Using the immuno-fluorescence and immuno-gold electron microscope technology, localization of ni- tric oxide synthase (NOS)-like proteins was determined in guard cells of Vicia faba L. NOS is mainly localized in nucleus, cytoplasm, chloroplast, mitochondria and the cell wall of guard cells. Scorch and exogenous JA can enhance the level of nitric oxide (NO) and increase NOS activity in both leaf and epidermis, and the changing pattern of NOS activity was consistent with the change of NO. NOS in- hibitor, L-NAME, inhibited JA-induced NO generation. From the results, we presumed that NO genera- tion from NOS pathway is the main pathway in the stress and JA responses. The pharmacological ex- periment showed that increasing the Ca2 at a suitable concentration promoted leaf NOS activity and the NO level, indicating that NOS activity together with the distribution of NO is Ca2 -dependent. NOS and NO are possibly involved in the regulation of stomatal movement thus playing an important role in plant stress responses.  相似文献

NO and N2O fluxes from agricultural soils in Beijing area   总被引:2,自引:0,他引:2  
Chinese agriculture represents one of the most intensively managed agroecosystems in the world. Typical nitrogen fertilization rates are more than three times those of the world' s average, resulting in dramatically accelerated nitrogen cycling in China. In this study, we have examined NO and N2O exchange in the upland agricultural systems of Beijing area. Inorganic and organic fertilizer treatments were arranged in order to evaluate their impact on the magnitude and proportion of trace gas emissions. Increasing inorganic fertilization rates showed a highly significant impact upon emissions of both NO and N2O. Organic matter amendment did not have a statistically significant impact on the N-gas fluxes examined here. Overall losses of added nitrogen by NO and N2O emission averaged 1.24% and 0.22% respectively over the range of treatments in this study. Results from our field study indicate that compared with other studies done elsewhere, emissions of reactive nitrogen from agricultural systems in Beijing area are not so large as expected before.  相似文献

Nitric oxide (NO) and Jasmonic acid (JA) are two key signaling molecules involved in many and diverse biological pathways in plants. Growing evidence suggested that NO signaling interacts with JA signaling. In this work, Our experiment showed that NO exists in guard cell of Vicia faba L., and NO is involved in signal transduction of JAinduced stomata closuring: ( i ) JA enhances NO synthesis in guard cell; ( ii ) both JA and NO induced stomatal closure, and had dose response to their effects; ( iU ) there are synergetic correlation between JA and lower NO concentration in regulation of stomatal movement; (iV) JA-induced stomatal closure was largely prevented by 2-phenyl-4,4,5,5-tetramethylimidazoline-l-oxyl-3-oxide (PTIO), a specific NO scavenger. An inhibitor of NO synthase (NOS) in mammalian cells, N^G-nitro-L-Arg-methyl eater (L-NAME) also inhibits plant NOS, repressing JA-induced NO generation and JA-induced stomatal closure. We presumed that NO mainly comes from NOS after JA treatment.  相似文献

To account for the effects of both chemistry and flow turbulence, the present study proposes an integrated NO sub-model that combines the extended Zel'dovich mechanism and engine CFD computations to simulate the NO histories in a diesel engine. NOx sub-model parameters and pollutant formation mechanisms can be more easily investigated by solving the NOx sub-model. The new NO formation model incorporating the effects of both chemical kinetics and turbulent mixing was applied to simulate a diesel engine with a quiescent combustion chamber, and one with a re-entrant combustion chamber; the premise of the model being the reaction rate is mainly determined by a kinetic timescale and a turbulent timescale. The results indicate that the predicted NO formulation from the new model agrees well with the measured data. As the utilization of fossil fuels continues to increase, the control of NOx emissions is a worldwide concern; and it is imperative to understand fully the NOx reaction processes in combustion systems. This technology has the potential to enhance the application of various combustion techniques used to reduce NOx emissions from practical combustion systems.  相似文献

To avoid unfavorable environmental condi- tions, Scrippsiella trochoidea is capable of forming a resting cyst in the process of growth. In the present study, we investigated the effects of nitrate and ammonium on the growth and encystment of S. trochoidea. We incubated S. trochoidea in modified f/2 media without nitrogen and silicate in flasks. The flasks were divided into two groups. Nitrate was added as a nitrogen source in the first group, and ammonium was added in the second group. The concentrations of the nitrogen compounds were 0, 10, 30, 60, and 90 μmol/L. The results indicate that NO3--N favors cell growth, and the cultures with a higher concentration of NO3--N were ineffective at forming cysts. In contrast, NH4+-N promoted cell growth and cyst formation. At similar concentrations as NO3--N, NH4+-N had a toxic effect on cell growth and increased the cyst formation ratio. Thus, the NH4+-N concentration is an important factor for controlling encystment. We believe that the impact of NH4+-N in inducing cyst formation may be a useful feedback mechanism in ecological systems.  相似文献

The fecC gene encoding a putative iron (Ⅲ) dicitrate transporte rwas cloned from nitrogen-fixing cyanobacterium Anabaena sp. PCC 7120, and inactivated. The mutant grows normally in medium with NO3^- , NH1^- or without combined nitrogen. But in iron-deficient medium, the mutant grows slowly. Photosynthetic properties were compared between the mutant and the wildtype strain, the content of photosynthetic pigments in the mutant is lower than that of the wild-type. The results of RT-PCR experiments show that the fecC gene is expressed under iron-deficient conditions, but is not expressed under iron-replete conditions. These results revealed that fecC gene product is required for optimal growth under iron-deficient conditions in Anabaena sp. PCC 7120.  相似文献

To examine the community structure of β-Proteobacterial ammonia-oxidizing bacteria (AOB) in prawn farm sediment, the 16S rRNA gene library was constructed with β-Proteobacterial AOB-specific primers. The library was screened by PCR-restriction fragment length polymorphism (RFLP) analysis and clones with unique RFLP patterns were sequenced. Two groups of β-Proteobacterial AOB, the Nitrosomonas and the Nitrosospira, were detected. The Nitrosomonas occupied an absolute dominant position, accounting for more than 90% of total clones in the clone library, while the Nitrosospira accounting for 5.48%. Nitrosomonas-affiliated clones were grouped into the Nitrosomonas marina and the Nitrosomonas sp. Nm143 clusters, and Nitrosospira-affiliated clones were grouped into the Nitrosospira cluster 1. No other clusters of β-Proteobacterial AOB were found. The results enriched our knowledge of AOB diversity in the prawn farm sediment and provided important foundational data for further functional studies of these microbes in mariculture environments.  相似文献

利用环境16S rRNA基因序列分析技术,对福建漳江口红树林沉积物中存在的古菌类群进行检测。首先提取红树林沉积物的总DNA,并进一步构建环境16S rRNA基因克隆文库。随机挑选文库克隆,根据Amplified rDNA restriction analysis(ARDRA)分析分型后进行测序。对所测序列进行系统进化分析。结果显示,文库中的大部分古菌克隆(95%)分别属于广古菌门(Euryarchaeota)中的Marine benthic group D和泉古菌门(Crenarchaeota)的Miscellaneous crenarchaeotic group和Marine group I等3个已描述的古菌类群, 另有小部分克隆(5%)可能代表广古菌门中新的古菌类群。Miscellaneous crenarchaeotic group类群古菌是红树林沉积物中古菌的优势类群。上述古菌类群的检测加深了人们对于红树林古菌群落结构复杂性及其生态学意义的认识。  相似文献

通过生态群落16S rRNA基因库的重建并采用限制性酶切长度多态性(RFLP)分析的方法,对铜绿山铜矿酸性矿水中微生物群落的多样性进行研究。通过RFLP对扩增出的327个16S rRNA PCR产物进行分析,对44个16S rRNA的克隆子进行测序。每个样点各有2~6个主要操作分类单元,这些操作分类单元所对应的克隆子数分别占各样点克隆子总数的56.3%~69.6%。93.8%的克隆子序列与现在的数据库中序列相似性大于90%。样点酸性矿坑水主成分分析与系统发育分析结果显示:在低pH值、高离子浓度的样点tls1与tls2中多数克隆子属于gamma-Proteobacteria(主要为Acidithiobacillus ferrooxidans)和Nitrospira科(主要为Leptospirillum);而在较高pH值、低离子浓度的样点tls3的克隆子多属于gamma-Proteobacteria(没有A.ferrooxidans)和alpha-Proteobacteria。在古矿井区域酸性矿坑水中微生物群落的多样性很低,生物地球化学特性相近的酸性矿坑水由具有相近的微生物群落组成。  相似文献

cDNA fragment of the gene (dehydration induced,di1) of wheat (Triticum aestivum. L) induced by 30% PEG-6000 (−1.13 MPa) treatment was isolated with mRNA differential display technique. Northern blot analysis showed that the expression ofdi1 gene improved at 10 h reached the highest at 48 h under 30% PEG-6000 treatment. cDNA fragment ofdi1 gene has been cloned and sequenced (211 bp).  从青海柯柯盐湖泥样中分离的16株细菌,用2种限制性内切酶酶切进行ARDRA分析,其中12株菌的酶切带谱有较大差异.根据这12株菌的16SrDNA序列进行系统发育分析结果表明,12株菌有11株分布于芽孢杆菌科中的Halobacillus,Gracibacillus,Amphibacillus,Virgibacillus,Bacillus,Salibacillus和1个潜在的新属中,另1株属于Proteobacteria的Gamma亚群Halomonadaceae科,Halomonas属.而且16SrDNA系统发育分析提示这些菌株中存在3个新种和1个新属的可能.本研究得出的结论为:应用ARDRA来初步筛选菌株是可行的;青海柯柯盐湖这一极端环境中存在较丰富的新的芽孢菌种类.  相似文献

To study the promoter activities of genes cpcT 2 and cpcS 2,several upstream DNA fragments of these two genes with different lengths were selected.These fragments were fused with promoterless gfp(reporter gene) for constructing five recombinant plasmids.Then,these recombinant plasmids were transferred into Nostoc PCC 7120 by conjugation.The promoter activities of genes cpcT 2 and cpcS 2 were determined by observing the fluorescence of green fluorescent protein(GFP) with a fluorescence microscope.The result showed that the 1 300 bp(-1 300 to 0 bp) and 2 600 bp(-2 600 to 0 bp) upstream fragments of cpcT 2 had strong promoter activity and the promoter activity of the 680 bp(-680 to 0 bp) fragment was weaker than that of two above fragments of cpcT 2.The 2 000 bp(-2 000 to 0 bp) upstream fragment of cpcS 2 revealed weak promoter activity.This showed that a strong promoter of cpcT 2 was located in the upstream fragment between-680 and-1 300 bp.  相似文献

以现有菠菜基因组信息为基础,通过生物信息学方法筛选鉴定16个菠菜SoSWEET蛋白家族成员,命名为SoSWEET1~SoSWEET16.氨基酸残基数量在648~1 140之间,分子质量在54 070.32~95 868.64 u之间,理论等电点(pI)在5.06~5.19之间.亚细胞定位预测有6个SoSWEET蛋白定位于细胞膜,5个SoSWEET蛋白定位于内质网,5个SoSWEET蛋白定位于细胞膜、内质网.系统进化分析将菠菜SoSWEET蛋白家族分成4个亚族,在此基础上对基因结构、保守基序、顺式作用调控元件等进行分析.共鉴定了10个高度保守基序,其中所有菠菜SoSWEET蛋白都包含基序1,2和4,是构成菠菜SoSWEET蛋白中最高度保守的部分.所有菠菜SoSWEET蛋白家族成员都含MtN3_slv和PQ-loop superfamily结构域.大多数SoSWEET蛋白家族的基因含有5个内含子.顺式作用元件预测结果表明,菠菜SoSWEET基因启动子上包含光响应、生长发育、植物激素响应和逆境胁迫响应等顺式作用元件.组织表达分析表明,所有SoSWEET基因在根、茎、叶和叶柄中都有表达,霜霉病胁迫处理后16个基因表现出不同响应变化.本研究为后续深入研究菠菜SoSWEET蛋白家族成员的功能提供了重要参考.  相似文献

Metabotropic glutamate receptor 7, coupled with a chemical neurotransmitter L-glutamate, plays an important role in the development of many psychiatric and neurological disorders. To study the biological and genetic mechanism of the mGluR7-related diseases, a physical map covering the full-length mGluR7 genomic sequence has been constructed through seed clone screening and fingerprinting database searching. These BAC clones in the physical map have been sequenced with shotgun strategy and assembled by Phred-Phrap-Consed software; the error rate of the final genomic sequence is less than 0.01%. mGluR7 spans 880 kb genomic region, the GC content and repeat content of mGluR7 genomic sequence are 38% and 37.5% respectively. mGluR7 has a typical “house-keeping” promoter and consists of 11 exons, with introns ranging from 6 kb to 285 kb. mGhiR7a and mGluR7b are two known alternatively splicing variants. Comparing the genomic structures of extracellular domains of mGluR family, their genomic structures can be subdivided into three groups, which are consistent with that of proteins. Although the genomic organization of mGluR7’s group is conserved, the majority of introns in the extracellular segments vary dramatically. It is an obvious trend of the increasing intron size inverse proportion to phylogenetic time. Variation of genomic structure is higher than that of protein, which is attributed to the species characteristic regulation of gene expression.  相似文献

Four strains with typical morphology of actinomycete genus ofFrankia were isolated from root nodules ofCoriatia nepalensis. They were shown to nodulate the seedlings of host plant and hybridize withFrankia 16S rRNA targeted olionucleotide probes, indicating that they did belong to the genusFrankia. Furthermore, by nifHDK probe hybridizations, the homologous fragments of nifHDK genes were detected among the bacteria, and they were located in various sizes of restriction fragments of total DNA, showing diverse patterns of restriction fragment length polyrnorphisms of nifHDK gene (nifHDK-RFLPs). The PCR-based amplification and cloning of nifH gene throw light on the molecular phylogeny ofCoriaria -infectiveFrankia.  相似文献
