类矿物含量增加,胶结程度下降,孔隙逐渐发展成为横向微裂隙,导致样品的性能出现不同程度的劣化现象。循环作用对岩体性能的影响程度从大到小依次为冻融循环、耐盐循环、耐碱循环、耐酸循环和温湿循环。  相似文献

Proinsulin C-peptide is known to bind specifically to cell membranes and to exert intracellular effects, but whether it is internalized in target cells is unknown. In this study, using confocal microscopy and immunostained or rhodamine-labeled peptide, we show that C-peptide is internalized and localized to the cytosol of Swiss 3T3 and HEK-293 cells. In addition, transport into nuclei was found using the labeled peptide. The internalization was followed at 37°C for up to 1 h, and was reduced at 4°C and after preincubation with pertussis toxin. Hence, it is concluded to occur via an energy-dependent, pertussis toxin-sensitive mechanism and without detectable degradation within the experimental time course. Surface plasmon resonance measurements demonstrated binding of HEK-293 cell extract components to C-peptide, and subsequent elution of bound material revealed the components to be intracellular proteins. The identification of C-peptide cellular internalization, intracellular binding proteins, absence of rapid subsequent C-peptide degradation and apparent nuclear internalization support a maintained activity similar to that of an intracrine peptide hormone. Based on the researches on rock type, the mode of occurrence, diagnostic minerals and creatures, the sedimentary geochemistry and organic facies of Chinese marine source rocks from wells and outcrops, and on the research findings of developmental modes of foreign marine source rock, the authors consider that it is impossible to objectively make clear the formation mechanism of hydrocarbon source rock with high organic matter abundance by either single mode of preservation or high organic matter productivity. According to the Chinese geological features, the formation mechanism of the Neoproterozoic-Lower Paleozoic marine source rock is generalized into four modes, namely, thermal water activity-upwelling flow-anoxic; carbonate gentle slope-upwelling flow; xerothermic climate-brine euxinic milieu; and humid climate-retained euxinic milieu; as the Lower Cambrian undercompensation basin organic facies (the Tarim Basin, South China and southwestern margin of North China), carbonate gentle lime mud bound organic facies (the Upper Ordovician in Tazhong region of the Tarim Basin and the Lower Silurian in the Upper Yangtze Platform), the Middle Cambrian evaporation laggon organic facies (the Tarim Basin and the Upper Yangtze Platform), enclosed undercompensation terrigenous bay organic facies (the Middle-Upper Ordovician in the west of the Tarim Basin, the Lower Silurian Long- maxi Formation in the Upper Yangtze platform). Chinese marine sedimentations with lower organic matter abundance are generalized into two modes of consumption-dilution mode of open epicontinental sea and depletion-dilution mode of supercompensation basin.  相似文献

Generally, there are some anhydrites in carbonate reservoir, as H2S is also familiar in carbonate oil and gas reservoirs. Nowadays, natural gas with high H2S concentration is usually considered as TSR origin, so there is close relationship between H2S and anhydrite. On the contrary, some carbonate rocks with anhydrite do not contain H2S. Recently, researches show that H2S is only a necessary condition of H2S formation. The reservoir porosity, sulfate ion content within formation water, reservoir temperature, oil/gas and water interface, hydrocarbon and some elements of reservoir rock have great controlling effects on the TSR occurrence. TSR deoxidizes hydrocarbon into the acidic gas such as H2S and CO2, and the H2S formation is controlled by TSR occurrence, so the relationship among reaction room, the contact chance of sulfate ion and hydrocarbon, the reservoir temperature has great influence on the TSR reaction. H2S has relatively active chemical quality, so it is still controlled by the content of heavy metal ion. Good conditions of TSR reaction and H2S preservation are the prerequisite of H2S distribu- tion prediction. This paper builds a predictive model based on the characteristic of natural gas reservoir with high H2S-bearing. In the porosity reservoir with anhydrite, the formation water is rich in sulfate and poor in heavy metal ion. Oil and gas fill and accumulate in the gas reservoir with good preservation conditions, and they suffered high temperature later, which indicates the profitable area of natural gas with high H2S-bearing.  相似文献

分析了元宝山露天煤矿水文地质特征与基岩抽水试验情况,提出了元宝山露天矿基岩疏干设计方案,通过该方案的实施,解决了元宝山露天煤矿在剥离工作中出现陷车、陷镐、粘车、粘皮带这一难题,使剥离速度大大加快,缓解了采剥失调、生产接续紧张局面。不仅创造了可观的直接效益,而且间接效益更大,同时为今后的基岩疏干提供了宝贵经验。  相似文献
