利用已构建的基因工程菌株BL21/pET-28a-pilA和BL21/pET-28a-ompC,经IPTG诱导,SDS-PAGE分析分别可见表达的20ku和40.9ku的特异条带,确定蛋白以包涵体的形式存在.Western blot结果表明,表达的Ⅰ型菌毛蛋白(pilA基因编码)和外膜蛋白(ompC基因编码)可与抗体发生特异性结合,说明其具有良好的反应原性.将表达特异蛋白的重组菌株BL21/pET-28a-pilA和BL21/pET-28a-ompC分别制成基因工程包涵体疫苗,免疫小鼠后,具有很好的保护能力.ELISA结果显示,免疫的小鼠体内已产生相应的抗体,效价为1∶400,说明这2种疫苗有很好的免疫原性.表明这2株基因工程菌株有望作为鸡致病性大肠杆菌基因工程疫苗的候选生产菌株.  相似文献

人心肌肌钙蛋白Ⅰ(hcTnI)是临床检测心肌损伤及预后提供诊断的生物学指标,由于来源有限,采用基因重组技术,以期获得高表达量的人心肌肌钙蛋白Ⅰ.人工合成hcTnI基因,将其插入pET-11a载体中,通过酶切鉴定正确后转入表达宿主菌BL21(DE3)中,诱导表达目的蛋白.采用蛋白免疫印迹反应(Western Blot,WB)鉴定表达目的蛋白的免疫特异性.经SDS-PAGE证实重组蛋白的相对分子质量约为2.6×104,凝胶密度扫描软件检测到目的蛋白占总蛋白比例为30.1%,WB实验验证诱导后目的蛋白特异性良好.成功构建了重组hcTnI基因在大肠杆菌表达的工程菌株,并获得表达,为制备高特异性的抗体及临床检测应用和测定标准化奠定了基础.  相似文献

为了制备抗人心肌肌钙蛋白I(cTnI)单克隆抗体、建立检测人cTnI链酶亲和素-生物素双抗体夹心酶联免疫吸附测定(ELISA)方法,用基因工程表达的人cTnI免疫雌性BALB/c小鼠,以杂交瘤技术制备得到三株特异性抗人cTnI的单克隆抗体记为6G3,6A6,3G9.三株杂交瘤细胞中6G3和6A6为IgG1,3G9为IgG2b,腹水效价分别为4×10-5,4×10-5和10-5.亲和常数为5.0×109,6.4×109和3.8×109mol-1.Westernblotting结果表明6G3和6A6能够识别cTnI-C复合物.把6G3抗体生物素化并制备抗cTnI、cTnC和cTnI-C的大鼠多克隆抗体.以多抗作为固相捕捉抗体,生物素化6G3抗体作为检测抗体,建立检测cTnI的双抗体夹心ELISA方法并初步应用于临床病人标本检测.以cTnI-C复合物多抗捕捉检测方法具有良好的灵敏度、特异性和准确性,灵敏度为0.9ng/mL,较包被其他抗体捕捉方法提高1~2倍.批内变异系数为5.03%,回收率为94.56%.测定17例AMI病人和18例血清样本临床诊断结果符合率为100%.  相似文献

在心脏发育过程中,相关基因的正常表达是有功能心脏形成的关键.斑马鱼CFL基因是从斑马鱼胚胎的cDNA文库中克隆出来的一个心脏发育候选基因.生物信息学分析表明:CFL基因编码278个氨基酸,含有一个CXXC结构域.为进一步研究该基因的功能,采用生物信息学结合RT-PCR的方法获得了该基因.将所得片段插入原核表达载体pET-28a载体中,经测序鉴定正确后转化入Rosetta菌株中,用IPTG诱导表达出pET-28a-CFL融合蛋白,表达的融合蛋白占菌体总蛋白的71%,融合蛋白相对分子质量约为30kD.经包涵体纯化后,免疫新西兰大白兔制备了多克隆抗体.经验证,具有较好的特异性和较高的效价,可以用作western-blot免疫印迹等实验分析.  相似文献

人溶菌酶是人体内的一种非特异性免疫物质,具有杀菌消炎、免疫调节、改善胃肠道菌群等作用,在抗生素替代方面具有广阔的应用前景。已报道人溶菌酶基因c DNA在内源的αS2酪蛋白基因启动子调控下的表达量不高(23~31μg/m L),特尝试密码子优化策略以改善表达量。优化了人溶菌酶基因c DNA的密码子,连入以小鼠乳清酸蛋白(m WAP)基因座序列为调控元件的乳腺表达载体,验证其表达效率。通过原核注射制备转基因小鼠,PCR检测发现,出生的21只小鼠中有4只为转基因阳性,且能按孟德尔遗传规律传递给下一代;Western blot检测表明,小鼠乳汁中有微量的重组人溶菌酶表达。因此,密码子优化后的人溶菌酶基因可以在小鼠乳腺中获得有效表达,这可为外源基因定点整合的人溶菌酶转基因动物的高效表达研究奠定基础。  相似文献

为了提高流感病毒血凝素(HA)基因的表达量及其免疫原性,按照哺乳动物偏爱密码子将A/Goose/HLJ/QFY/04(H5N1)流感病毒的血凝素基因进行优化改造,然后插入到真核表达载体pCI—neo中,构建了真核表达质粒pCI—opti—HA.将此质粒和含野生HA基因的真核表达质粒pCI—wt—HA分别转染Vero或293T细胞,通过免疫过氧化物酶单层细胞试验(IPMA)和蛋白免疫印迹(WB)实验比较HA蛋白的表达量.将两种重组质粒免疫Balb/c小鼠,比较两者诱导的抗体产生水平.三次免疫后两周用强毒攻击,通过体温和体重变化评价两种重组质粒的免疫保护效果.IP—MA和WWWB实验结果表明:密码子优化后,HA蛋白在哺乳动物细胞中的表达水平显著提高.酶联免疫吸附实验(ELISA)结果表明:小鼠免疫后密码子优化的重组质粒诱导的特异性抗体效价较高.攻毒试验结果表明:两种重组质粒均能够提供较好的保护.  相似文献

文章构建小鼠肥大细胞蛋白酶4(mMCP-4)原核表达体系,获得原核表达蛋白,并制备兔抗mMCP-4多克隆抗体,提取小鼠脾细胞总 RNA ,运用 RT-PCR技术获得目的基因 mMCP-4,构建重组原核表达载体pET28a-mMCP-4,将其转化到大肠杆菌BL21中诱导表达。mMCP-4融合蛋白经Ni亲和层析法纯化后,作为抗原免疫兔子获得mMCP-4多克隆抗体。采用ELISA法测抗体效价,western blot检测抗体特异性。结果表明,构建重组表达质粒pET28a-mMCP-4,经大规模原核表达获得大量mMCP-4融合蛋白,制备的兔抗血清效价达1∶128000,并表现出较好特异性。研究结果为进一步研究mMCP-4生物学功能奠定了基础。  相似文献

构建外毒素结构域Ⅰ a基因(EPA103)与Ⅱ型单纯疱疹病毒糖蛋白(HSV-2-gD)嵌合蛋白表达载体(pET-EgD),得到嵌合表达蛋白.利用外毒素结构域Ⅰ a基因表达蛋白的佐剂效应,以pET-EgD表达嵌合蛋白免疫Balb/c小鼠.结果显示:嵌合表达蛋白(pET-EgD)免疫小鼠,可在小鼠体内诱导产生抗HSV-2特异性抗体.嵌合表达蛋白(pET-EgD)具有较强的免疫原性,为开发研究Ⅱ型单纯疱疹核酸疫苗奠定了基础.  相似文献

将鸡白细胞介素-2(ChIL-2)基因插入酵母分泌型表达载体pPIC9K 构建重组表达载体pPIC9K/ChIL-2,通过电转化构建毕赤酵母(Pichia methanolica)GS115 甲醇利用型重组表达菌株.经SDS-PAGE与Western blot分析,证实ChIL-2基因在重组GS115中成功表达出约16 ku与14 ku两条特异性条带,这与天然ChIL-2相对分子质量大小一致,提示ChIL-2可能为糖基化蛋白质.  相似文献

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. There are numerous geometric objects stored in the spatial databases. An importance function in a spatial database is that users can browse the geometric objects as a map efficiently. Thus the spatial database should display the geometric objects users concern about swiftly onto the display window. This process includes two operations:retrieve data from database and then draw them onto screen. Accordingly, to improve the efficiency, we should try to reduce time of both retrieving object and displaying them. The former can be achieved with the aid of spatial index such as R-tree, the latter require to simplify the objects. Simplification means that objects are shown with sufficient but not with unnecessary detail which depend on the scale of browse. So the major problem is how to retrieve data at different detail level efficiently. This paper introduces the implementation of a multi-scale index in the spatial database SISP (Spatial Information Shared Platform) which is generalized from R-tree. The difference between the generalization and the R-tree lies on two facets: One is that every node and geometric object in the generalization is assigned with a importance value which denote the importance of them, and every vertex in the objects are assigned with a importance value,too. The importance value can be use to decide which data should be retrieve from disk in a query. The other difference is that geometric objects in the generalization are divided into one or more sub-blocks, and vertexes are total ordered by their importance value. With the help of the generalized R-tree, one can easily retrieve data at different detail levels.Some experiments are performed on real-life data to evaluate the performance of solutions that separately use normal spatial index and multi-scale spatial index. The results show that the solution using multi-scale index in SISP is satisfying.  相似文献

The discovery of the prolific Ordovician Red River reservoirs in 1995 in southeastern Saskatchewan was the catalyst for extensive exploration activity which resulted in the discovery of more than 15 new Red River pools. The best yields of Red River production to date have been from dolomite reservoirs. Understanding the processes of dolomitization is, therefore, crucial for the prediction of the connectivity, spatial distribution and heterogeneity of dolomite reservoirs.The Red River reservoirs in the Midale area consist of 3~4 thin dolomitized zones, with a total thickness of about 20 m, which occur at the top of the Yeoman Formation. Two types of replacement dolomite were recognized in the Red River reservoir: dolomitized burrow infills and dolomitized host matrix. The spatial distribution of dolomite suggests that burrowing organisms played an important role in facilitating the fluid flow in the backfilled sediments. This resulted in penecontemporaneous dolomitization of burrow infills by normal seawater. The dolomite in the host matrix is interpreted as having occurred at shallow burial by evaporitic seawater during precipitation of Lake Almar anhydrite that immediately overlies the Yeoman Formation. However, the low δ18O values of dolomited burrow infills (-5.9‰~ -7.8‰, PDB) and matrix dolomites (-6.6‰~ -8.1‰, avg. -7.4‰ PDB) compared to the estimated values for the late Ordovician marine dolomite could be attributed to modification and alteration of dolomite at higher temperatures during deeper burial, which could also be responsible for its 87Sr/86Sr ratios (0.7084~0.7088) that are higher than suggested for the late Ordovician seawaters (0.7078~0.7080). The trace amounts of saddle dolomite cement in the Red River carbonates are probably related to "cannibalization" of earlier replacement dolomite during the chemical compaction.  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献

The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.  相似文献
