为探究Rnd1基因在大黄鱼免疫应答过程中的作用,采用实时荧光定量PCR技术(qRT-PCR)对该基因的表达模式进行分析,同时构建原核表达载体pET-28a-Rnd1,并转化进大肠杆菌后诱导该蛋白的融合表达。研究结果表明:大黄鱼Rnd1基因ORF为699 bp,编码232个氨基酸;经序列多重比对和进化树构建发现,Rnd1的氨基酸序列高度保守;在健康大黄鱼的8个免疫组织中,Rnd1在肝脏中相对表达量最高,其次为脑,而在肠中表达量最低;变形假单胞菌攻毒后,Rnd1在肝脏中48 h时达到最高值,为对照组的25倍;在脾脏中则持续上调表达,尤其在48 h后升高更为明显。  相似文献

克隆得到了调节雌雄激素平衡的大黄鱼cyp19a/b基因,cyp19a基因c DNA全长1805 bp(NCBI登录号:FJ800566),其中开放阅读框1557 bp,编码518个氨基酸;cyp19b基因c DNA全长2268 bp(NCBI登录号:FJ800567),其中开放阅读框1503 bp,编码500个氨基酸.荧光定量PCR分析显示cyp19a基因在性腺中有明显的表达,且在卵巢中的表达量极显著且高于精巢;在肌肉、脑、肝脏、肾脏、脾脏等组织器官中基本不表达.而cyp19b基因在脑、脾脏和肝脏中有较高表达,在性腺和其他器官中基本不表达.  相似文献

铜蓝蛋白(Ceruloplasmin,Cp)是一种重要的铜转运蛋白,合成于肝脏并参与生物体铁的代谢,在医学上是各种炎症、感染、中毒及癌症疾病的标志性蛋白.铜蓝蛋白的研究已在多种真骨鱼类中被报道,文中第一次在稀有鮈鲫(Gobiocypris rarus)中报道此基因.采用cDNA末端快速扩增技术(rapid amplification of cDNA ends,RACE)克隆了稀有鮈鲫铜蓝蛋白基因,使用荧光定量PCR的方法构建了该基因组织表达谱.序列分析表明稀有鮈鲫铜蓝蛋白基因包含3 264bp全长编码序列,该序列编码1 087个氨基酸,其核苷酸和氨基酸序列与斑马鱼同源性最高(分别为88.1%和90.3%).理论相对分子质量和等电点分别为124 429.1D和6.41.荧光定量PCR检测表明该基因在肝脏和脾脏中相对表达量最高,在肌肉和鳃中相对表达量最低.使用氨基酸序列进行蛋白结构保守域分析,结果表明铜蓝蛋白基因在脊椎动物中是相对保守的,推测其功能也与其他物种相似.这为进一步研究稀有鮈鲫该基因的功能及其应用奠定了基础.  相似文献

从黄姑鱼转录组数据库中筛选出NK-lysin基因,命名为YdNkl-2。YdNkl-2基因cDNA全长600 bp,其中开放阅读框为456 bp,编码151个氨基酸。YdNkl-2具有一个信号肽和一个鞘脂激活蛋白B(saposin B)结构域。亚细胞定位结果显示,YdNkl-2分布于细胞质和细胞核。实时荧光定量PCR结果表明,YdNkl-2的mRNA广泛分布于黄姑鱼各组织,其中在鳃和脾脏中的表达量最高。经哈维氏弧菌感染后,头肾、肝脏和脾脏中的YdNkl-2表达量均明显升高,提示YdNkl-2在哈维氏弧菌感染黄姑鱼过程中发挥重要作用。  相似文献

【目的】考察棘 腹 蛙(Rana boulengeri)白 细 胞 介 素-34(Interleukin-34,IL-34)基 因 的 信 息 和 该 基 因 组 织 表 达 特 性。【方法】基于实验室构建的棘腹蛙皮肤转录组数据库,采用反转录PCR从棘腹蛙皮肤组织中克隆IL-34 基因的全长c DNA序列,并用荧光定量PCR法检测IL-34 基因在棘腹蛙各组织中的表达情况。【结果】棘腹蛙IL-34 基因的c DNA全长为864bp,包含564bp的开放阅读框,编码187个氨基酸;预测IL-34理论分子量大小为70.56k Da,理论等电点为4.85。荧光定量PCR分析表明IL-34 基因在棘腹蛙皮肤、脑、肝脏、脾脏、胃、肌肉等组织中均有表达,在脾脏组织中表达量最高。【结论】成功克隆了棘腹蛙IL-34 基因并探明了它的组织表达模式,为深入研究棘腹蛙IL-34的生物学功能提供了理论依据。

应用荧光定量PCR法分析抗菌肽hepcidin、c型溶菌酶和MHCⅠ基因在健康革胡子鲶肝胰脏、脾脏、胃、肠道、肾脏、头肾、心脏、脑、鳃、鳃上器官、背部皮肤、背部肌肉和尾鳍13种组织中的表达.结果显示:3种基因在以上组织中均有表达,Hepcidin基因在脾脏中表达最高,显著高于除肾脏以外的其它组织(P0.05);c型溶菌酶基因在胃中表达量最高,与其余组织的表达差异显著(P0.05);而MHCⅠ基因在脾脏和鳃中表达量相对较高,但13种组织间的表达差异不显著(P0.05).  相似文献

为了解黑鲷(Acanthopagrus schlegelii)在盐度胁迫过程中的适应机制,本研究利用基因扩增、聚类分析、荧光定量PCR等技术对NKCC1基因进行生物信息学分析以及在不同组织中的表达特征研究,并探讨其在急性盐度胁迫下的表达机制。结果表明NKCC1基因的开放阅读框(Open Reading Frame, ORF)为3 504 bp,共编码1 167个氨基酸,预测蛋白分子量为127.19 kDa,理论等电点为5.75。结构域分析表明黑鲷NKCC1分子为跨膜蛋白,并包含一个典型的Na~+-K~+-Cl~-共同转运蛋白SLC12A结构域,且在不同物种中具有很高的保守性;序列比对分析表明黑鲷NKCC1氨基酸序列与金头鲷NKCC1a的同源性最高,为97.8%;组织表达分析表明黑鲷NKCC1基因在所检测的12个组织,即肝脏、肾脏、心、脑、眼、鳃、鳍条、脾脏、皮肤、肠、性腺和白肌中均有表达,其中在肾脏、鳃和肠中表达量最高,并显著高于其他组织(P0.05);在低盐度胁迫下,NKCC1基因在鳃组织中的表达量对盐度变化响应十分迅速,而在肾脏组织中,低盐度和高盐度胁迫都会在不同时间点抑制NKCC1基因的表达量。综上所述,本研究为解析NKCC1基因在黑鲷调节渗透压和离子平衡过程中的功能机制奠定了基础,并为促进黑鲷养殖产业的可持续发展提供一定的技术理论。  相似文献

采用Illumina MiSeq二代测序方法筛选得到青蛤丝裂原活化蛋白激酶(MAPK/p38)基因相似序列,进一步克隆得到全长cDNA.利用荧光定量PCR技术检测该基因在青蛤各组织中以及在鳗弧菌胁迫下的组织表达特征.结果显示,该基因全长共1781bp,开放阅读框长度为1104bp,编码367个氨基酸.在正常条件下该基因在青蛤体各组织中都表达,其中血淋巴中表达量最高,肝脏次之,鳃中最少.经菌液侵染后12h青蛤血淋巴中表达量即达到峰值,随后表达量逐渐降为正常水平.  相似文献

采用垂直板聚丙烯酰胺凝胶电泳方法,对沙塘鳢(Odontobutis abscura)的消化道、心脏、肌肉、脾脏、肾脏、脑、肝脏、生殖腺和晶体9种组织的4种同工酶(EST、LDH、ADH、MDH)进行了研究,讨论了这几种同工酶在沙塘鳢不同组织中的分布以及遗传基础。EST有5个基因位点,单体酶;LDH只有1个基因位点,为四聚体酶;MDH为由3个基因位点控制的二聚体酶;ADH亦为二聚体酶,但仅在肝脏中得到表达。结果表明,同工酶的表达具有组织特异性。  相似文献

为进一步了解内质网蛋白44基因(ERP44)在石斑鱼免疫反应中的作用,本研究根据实验室石斑鱼转录组数据中ERP44的表达序列标签(EST)设计引物,克隆了斜带石斑鱼(Epinephelus coioides)Ec-ERP44基因的开放阅读框序列(ORF),利用生物信息学手段分析Ec-ERP44基因及其编码蛋白的序列结构和特征,并通过实时荧光定量PCR检测该基因的组织分布特征,以及脂多糖(Lipopolysaccharide, LPS)、聚肌胞苷酸(Polyinosinic-polycytidylic acid, Poly I:C)刺激后该基因在石斑鱼脾脏中的表达变化。研究结果表明,Ec-ERP44基因ORF全长1 233 bp,编码410个氨基酸;该蛋白具有蛋白质二硫键异构酶(PDI)家族保守的硫氧还蛋白结构域。同时,Ec-ERP44基因在健康石斑鱼体内的多种组织均有表达,其中在脑组织中的表达量最高;在LPS与Poly I:C刺激后,石斑鱼脾脏中Ec-ERP44基因的表达显著上调。本研究结果表明,Ec-ERP44基因参与了石斑鱼抗病原感染的免疫反应。  相似文献

From the discussion of the features of women's language, more attention should be paid to language use in social context. The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.  相似文献

There are numerous geometric objects stored in the spatial databases. An importance function in a spatial database is that users can browse the geometric objects as a map efficiently. Thus the spatial database should display the geometric objects users concern about swiftly onto the display window. This process includes two operations:retrieve data from database and then draw them onto screen. Accordingly, to improve the efficiency, we should try to reduce time of both retrieving object and displaying them. The former can be achieved with the aid of spatial index such as R-tree, the latter require to simplify the objects. Simplification means that objects are shown with sufficient but not with unnecessary detail which depend on the scale of browse. So the major problem is how to retrieve data at different detail level efficiently. This paper introduces the implementation of a multi-scale index in the spatial database SISP (Spatial Information Shared Platform) which is generalized from R-tree. The difference between the generalization and the R-tree lies on two facets: One is that every node and geometric object in the generalization is assigned with a importance value which denote the importance of them, and every vertex in the objects are assigned with a importance value,too. The importance value can be use to decide which data should be retrieve from disk in a query. The other difference is that geometric objects in the generalization are divided into one or more sub-blocks, and vertexes are total ordered by their importance value. With the help of the generalized R-tree, one can easily retrieve data at different detail levels.Some experiments are performed on real-life data to evaluate the performance of solutions that separately use normal spatial index and multi-scale spatial index. The results show that the solution using multi-scale index in SISP is satisfying.  相似文献

The discovery of the prolific Ordovician Red River reservoirs in 1995 in southeastern Saskatchewan was the catalyst for extensive exploration activity which resulted in the discovery of more than 15 new Red River pools. The best yields of Red River production to date have been from dolomite reservoirs. Understanding the processes of dolomitization is, therefore, crucial for the prediction of the connectivity, spatial distribution and heterogeneity of dolomite reservoirs.The Red River reservoirs in the Midale area consist of 3~4 thin dolomitized zones, with a total thickness of about 20 m, which occur at the top of the Yeoman Formation. Two types of replacement dolomite were recognized in the Red River reservoir: dolomitized burrow infills and dolomitized host matrix. The spatial distribution of dolomite suggests that burrowing organisms played an important role in facilitating the fluid flow in the backfilled sediments. This resulted in penecontemporaneous dolomitization of burrow infills by normal seawater. The dolomite in the host matrix is interpreted as having occurred at shallow burial by evaporitic seawater during precipitation of Lake Almar anhydrite that immediately overlies the Yeoman Formation. However, the low δ18O values of dolomited burrow infills (-5.9‰~ -7.8‰, PDB) and matrix dolomites (-6.6‰~ -8.1‰, avg. -7.4‰ PDB) compared to the estimated values for the late Ordovician marine dolomite could be attributed to modification and alteration of dolomite at higher temperatures during deeper burial, which could also be responsible for its 87Sr/86Sr ratios (0.7084~0.7088) that are higher than suggested for the late Ordovician seawaters (0.7078~0.7080). The trace amounts of saddle dolomite cement in the Red River carbonates are probably related to "cannibalization" of earlier replacement dolomite during the chemical compaction.  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
