利用已建成的人肠粘液腺癌裸鼠移植瘤标本,制备成癌细胞悬液,进行裸鼠腹腔接种,建立了人肠粘液腺癌裸鼠腹腔移植瘤模型,至今巳传14代,接种方法简单,生长稳定,移植成功率达98.8%。病理组织学和超微结构形态证实腹腔移植瘤保持了人肠粘液腺癌的结构和特点,染色体分析显示人类恶性肿瘤染色体特征。血清癌胚抗原(CEA)含量测定表明,该移植瘤具有高分泌CEA的能力,平均63.4ng/ml,最高达111ng/ml。该腹腔移植瘤的建立,为人体肿瘤的基础研究、抗肿瘤药物的筛选和肿瘤的腹腔内化疗提供了较理想的动物模型。  相似文献

作者将SGC-7901人胃癌细胞接种至棵鼠皮下,建立皮下移植模型并反复传代,以第八代移植瘤为材料,将瘤组织块种植到裸鼠胃壁,成功地建立了裸鼠人胃癌原位移植模型。该模型原位瘤沿胃壁呈浸润性生长,潜伏期为3-4周,以后迅速增长并与网膜粘连,约12周以后部分动物出现肝转移,以后逐渐消瘦,生存期为16-24周,本方法原位成瘤率及局部浸润发生率均为100%,且可出现肝脏转移,肿瘤生长过程与临床胃癌特点很类似,适用于人胃癌转移的研究。  相似文献

介绍纯合子Swiss裸鼠自发恶性淋巴瘤的移植特性和病理形态观察。取肿瘤组织悬液接种无T淋巴细胞的纯合子(nu/nu)小鼠皮下,每点注射0.2ml,肿瘤发生率100%;而接种有T淋巴细胞的小鼠均不生长。该肿瘤共传5代,其特性均与原自发瘤一样。光镜检查为B细胞型恶性淋巴瘤,电镜检查符合B细胞恶性淋巴瘤。  相似文献

胃癌的基础与临床研究需要能真正模拟胃癌在人体内自然生长、侵袭及转移全部过程的动物模型。目前,人胃癌裸鼠移植模型是人体外最接近人类胃癌的整体实验模型,并且造模时间短,成功率高,按移植部位可以分为皮下移植、原位移植、腹腔移植以及转移模型。影响裸鼠移植模型建立的因素主要有人胃癌细胞或外科标本的特性、移植部位及移植、癌细胞数量、裸鼠的品系和周龄以及生长环境和其他因素的影响。  相似文献

探讨了南赤瓟醇提物对裸鼠宫颈癌Caski移植瘤的抑制作用,并在此基础上探寻其可能的机制.将0.3mL Caski(4×107个/mL)细胞悬液接种于裸鼠右侧后腿背部皮下,接种2次,间隔时间1周,建立裸鼠宫颈癌Caski移植瘤模型.将移植瘤裸鼠随机分为模型组、南赤瓟醇提物组(200和400mg/kg)组,每组5只,每天给药1次,连续给药4周,停药次日处死裸鼠,测量裸鼠体重、移植瘤体积、瘤重,计算抑瘤率;光镜下观察移植瘤组织形态学变化;Annexin V-FITC/PI染色测定细胞凋亡率,荧光定量PCR测定Bcl-2、Bax和Caspase-3、Caspase-9mRNA表达,并计算Bcl-2与Bax比值.结果表明南赤瓟醇提物可显著降低移植瘤体积和抑瘤率,上调Bax和Caspase-3、Caspase-9mRNA表达,下调Bcl-2mRNA表达和Bcl-2与Bax比值,与模型组比较具有显著性差异(P0.05或P0.01).南赤瓟醇提取物对人宫颈癌Caski细胞移植瘤生长具有明显抑制作用,其作用机制与调节Bcl-2、Bax和Caspase-3、Caspase-9基因表达有关.  相似文献

 为建立裸鼠胫骨原位骨肉瘤模型,并比较分析MG-63细胞悬液局部注射以及肿瘤组织块移植两种方法的成瘤特性,在注射组体内连续传代筛选MG-63细胞系获得高成瘤率的细胞,将MG-63细胞悬液(25μL,约1×105个)注射于裸鼠胫骨上段髓腔内;组织块移植组将MG-63细胞皮下成瘤瘤组织块接种于裸鼠胫骨上段髓腔内。随后观察两种方法建立的胫骨骨肉瘤原位模型不同的成瘤特性,并采用影像学等手段比较两种方法的成瘤率和成瘤特点。结果表明,术后3周左右可见局部肿瘤形成,4周后通过小动物X线技术可见胫骨中上段溶骨与肿瘤样骨形成,同时在光学显微镜下可见典型的骨肉瘤病理特征。细胞悬液法成瘤率85%,组织块法成瘤率95%,两种方法成瘤率的差异有统计学意义(P<0.05),模型组血清碱性磷酸酶(ALP)明显高于对照组(P<0.01)。两种方法均可形成裸鼠胫骨原位骨肉瘤模型,采用组织块移植法成瘤率较高,瘤体生长速度较快,而且对骨皮质及周围软组织的破坏力较强,转移率高。  相似文献

利用l988年建立的人视网膜母细胞瘤细胞系—SO-Rb_(50),作裸鼠移植瘤的实验研究。在手术显微镜下,用微量注射器吸取SO-Rb_(50)的细胞悬浮液0.45—0.5μl,含细胞数1.5—2.5×10~7个,注入裸鼠眼前房内。共注射7只裸鼠的右眼,左眼为对照。实验眼于注射后第3天瞳孔区即出现肉眼可见到的灰白色混蚀,以后逐日增多,或在前房,或进入玻璃体内。约于注射后第7周出现右眼球突出,逐渐加剧。长期观察的实验鼠左眼也逐渐丧失视力,最后小鼠衰竭死亡。病理检查见典型的视网膜母细胞瘤细胞充满眼球腔,伴有不同程度变性坏死,部分眼内移植瘤瘤细胞呈菊形团样结构。肿瘤侵入眼球各层组织,破坏眼球壁扩展至眼眶内。长期观察的实验鼠肿瘤细胞除侵犯视乳头、视神经外,可沿着视神经侵犯视交叉等。另外,将SO-Rb_(50)的细胞悬浮液0.5ml,含细胞1×10~8 个,注入7只裸鼠右颈背部皮下,只有1只于注射后第4周长出皮下移植瘤,其余6只观察50~90天均未出现肿瘤。可见SO-Rb_(50)细胞接种于眼前房内比接种于皮下更易形成移植瘤。移植瘤的组织学及免疫组织化学检查,与人类视网膜母细胞瘤相一致。  相似文献

一个高转移的人肺巨细胞癌移植瘤PG已在无胸腺裸鼠体内建立。在各种遗传背景的裸鼠体内,包括BALB/cA、NIH、C57BL/6J、615/PBI等,PG肿瘤细胞皮下接种后,均可发生自发转移,局部淋巴结转移率为94%(110/117),肺转移率为83%(97/117)。这种高转移特性在五年多时间内连续100多代移植过程中仍保持不变。为了研究肿瘤转移的遗传机制,我们从PG瘤细胞中提取高分子量DNA片段,用磷酸钙共沉淀的方法转染NIH/3T3小鼠纤维母细胞。经过连续两轮转染,获得的转化细胞能在裸鼠体内成瘤。其中一个转化克隆TF87-2-10皮下接种裸鼠后,能自发转移至肺。DNA分子杂交显示该细胞基因组中整合有人DNA序列。说明该转化细胞转移表型的  相似文献

为探讨人卵巢癌进展机制的相关研究,构建适宜的体内动物模型。采用慢病毒转染技术,构建稳定过表达绿色荧光报告基因(GFP)的人EOC细胞模型:SKOV3(人卵巢浆液性腺癌细胞株)和ES-2(人卵巢透明细胞癌细胞株);异氟烷吸入麻醉4-6周龄BALB/C(nu/nu)雌性裸鼠,从裸鼠背部正中纵切口进入腹腔,显微多点注射法分别将细胞模型悬液,移植于裸鼠的单侧卵巢,每组6只裸鼠;Night OWL活体成像系统动态观察裸鼠移植瘤生长和转移情况;移植术后4-6周解剖裸鼠,取卵巢和转移瘤组织行HE染色。结果显示,携带GFP的EOC细胞模型SKOV3和ES-2,均可成功构建裸鼠卵巢原位移植瘤模型,裸鼠未发生手术原因死亡,成功率均100%;活体成像系统通过检测GFP的荧光强度和范围,能够动态监测裸鼠移植瘤生长情况;SKOV3细胞模型构建的卵巢癌和转移瘤组织,HE染色观察病理特征符合人卵巢浆液性腺癌;ES-2细胞模型构建的卵巢癌和转移瘤组织,HE染色观察病理特征符合人卵巢透明细胞癌。由此可知,异氟烷吸入麻醉、从背部正中纵切口进入腹腔、显微多点注射法移植携带GFP的EOC细胞模型,能成功构建可动态监测的裸鼠卵巢原位移植瘤动物模型;该动物模型与人卵巢癌生物学特征类似,适宜作为研究人卵巢癌演变进展的体内模型。  相似文献

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.  相似文献

The discovery of the prolific Ordovician Red River reservoirs in 1995 in southeastern Saskatchewan was the catalyst for extensive exploration activity which resulted in the discovery of more than 15 new Red River pools. The best yields of Red River production to date have been from dolomite reservoirs. Understanding the processes of dolomitization is, therefore, crucial for the prediction of the connectivity, spatial distribution and heterogeneity of dolomite reservoirs.The Red River reservoirs in the Midale area consist of 3~4 thin dolomitized zones, with a total thickness of about 20 m, which occur at the top of the Yeoman Formation. Two types of replacement dolomite were recognized in the Red River reservoir: dolomitized burrow infills and dolomitized host matrix. The spatial distribution of dolomite suggests that burrowing organisms played an important role in facilitating the fluid flow in the backfilled sediments. This resulted in penecontemporaneous dolomitization of burrow infills by normal seawater. The dolomite in the host matrix is interpreted as having occurred at shallow burial by evaporitic seawater during precipitation of Lake Almar anhydrite that immediately overlies the Yeoman Formation. However, the low δ18O values of dolomited burrow infills (-5.9‰~ -7.8‰, PDB) and matrix dolomites (-6.6‰~ -8.1‰, avg. -7.4‰ PDB) compared to the estimated values for the late Ordovician marine dolomite could be attributed to modification and alteration of dolomite at higher temperatures during deeper burial, which could also be responsible for its 87Sr/86Sr ratios (0.7084~0.7088) that are higher than suggested for the late Ordovician seawaters (0.7078~0.7080). The trace amounts of saddle dolomite cement in the Red River carbonates are probably related to "cannibalization" of earlier replacement dolomite during the chemical compaction.  相似文献

There are numerous geometric objects stored in the spatial databases. An importance function in a spatial database is that users can browse the geometric objects as a map efficiently. Thus the spatial database should display the geometric objects users concern about swiftly onto the display window. This process includes two operations:retrieve data from database and then draw them onto screen. Accordingly, to improve the efficiency, we should try to reduce time of both retrieving object and displaying them. The former can be achieved with the aid of spatial index such as R-tree, the latter require to simplify the objects. Simplification means that objects are shown with sufficient but not with unnecessary detail which depend on the scale of browse. So the major problem is how to retrieve data at different detail level efficiently. This paper introduces the implementation of a multi-scale index in the spatial database SISP (Spatial Information Shared Platform) which is generalized from R-tree. The difference between the generalization and the R-tree lies on two facets: One is that every node and geometric object in the generalization is assigned with a importance value which denote the importance of them, and every vertex in the objects are assigned with a importance value,too. The importance value can be use to decide which data should be retrieve from disk in a query. The other difference is that geometric objects in the generalization are divided into one or more sub-blocks, and vertexes are total ordered by their importance value. With the help of the generalized R-tree, one can easily retrieve data at different detail levels.Some experiments are performed on real-life data to evaluate the performance of solutions that separately use normal spatial index and multi-scale spatial index. The results show that the solution using multi-scale index in SISP is satisfying.  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
