Effects of cadmium on the immune system of mice

Summary Chronic oral exposure of mice to Cd⁺⁺ inhibits cell-mediated immunity of delayed type hypersensitivity induced by sheep red blood cells (SRBC). No effect was detected on humoral immune response to SRBC. Spleen cells derived from mice exposed to Cd⁺⁺ showed in vitro enhanced response to T and B cell mitogens. Effect of NPV of the armywormMythimna (Pseudaletia) separata on the silkwormBombyx mori

Summary Nuclear polyhedrosis virus (NPV) material of the armywormMythimna (Pseudaletia) separata was administered in the form of PIBs and free virus rods to the larvae of silkwormBombyx mori. Routes used for administration were topical, intrahaemocoelic and oral. The larvae were treated with following concentrations: 10×10⁵ PIBs/L, 10×10⁶ PIBs/L, 10×10⁷ PIBs/L, 10×10⁸ PIBs/L. In all the 3 experiments, the larvae showed neither any signs and symptoms, nor mortality due to polyhedrosis. Thus it appears that the NPV ofM. (P.) separata is safe for the silkwormB. mori.Acknowledgments. Loss of antibody production accompanied by chromosome loss in a cloned hybrid line secreting antibodies to sheep red blood cells

Summary Somatic cell hybrids between Sp2/O-Ag14 mouse myeloma cells and lymphocytes derived from BALB/c mice hyperimmunized with sheep red blood cells (SRBC) were produced. One hybrid producing IgG₁ antibody to SRBC was selected, cloned twice and subsequently transferred to BALB/c mice. After a number of transfers it was found that the antibody titer in ascitec fluid gradually decreased. Cytogenetic analysis revealed gradual chromosome loss in the hybrid clone, which produced progressively less antibody.     

Phosphoramidon enhances allatostatin-mediated inhibition of juvenile hormone biosynthesis in the corpora allta of the cockroach,Diploptera punctata

Use of the enkephalinase inhibitor phosphoramidon in the in vitro radiochemical assay for juvenile hormone biosynthesis enhanced allatostatin-mediated inhibition of hormone production by corpora allata of the cockroach,Diploptera punctata. Significant increases in inhibition in day 2 virgin female CA by AST 1 (at 10^–7 M) and AST 4 (10^–8–10^–7 M) were observed in the presence of phosphoramidon (10^–5M or greater). No significant increases in inhibition were seen in CA from day 6 mated females with AST 4 (10^–9–10^–7M) and phosphoramidon combined. Phosphoramidon alone had no effect on JH biosynthesis. Analysis of allatostatin content of the CA, as determined by ELISA, revealed that addition of phosphoramidon to the medium increased the endogenous allatostatin conten in CA of virgin and mated females. The similarity in primary structure between allatostatins and enkephalin-like peptides and their similar distribution makes it probable that phosphoramidon acts by preventing breakdown of allatostatins within the CA.     

Inhibition and sex specific induction of spawning by serotonergic ligands in the zebra musselDreissena polymorpha (Pallas)

Serotonin (5-hydroxytryptamine, 5-HT) stimulates spawning in the zebra mussel (Dressena polymorpha), a macrofouling European bivalve that has recently invaded North America. To develop methods of controlling zebra mussel spawning, two vertebrate serotonin antagonists, methiothepin and metergoline, known to bind with high affinity to snail 5-HT receptors, were tested for their ability to block 5-HT-induced spawning in zebra mussels. Methiothepin inhibited 5-HT-induced spawning at concentrations as low as 10^–6 M. Metergoline (10^–4 M) inhibited 5-HT-induced spawning; however, at lower concentrations (10^–8 to 10^–5 M), metergoline by itself significantly induced spawning in male, but not female zebra mussels. Metergoline (10^–5 M)-induced male spawning was inhibited by 10^–5 M methiothepin. Thus, methiothepin is the most effective inhibitor and metergoline the most powerful inducer of spawning yet tested in zebra mussels.     

Effect of strychnine on the electroretinogram of the isolated rabbit retina

Zusammenfassung Strychnin in einer Konzentration von 3×10^–6 bis 3,5×10^–5 M/l verursacht eine vorübergehende Vergrösserung derb-Welle des ERG der isolierten Kaninchennetzhaut, in einer Konzentration von 7×10^–5 bis 7×10^–4 M/l verkleinert es dieb-Welle. Aus den Ergebnissen wird geschlossen, dass die Substanz zumindest teilweise schon auf die Schicht des zweiten Neurons wirkt.     

Different marrow cell number requirements for the haemopoietic colony formation and the cure of the W/Wv anemia

Summary The lowest cell number in the normal marrow transplant, which allows the cure of W/W^v anemia was found to be between 10⁴ and 10⁵. This exceeds by several times the lowest cell number necessary for the haemopoietic colony formation. Therefore, either the colony forming cell is not the haemopoietic stem cell but rather its progeny, or this cell requires an aid from some other cells to exert its activity.Acknowledgments. Growth ofNostoc muscorum mutants in the presence of diuron (DCMU) and L-methionine-DL-sulfoximine

Summary Diuron (DCMU) is inhibitory to the photoautotrophic and photoheterotrophic growth of the N₂-fixing blue-green algaNostoc muscorum at concentrations of 1.0×10^–5 M and 2.0×10^–5 M, respectively. A mutant of this organism resistant to 5.0×10^–5 M DCMU under its photoheterotrophic growth conditions, with the ability to utilize DCMU as a carbon and nitrogen source for growth, and complete inability to grow photoautotrophically has been isolated. With the apparent defect in its photosynthetic ability, it is suggested that theDCMU ^r mutant lacks the step inhibited by 1.0×10^–5 M DCMU, and metabolizes DCMU by an existing enzyme system in the absence of such inhibition. That this enzyme may be glutamine synthetase (GS) is explained with the help of a L-methionine-DL-sulfoximine (MSO)-resistant mutant ofN. muscorum which is able to grow faster with 2.0×10^–5 DCMU and is known to contain an altered GS.Thanks are due to the Council of Scientific and Industrial Research, CSIR Complex, Govt. of India, New Delhi-110012, for appointing the author to the Scientists' Pool for undertaking researches on the physiological and genetic controls of nitrogen metabolism in blue-green algae, a part of which is presented in this literature.     

In vitro effects of methionine-enkephalin,somatostatin and insulin on cultured gonadal cells of the snailHelix aspersa

Isolated snail gonadal cells were cultured in the presence of synthetic neuropeptides in order to determine the subsequent effect of these substances on gonadal synthetic activities. Gonadal cells were incubated for 24 h in concentrations of methionine-enkephalin, somatostatin and insulin ranging from 10^–4 M to 10^–9 M, in medium 199 supplemented with 6% Ultroser G. Synthesis of DNA and protein by the cultured cells were simultaneously estimated by measuring incorporation of³H thymidine and³⁵S methionine. The rate of labelled precursor incorporation was measured using the liquid scintillation technique. All substances tested exerted a dose-dependent effect. The synthetic activity of the cultured cells was highest when the concentration of the peptides added to the medium approximated the physiological levels. Methionine-enkephalin, somatostatin and insulin at 2×10^–8 M significantly increased³H thymidine incorporation, by 62%, 69% and 69% respectively, and protein synthesis by 42%, 57% and 57%, respectively. In the case of juvenile gonadal cultured cells, a similar increase in³H and³⁵S incorporation was registered for a 10^–7 M peptide concentration. Both lower and higher peptide concentrations inhibited³H thymidine and³⁵S methionine incorporation. Pharmacological studies suggest the existence of methionine-enkephalin and somatostatin-like receptors on snail gonadal cells. These results indicate that our gonadal cell culture model provides a useful tool for the study of the neuroendocrinological control of the activity of snail gonadal cells.     

Calcium and the contractile response to prostaglandin in the smooth muscle of guinea-pig stomach

Summary In the longitudinal smooth muscle of guinea-pig stomach, verapamil (10^–5 M) which showed marked suppression of high K-induced contractures, did not suppress the contractile response to PGE₁ (1.5×10^–9 to 10^–6 M) markedly. These results suggest that the contractile mechanism of PGE₁ in guinea-pig stomach may mainly depend on a release of bound Ca in the cell and partly depend on a Ca influx from the extracellular origin.     

A histochemical demonstration of the Na++K+-ATPase activity in the thyroid and the effect of cyclic adenosine monophosphate (c-AMP)

Summary With a suitable modification of the Farquhar and Palade technique the Na⁺+K⁺-ATPase activity in guineapig thyroid is demonstrated. The addition of c-AMP (5×10^–6 M or 1.5×10^–5 M) to the incubation media produced an apparent intensification of the Na⁺+K⁺-ATPase activity in the thyroid.This work was supported by a grant from ZMNU of Serbia.     

Effect of enkephalins in the presence of the antibiotic bacitracin in the longitudinal muscle strip preparation from guinea-pig ileum

Summary The antibiotic bacitracin (5×10^–5–4×10^–4 M) increases the inhibition of the contractile response caused by both enkephalin release and direct application of Met-enkephalin 5×10^–7 M in the longitudinal muscle strip preparation from guinea-pig ileum. This effect is attributed to an inhibition of enkephalin degrading peptidases by bacitracin.     

The effect of perchlorate ion and some pharmacological agents on the sensitivity of the frog (Rana pipiens) rectus abdominis to acetylcholine

Zusammenfassung Die Reaktion des mit Eserin vorbehandelten musculus rectus abdominis des Frosches auf Acetylcholin liess sich durch Oxotremorin (2.0·10^–5 M), Arecolin (2.0·10^–5M), Aceclidin (1.0·10^–4 M) und Carbachol (2.0·10^–7 M) verstärken, während Atropin (1.0·10^–5 M) hemmend wirkte. Perchlorationen in Konzentrationen über 2 · 10^–4 M sensibilisierten den Froschmuskel erheblich und reduzierten die Grenzdosis, bei der eine Potenzierung durch Substanzen mit muskarinähnlicher Wirkung beobachtet wird. Alle beschriebenen Reaktionen liessen sich durch Gehirnextrakte verstärken, in denen zuvor Acetylcholin durch Kochen in alkalischer Lösung zerstört worden war.

Parameters affecting the maximum cell concentration ofTetrahymena

Summary In cultures with efficient aeration a maximum cell concentration (MCC) of 6×10⁵ cells/ml (defined medium) and 5.5×10⁶ cells/ml (broth) can be reached. By culturing within Millicells with excess supply of medium and efficient removal of waste products a physical limit for MCC of about 13×10⁶ cells/ml is reached.     

Changes induced by the acute administration of iodide on secretion of iodinated components by the rat thyroid gland perfused in situ

Résumé Nous avons étudié l'effet de l'iodure 127 (10^–3 et 10^–4 M) sur la sécrétion de T₄ ^*, T₃ ^* et –I^* par la thyroïde de rat, prémarquée à l'iode 131 et perfusée in situ avec du sang non radioactif contenant 12 mU de TSH/ml. Il n'a pas été noté de changement significatif dans la sécrétion de T₄ ^* ou T₃ ^* jusqu'à 45 min après le début de l'administration d'iodure 127, qu'il y ait ou non du méthimazole (10^–3 M) dans le sang perfusé. Il y avait par contre une augmentation de la sécrétion d'-I^* pendant la perfusion de 10^–3 M d'ïodure, indiquant une décharge d'-I^* pendant la perfusion de 10^–3 M d'ïodure, indiquant une décharge d'-I^* de provenance intrathyroïdienne. Cet effet est similaire à celui noté auparavant pendant le perfusion de 10^–3 M de ClO ₄ ^– .     

Interaction of polyamines or their precursors with the calcium-controlled secretion of peroxidase by sugarbeet cells

Summary Three polyamines tested (cadaverine, spermidine and spermine) and their 2 precursors (the amino acids arginine and ornithine) inhibit the Ca²⁺-mediated secretion of peroxidases by sugarbeet cells in suspension culture at concentrations ranging from 10^–15 to 10^–5M. In the absence of exogenous Ca²⁺, spermine added at higher concentrations mimics the activatory effect of Ca²⁺, the other polyamines being without effect.Supported by the Belgian FRFC (grant No. 2.9009.75 to T.G.) and the Swiss National Foundation for Scientific Research (grant No. 3.140-0.81 to C.P. and H.G.).     

Differential sensitivity of the two phases of ear artery contraction to intimal and adventitial norepinephrine

Summary The biphasic contraction of the rabbit ear artery to norepinephrine (NE) was investigated in the normal (adventitial stimulation) and the everted (intimal stimulation) segment of ear artery. The 2nd phase response showed an intimal ED₅₀ of 8.2×10^–8 M which was significantly (p<0.05) lower than the adventitial ED₅₀ of 42.6×10^–8 M. This difference was abolished by inhibition of neuronal and extraneuronal uptake for NE. The 1st phase response also showed an ED₅₀ for the intimal stimulation (6.9×10^–8 M) which was significantly (p<0.05) lower than adventitial (65.5×10^–8 M). This difference was reduced but not abolished by NE uptake inhibition. This suggsets that some feature of the adrenergic neuroeffector apparatus is asymmetrically arranged to favor fast responses to blood borne NE.Supported by American Heart Association-Greater Los Angeles Affiliate Grant No. 602. Influence of turbulence on electrokinetic phenomena; experimental determination of the thickness of the diffuse part of the double layer

Zusammenfassung Durch Messungen in 10^–5, 10^–4 und 10^–3 N benzolischen Lösungen von Zn-di-i-propylsalizylat haben wir einen beträchtlichen Einfluss von Turbulenz auf Strömungsströme feststellen können. Dieser Einfluss kann ausgedrückt werden durch einen Faktorf, der in einem unserer Experimente einen Wert vonf=66 erreichte; aus den Werten vonf kann die Dicke der diffusen Doppelschicht in der Flüssigkeit berechnet werden.     

Nervous activity of the frog's epiphysis cerebri in relation to illumination

Zusammenfassung Die proximale Anlage des lichtempfindlichen Pinealorgans adulter Anuren zeigt eine Daueraktivität, die durch Belichtung gehemmt wird. Die mit Hilfe von Stahlmikroelektroden bestimmte absolute Schwelle einzelner Neurone des Organs liegt zwischen 10^–5 und 10^–6 lm/m². Bei Belichtung vermindert sich die Impulsfrequenz der Daueraktivität linear mit dem Logarithmus der Beleuchtungsstärke. Der Halbwert der Entladungsfrequenz liegt bei 10^–2 lm/m². Oberhalb 10 lm/m² ist keine Impulsaktivität zu beobachten.     

Role of calcium in the histamine release induced by D-galactosamine from rat mast cells

Summary Rat peritoneal mast cells were isolated and purified by differential centrifugation in Ficoll. Cells pooled from three to four rats were suspended at approximately 10⁶ cells/ml in a buffered salt solution and incubated for 1 h at 37°C in 300 l volumes in the absence or presence (9×10^–4 M) of calcium chloride. Addition of D-galactosamine hydrochloride (DGM; 2.8×10^–4 M) caused (in addition to basal release) a mean ±SEM percent histamine release of 15.7±5.2 in the presence of Ca⁺⁺ and 19±4.9 in the absence of Ca⁺⁺ (p>0.05). It is suggested that D-galactosamine does not require extracellular Ca⁺⁺ for the release of histamine from the rat mast cell.A preliminary analysis of these results was presented at the International Symposium on calcium entry blockers and tissue protection, Rome, 15–16 March 1984.
