通过PCR扩增及定点突变等构建了pET-DsbA/NAP1 (18~44)融合表达载体,将其转化入大肠杆菌BL21后获得可溶表达.通过镍亲和柱,Thrombin酶切及G50分子筛对鼻咽癌NAP1(18~44)进行了纯化并进行了质谱鉴定.NAP1(18~44)的圆二色谱和2D 1HNMR DQF-COSY谱表明鼻咽癌肽段在TFE溶液中能形成一定的α螺旋.这些工作为进一步用核磁共振技术研究NAP1(18~44)打下了基础.  相似文献

采用傅里叶变换红外光谱和圆二色谱,测定鲑鱼降钙素在不同pH值水溶液中的构象,并对各种二级结构的含量进行计算.结果表明pH值对鲑鱼降钙素溶液构象有较大影响,pH值从2.0升高到8.0时鲑鱼降钙素溶液中存在β-折叠、β-转角和无序结构,无α-螺旋结构,且各种二级结构的含量随pH值增加而发生变化.当pH值增加到9.0时鲑鱼降钙素溶液中出现α-螺旋结构,且β-折叠和无序结构含量降低到最小值,β-转角含量增加到最大值.以上研究结果提示较低的pH值有助于鲑鱼降钙素的稳定,为进一步阐明pH值对鲑鱼降钙素稳定性的影响提供了依据.  相似文献

用圆二色性谱(CD谱)研究了猪胰蛋白酶及其自溶活性产物在溶液中的构象,并观察了在十二烷基硫酸钠(SDS)溶液中酶分子构象的变化。 猪胰蛋白酶的CD谱显示有相当量的α-螺旋成份及少量β-折叠成份,它在190nm附近的[θ]值远大于牛胰蛋白酶。其自溶活性产物的CD谱显示α-螺旋的成份减少5—6%,β-折叠成份上升3—6%,而有序部分所占比例基本不变,提示自溶后α-螺旋转化为β-折叠。 在SDS溶液中,α-螺旋和β-折叠在胰蛋白酶分子中所占的比例提高,而α-螺旋成份增加更为明显,表明酶分子的有序度提高。 参照Chou与Fasman以及Chen,Yang与Martinez等人的方法,预测猪胰蛋白酶有36%的氨基酸残基可以形成α-螺旋,由实测CD谱计算,亦在37%左右,两者相符。  相似文献

黄精凝集素II(Polygonatum cyrtonema Hua.Lectin II,PCLII)具有323nm的荧光发射峰和281.6nm的激发峰,其远紫外圆二色谱(CD谱)显示224nm负峰,分子中含有17.3%的α螺旋和45.8%的β折叠,在10mmol/L的SDS中,PCL II完全丧失兔红细胞凝集能力及促T-淋巴细胞有丝分裂活性,CD谱呈现双负峰的a螺旋构象,去除SDS后,则呈无规卷曲构象,此时,荧光发射峰红移而激发峰蓝移,PCL II仍完全失活,在EDTANa2溶液中,PCL II凝集红细胞能力及促淋巴细胞有丝分裂活性完全丧失,去除EDTA后,红细胞凝集活性完全恢复,但仍失去促淋巴细胞有丝分裂能力,PCL II二级结构变化不大,荧光光谱的变化表明微环境受到影响。  相似文献

为了探明重金属Hg^2+进入体内,对血清蛋白固有结构和功能的影响,我们用Hg^2+对NBS修饰前后血清蛋白HSA和BSA进行处理,然后分析血清蛋白的圆二色谱特性,结果表明,近紫外区未修饰的HSA和BSA均具有典型的α螺旋构象;Hg^2+处理或NBS修饰BSA溶液,均引起α-螺旋含量减少和三级结构构象的改变.Hg^2+处理HSA,α-螺旋含量略有增加,构象也有变化.而NBS修饰导致α簇结构的严重破坏,蛋白构象无序化程度加深.修饰对BSA、HSA光谱特性的影响不同,可能与这两种蛋白的结构差异有关.  相似文献

研究了二价铜离子对牛朊蛋白的二级结构的影响.牛朊蛋白(BoPrPc)的小分子(BoPrPcS)和大分子(BoPrPcL)经初步纯化后,在含Cu2+的碱性溶液中氧化,用反相柱进行氧化态和还原态的BoPrPcL和BoPrPcS的分离.质谱分析证明氧化态的Bo-prPcL和BoPrPcS分子质量分别为24.064,15.819ku.用远紫外圆二色谱(Far-UVCD)分析显示,BoPrPcS在208和222m处出现两个负峰,为典型的α-螺旋型结构;而Bo-PrPcL在222nm处负峰减小.与未经氧化的BoPrPcL的质谱和CD结果比较后发现,氧化后的BoPrPcL在八肽重复区结合了6个Cu2+,其β-折叠含量明显增加.而Cu2+对BoPrPcS二级结构基本无影响.BoPrPcL结合Cu2+的特性及其二级结构的相应变化可能与细胞内抗Cu2+毒性与氧化压力有关.  相似文献

本文报告了对猪血浆α_1抗胰蛋白酶的紫外吸收光谱和园二色谱的测定结果。从紫外光谱分析得知猪α_1抗胰蛋白酶表现出典型的蛋白质吸收特征,其摩尔消光系数E_(280nm)~1%1cm 等于5.25;从(?)二色谱分析得知该蛋白质分子中合有约8%的α螺旋,50%的β-折叠,其余均为无规则卷曲构象。本测定结果对猪α_1-抗胰蛋白酶的性质、结构和功能的研究打下了基础。  相似文献

用紫外-可见吸收光谱,荧光光谱和圆二色谱对细胞色素c和脱辅基细胞色素c在不同的φ(乙醇)溶液中的结构进行了表征。结果表明,乙醇对细胞色素c的α-螺旋二级结构影响很小,而对血红素配位的影响十分明显,可导致血红素Fe-Met80配位键断裂,另一方面,乙醇对脱辅基细胞色素c有明显的结构转换诱导作用,在φ(C2H5OH)=0.55(乙醇/水)溶液中,脱辅基细胞色素c的α-螺旋结构含量可以达到全蛋白的89%,研究结果表明:负电荷微环境的诱导对脱辅基细胞色素c的结构转换并不是必要的,改变溶剂与脱辅基细胞色素c之间的疏水和氢键相互作用力也可以诱导脱辅基细胞色素cα-螺旋的形成。  相似文献

采用刚果红实验判断白术多糖WAM在溶液中的构象特征,通过测定不同溶液体系中多糖分子的黏度、比旋度、紫外吸收光谱和圆二色谱的变化来研究多糖构象及其变化规律.实验结果表明:WAM不具有螺旋结构,温度、金属离子、变性剂均可以使WAM构象发生改变.WAM在溶液中以无规则线团形式存在.  相似文献

采用圆二色谱法表征了不同冻藏模式和冻藏时间对小麦面筋蛋白二级结构的影响.结果表明:在SDS溶液中,未经冻藏的面筋蛋白二级结构主要由β折叠和无规则卷曲组成;当冻藏时间小于60天时,两种冻藏模式对小麦面筋蛋白的二级结构影响不大;恒温冻藏模式中面筋蛋白二级结构在冻藏90天时变化最显著,易溶部分中α螺旋转化为β折叠和β转角,难溶部分中β折叠和β转角转化为α螺旋和无规则卷曲;冻融冻藏模式中面筋蛋白二级结构在冻藏60天时变化最显著,易溶部分中α螺旋转化为β折叠和无规则卷曲,难溶部分中β折叠和β转角转化为α螺旋和无规则卷曲.这说明,在恒温冻藏模式下,只有当冻藏时间达到或超过90天的时候,才对面筋蛋白的二级结构产生影响,而在冻融模式下,冻藏时间仅为60天时便可对面筋蛋白的二级结构产生影响.  相似文献

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. There are numerous geometric objects stored in the spatial databases. An importance function in a spatial database is that users can browse the geometric objects as a map efficiently. Thus the spatial database should display the geometric objects users concern about swiftly onto the display window. This process includes two operations:retrieve data from database and then draw them onto screen. Accordingly, to improve the efficiency, we should try to reduce time of both retrieving object and displaying them. The former can be achieved with the aid of spatial index such as R-tree, the latter require to simplify the objects. Simplification means that objects are shown with sufficient but not with unnecessary detail which depend on the scale of browse. So the major problem is how to retrieve data at different detail level efficiently. This paper introduces the implementation of a multi-scale index in the spatial database SISP (Spatial Information Shared Platform) which is generalized from R-tree. The difference between the generalization and the R-tree lies on two facets: One is that every node and geometric object in the generalization is assigned with a importance value which denote the importance of them, and every vertex in the objects are assigned with a importance value,too. The importance value can be use to decide which data should be retrieve from disk in a query. The other difference is that geometric objects in the generalization are divided into one or more sub-blocks, and vertexes are total ordered by their importance value. With the help of the generalized R-tree, one can easily retrieve data at different detail levels.Some experiments are performed on real-life data to evaluate the performance of solutions that separately use normal spatial index and multi-scale spatial index. The results show that the solution using multi-scale index in SISP is satisfying.  相似文献

The discovery of the prolific Ordovician Red River reservoirs in 1995 in southeastern Saskatchewan was the catalyst for extensive exploration activity which resulted in the discovery of more than 15 new Red River pools. The best yields of Red River production to date have been from dolomite reservoirs. Understanding the processes of dolomitization is, therefore, crucial for the prediction of the connectivity, spatial distribution and heterogeneity of dolomite reservoirs.The Red River reservoirs in the Midale area consist of 3~4 thin dolomitized zones, with a total thickness of about 20 m, which occur at the top of the Yeoman Formation. Two types of replacement dolomite were recognized in the Red River reservoir: dolomitized burrow infills and dolomitized host matrix. The spatial distribution of dolomite suggests that burrowing organisms played an important role in facilitating the fluid flow in the backfilled sediments. This resulted in penecontemporaneous dolomitization of burrow infills by normal seawater. The dolomite in the host matrix is interpreted as having occurred at shallow burial by evaporitic seawater during precipitation of Lake Almar anhydrite that immediately overlies the Yeoman Formation. However, the low δ18O values of dolomited burrow infills (-5.9‰~ -7.8‰, PDB) and matrix dolomites (-6.6‰~ -8.1‰, avg. -7.4‰ PDB) compared to the estimated values for the late Ordovician marine dolomite could be attributed to modification and alteration of dolomite at higher temperatures during deeper burial, which could also be responsible for its 87Sr/86Sr ratios (0.7084~0.7088) that are higher than suggested for the late Ordovician seawaters (0.7078~0.7080). The trace amounts of saddle dolomite cement in the Red River carbonates are probably related to "cannibalization" of earlier replacement dolomite during the chemical compaction.  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献

The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.  相似文献
