The effect of guanethidine treatment of testicular blood flow and testosterone production in rats

Testicular blood flow was measured by means of Xenon-133 clearance in control rats and rats treated with guanethidine for 3 weeks. Plasma and testis testosterone concentrations were also measured, and the effect of hCG-treatment was examined. No difference in testicular blood flow between the control group and the guanethidine-treated group was found. However, in guanethidine-treated rats, plasma and testis testosterone concentrations after hCG-treatment were significantly decreased. The results may indicate that adrenergic nerves are involved in the regulation of Leydig cell function.     

Capillary blood flow in the testes and testosterone secretion in the starved rat

Summary The average capillary blood flow in the testes was found to be 181 l/min/g testis tissue (n=19) in rats starved for 5 days and 273 l/min/g (n=18, p<0.01) in the control group. Plasma testosterone was significantly decreased in the starved animals (1.00±0.06 ng/ml vs 5.43±0.63 ng/ml). When starved and control rats were stimulated with human chorion gonadotropin, testosterone values in plasma were greatly increased in both groups. The capillary blood flow was not altered. The data indicate that human chorion gonadotropin can stimulate testosterone production in the starved rat without influencing the reduced capillary blood flow.     

Androgen levels in the rete testis fluid during sexual development

Summary The concentrations of testosterone (T) and 5-dihydrotestosterone were measured in fluid collected from the rete testis of immature and adult rats. The results indicate that adult levels of T are attained in the seminiferous tubules much earlier than in the peripheral circulation.This work was performed at the Worcester Foundation for Experimental Biology, Shrewsbury, Massachusetts, USA and supported by the National Institutes of Health through grants HD12641 and HD12642. Plasma steroid and protein hormone concentrations in patients with benign prostatic hypertrophy and in normal men.

Serum levels of testosterone, FSH, LH and prolactin were measured by RIA in patients with BPH (benign prostatic hypertrophy) and in control men. The testosterone concentrations in the BPH group were significantly lower than that of the control group. Serum concentrations of FSH, LH, prolactin and estradiol, did not differ significantly in the 2 groups.

Effect of immunization of male rabbits against androstenedione

Summary After active immunization of male rabbits against androstenedione and with increasing antibody titre the concentration of androstenedione and testosterone in peripheral blood increased more than 1000-fold resp. 20-fold above values of control animals. On histological examination of the testes a marked Leydig's cell hyperplasia was found, though there was no difference in testis weight.     

Radioimmunoassay of plasma progesterone, testosterone, total estrogen and immunoreactive gonadotropin in the nesting and non-nesting green sea turtle, Chelonia mydas (L.).

Plasma progesterone, testosterone, total estrogens and immunoreactive gonadotropin were measured in nesting and non-nesting sea turtles, Chelonia mydas. Progesterone and gonadotropin concentrations were significantly higher in nesting than in non-nesting turtles, testosterone was not significantly different in either group and total estrogens appeared to be slightly in the nesting group.     

Evolution of testosterone and dihydrotestosterone in plasma, testicle and ovary of the guinea pig throughout fetal life]

High testosterone concentrations were found in the plasma of male fetus at the stage of sexual differentiation. Then the fetal testis is the principal source of circulating testosterone; its action on the Wolffian duct would occur by the systemic pathway. In female fetus, androgens were synthesized by the gonad and the relatively high levels found in the plasma have probably another source.

Circadian variation of serum testosterone in the adult male rat with a late morning acrophase

Summary Serum testosterone concentrations were measured in adult male Sprague-Dawley rats. The relationship between high and low trait psychological stress,serum testosterone,and serum cortisol

Summary Psychological stress, as measured by the parameters of trait anxiety, hostility, and depression, was compared in the group of males age 30–55 with serum levels of testosterone. Those males who were classified as high psychological stress had significantly lower testosterone levels than did their low stress counterparts. However, comparison of their serum cortisol values revealed similar concentrations in both groups.     

Plasma, testicular, and adrenal cortex levels of androgens in the perinatal period and determination of neonatal hypertestosteronemia in guinea pigs]

The perinatal period is characterized in the male faetus and new-born Guinea-Pig by a prenatal increase in testis testosterone and by a neonatal peak in plasma testosterone. This transient rise in plasma testosterone is testis dependent and light dependent at birth.

Glomerular filtration rate (GFR), cardiac output, regional blood flow and kidney weight were measured in alloxan and streptozotocin diabetic rats at different times after the administration of diabetogen. A high GFR was found together with increased kidney weight and reduced blood flow.

The effect of gonadotropic hormones and the fetal hypophysis on testosterone production by the testis of 18 day mouse fetuses in organ culture]

The production of testosterone (measured by radioimmuno-assay) by the 18-day-old mouse fetal testis may be stimulated specifically by ovine LH (1 ng, p less than 0.005) and HCG in organ culture. A stimulation by FSH is observed only with high doses (10 mug, p less than 0.0005). Prolactin and ACTH have no effect. Age-matched fetal pituitaries increase significantly the testosterone production in the culture medium (p less than 0.0005).     

链霉素诱导的2型糖尿病大鼠模型的肾脏功能变化

目的通过观察链霉素诱导的2型糖尿病大鼠模型肾脏功能的变化,以探讨此种模型是否适用于糖尿病肾损害的研究。方法采用链脲佐菌素(streptozotocin,STZ)辅以高脂饮食建立2型糖尿病大鼠模型,筛选造模成功的大鼠为糖尿病组,并与正常大鼠比较,造模后8周观察两组大鼠血糖、血尿素氮(blood urea nitrogen,BUN)、血肌酐(creatinine,CR)、血糖基化终末产物(Advanced glycation end products,AGE)、尿微量白蛋白、肾重指数的变化以及AGE受体(Receptor of AGE,RAGE)、蛋白激酶C(protein kinase C,PKC)、蛋白激酶A(protein kinase A,PKA)在肾脏的表达变化。结果糖尿病组的血糖、血尿素氮、血肌酐、血AGE、尿微量白蛋白以及肾重指数均明显高于正常组(P0.05),RAGE、PKC的mRNA以及蛋白表达水平均较正常组高(P0.05),PKA的mRNA和蛋白表达水平则较正常组低(P0.05)。结论 STZ诱导的2型糖尿病大鼠模型在造模后第8周肾脏功能存在明显的损害,并且导致肾脏损害的上游重要介质RAGE、PKC、PKA在第8周都有着明显的变化。此种模型能够用于糖尿病肾损害的研究,并且是观察中早期糖尿病肾病的较好的模型。     

Inhibition of post-hemicastration hypertrophy of the remaining testis by testosterone or aqueous testicular extract treatments in the young rat]

In the 10 day old hemicastrated Rat daily injections of testosterone (total doses of 0.04 to 4 mg/Rat) or of proteinaceous extract of Rat testis (4 mg/Rat) inhibit compensatory hypertrophy of the remaining testis. Both testosterone and testicular proteinaceous extract could be involved in the growing processes of the testis, locally or by means of modification of gonadotropin secretion. Results have been analysed by covariance analysis to eliminate the influence of body weight variations.     

Effect of light on variations in blood testosterone in the ram: demonstration of a photosensitive phase in diurnal rhythm]

6 groups each of 5 adult rams were subjected in light controlled pens to a pretreatment of normal variations in daylength during May and June. Then one group followed the normal daylength and the other five different "skeleton photoperiods" of 8 hrs of light per day consisting of: 7 hrs continuous light (7 L) + 1 hr light (1 L) situated at different times of the dark period (D) according to groups. Blood samples were taken once each week and hourly during one 24 hr period in June, July and September. Peripheral plasma testosterone measured by radio-immunoassay was used as an index of testicular activity. There was increase in the level of testosterone resuting from a rise in the number of peak release during the 24 hrs. In the different light treatments tested, only the treatment (7L+9D+1L)+7D) stimulated testosterone secretion with the pattern of testosterone of this group being similar to that of the control group. This indicates that there exists a photosensitive phase at 16 to 17 hrs after the beginning of the principal light (the subjective dawn).     

解聚复肾宁对糖尿病肾病大鼠TNF—OL、MCP-1的影响

目的观察解聚复肾宁（Jiejufushenning，JJFSN）对糖尿病（Diabetes mellitus，DM）大鼠肾组织肿瘤坏死因子-α（Tumorneerosisfactor-alpha，TNF-α）、单核细胞趋化蛋白-1（Monocyte chemoattractant protein-1，MCP-1）和血清TNF-α的影响，探讨其肾保护作用机制。方法腹腔注射链脲佐菌素（Streptozotocin，STZ）建立SD大鼠DM模型，将成模DM大鼠随机分为3组：DM模型组（B组）、JJFSN组（C组）、厄贝沙坦（Irbesartan）组（D组），另设正常对照组（A组）。采用相应干预措施处理12周。常规方法测定12周末各组大鼠空腹血糖（FBG）、血尿素氮（BUN）、血肌酐（Scr）、肾重／体重（KW／BW）、24h尿蛋白（24huFro）；放免法测血清TNF-α含量：免疫组织化学方法测肾组织TNF—Ot、MCP-1的表达；PAS染色评估细胞外基质；电镜观察肾组织超微结构改变。结果与A组相比．B组大鼠FBG、BUN、Scr、KW／BW、24huPro及血清TNF-α升高（P〈0．05），肾组织TNF—Ot、MCP-1表达明显增高（P〈0．05）；肾组织超微结构明显异常；C组和D组上述指标显著改善（P〈0．05），肾组织超微结构异常改善。结论竹FSN能延缓DM大鼠肾损害进程，其机制可能与抑制TNF-α、MCP-1上调有关。     

Reversible reduction in bone blood flow in streptozotocin-diabetic rats

Tibial growth and blood flow were both found to be markedly reduced in anaesthetised streptozotocin-diabetic rats compared to controls. Insulin treatment restored tibial growth to approximately control values and increased tibial blood flow to above control values. The observations are likely to be related to reduced bone turnover in uncontrolled diabetes.
