参照毕赤巴斯德酵母(Pichia pastoris)的偏好密码子,改造克隆杂合抗菌肽CA(1~7)M(4~11)和CB(1~7)M(4~11)基因,将改造后的基因克隆到pPICZα-A 载体中,构建分泌型表达载体pPICZα-A-CAM和pPICZα-A-CBM,转化宿主菌Pichia pastoris X-33,在甲醇的诱导下进行表达.实验结果表明,杂合抗菌肽获得表达,两种表达产物具有明显的抗菌活性.此外,本文还比较了二者的抗菌谱,分析了杂合肽的性质,优化了酵母电转化条件.  相似文献

黑鲷抗菌肽hepcidin在毕赤酵母中的表达及其抗菌活性   总被引:2,自引:0,他引:2  
鱼类抗菌肽hepcidin具有广谱抗菌活性.利用PCR技术扩增获得了黑鲷抗菌肽hepcidin (AS-hepc2)的前体肽和成熟肽 cDNA 序列,片段大小约250 bp.将其与毕赤酵母(Pichia pastoris) pPIC9K 质粒连接,构建了分泌表达载体pPIC9K/AS-hepc2 .电击法转化重组表达载体,经过G418筛选和选择性培养基以及PCR鉴定,得到的克隆子都为Mut+.以甲醇诱导pPIC9K/AS-hepc2,分泌表达上清中获得10 ku左右的表达产物,与预期的目的蛋白黑鲷hepcidin的前体肽和成熟肽的大小相符.表达试验证明,随时间的延长,表达产物量增多,至120 h 达到高峰.表达产物具有很强的热稳定性.用抑菌圈法测定重组蛋白Pro-AS-hepc2 的抗菌活性,发现能抑制革兰氏阳性菌和革兰氏阴性菌的生长.  相似文献

采用PCR技术改造抗菌肽AD基因,将其C末端改造为Asn编码,改造后的抗菌肽Cecropin AD基因克隆到pPICZα-A载体上,构建成酵母重组分泌型表达载体,转化毕赤酵母(Pichia pastoris)受体菌GS115中,采用zerocin抗性标记筛选重组转化子,经摇瓶发酵,浓缩发酵液进行酸性聚丙烯酰胺凝胶电泳分析和抑菌活性检测。结果表明,改造后的Cecropin AD基因在毕赤酵母中获得了表达,表达产物经α-Factor信号肽引导分泌到胞外,具有较强的杀菌活性。  相似文献

利用逆转录聚合酶链式反应(RT-PCR)方法扩增得到肠道病毒71型(EV71SHZH03)外壳蛋白VP1基因,经序列测定证实后,构建重组表达质粒VP1/pPIC9K,转化Pichia pastoris 酵母宿主菌Gs115,以Myc-Tag多克隆抗体作为一抗,利用双层滤膜法筛选酵母转化子.甲醇诱导表达.SDS-PAGE分析显示:表达产物的分子量约为30000,与天然VP1大小一致,ELISA实验表明,表达上清液可与EV71患者急性感染期血清呈阳性反应,表明重组蛋白VP1具有免疫原性.  相似文献

为了构建人防御素HNP4重组毕赤酵母(Pichia pastoris)表达载体pPIC9K-HNP4,建立高效、稳定表达人防御素HNP4的毕赤酵母表达菌株,并对表达产物进行检测与分析.利用PCR技术从pUC18-HNP4上扩增人防御素HNP4,构建人防御素HNP4重组表达载体pPIC9K-HNP4.SacI线性化后电击法转化入毕赤酵母GS115中,再通过同源重组到酵母染色体上,用G418筛选高表达菌株,在三角摇瓶中0.5%甲醇诱导表达.SDS-PAGE和Western blotting检测表达产物,用标准大肠杆菌、金黄色葡萄球菌菌株混菌板鉴定活性.结果显示,SDS-PAGE和Western-blotting可检测到大小4.0 kDa左右的目的蛋白条带,表达量可达到500 mg/L,表达的重组人防御素HNP4具有较强的杀菌或抑菌活性.提示成功构建人防御素HNP4重组毕赤酵母表达载体pPIC9K-HNP4,并建立高效、稳定表达有明显抗菌活性的重组人防御素HNP4的毕赤酵母表达菌株.  相似文献

Ag85是结核分枝杆菌的一种分枝菌酸转移酶复合体,在结核分枝杆菌的细胞壁合成过程中起着重要作用Ag85B是该复合体的一个组分,具有较好的免疫原性通过连接多肽(G4S)4的编码序列,将α2b干扰素基因与Ag85B基因连成融合基因,然后克隆到毕赤氏酵母表达载体pPIC9上,转化Pichia pastoris SMD1168后获得重组酵母工程菌SMD1168/pPIC9-α2bAg85B,经甲醇诱导培养,发酵上清液经抗病毒活性测定,Western blot鉴定以及初步纯化,结果表明,分泌表达的融合蛋白具有一定的抗病毒活性.  相似文献

Hydramacin-1是来源于大乳头水螅(Hydra)的上皮防御组织的具有广谱抗菌特性的新型抗菌肽.本研究构建了外泌型重组酵母表达载体pPICZαA-Hydramacin-1,再将线性化的重组表达载体pPICZαA-Hydramacin-1电转入毕赤酵母宿主菌(Pichiapastoris)GS115中,成功构建了重组毕赤酵母表达菌株.在培养温度为28℃、甲醇体积分数为1.5%、诱导表达时长为120 h时,蛋白表达量达到最高,重组抗菌肽Hydramacin-1的表达量可达83 mg/L.经阳离子交换柱纯化,每升发酵液可得到约9 mg纯度为90%的重组抗菌肽Hydramacin-1.经最小抑菌浓度的测定,制备的重组抗菌肽具有广谱抗菌特性,相比于革兰氏阳性菌,革兰氏阴性菌对此抗菌肽更为敏感.本实验将Hydramacin-1抗菌肽首次成功地在毕赤酵母中高效表达,为以后该抗菌肽的理论研究和应用奠定了基础.  相似文献

通过PCR扩增得到组织型纤溶酶原激活剂突变体Reteplase(rPA)基因片断,将该基因片断插入到含有AOX1启动子和α分泌信号肽序列载体pPIC9K中,构建了重组表达质粒pPICgK/rPA,转化Pichia pastoris GS115宿主菌.通过比较转化子在MM和MD平板上的生长状况,筛选His^ Mut^s表型转化子.并在2.0mg/mL G418平板筛选得到多拷贝转化子GR10,GR11.摇瓶培养,甲醇诱导外源基因表达,表达产物经SDS-PAGE和Western blot分析,结果表明重组蛋白分子量约39ku,能与特异性单克隆抗体发生免疫反应;体外气泡法测定rPA活性,结果显示重组蛋白具有较好的纤溶活性.通过正交试验,优化发酵条件,表达活性最高为1050IU/mL.  相似文献

为提高南极假丝酵母脂肪酶B(Candida antarcticalipase B,CALB)的表达量,根据毕赤酵母(Pichia pastoris)密码子偏好性设计合成CALB基因,插入表达载体pPIC9K构建重组质粒pPIC9K-CALB.重组质粒转化毕赤酵母GS115,经过G418抗性筛选得到多拷贝转化子.摇瓶发酵120 h后上清液酶活力达到46 U/mL,通过金属螯合层析纯化发酵液将CALB纯化了5.23倍,比活力达到856.7 U/mg,去糖基化实验显示重组CALB比野生型CALB大5 ku.实验考察了不同反应温度、pH值和金属离子对重组CALB活性和稳定性的影响,发现重组CALB最适反应温度和pH分别为30℃和6.5,在pH 5.0~8.0之间以及40℃以下有较好稳定性,金属离子(10 mmol/L)Ca2+,Zn2+,Mn2+有助于CALB酶活力的提高,而Cu2+,Ag+,Fe3+强烈抑制酶催化反应.  相似文献

The discovery of the prolific Ordovician Red River reservoirs in 1995 in southeastern Saskatchewan was the catalyst for extensive exploration activity which resulted in the discovery of more than 15 new Red River pools. The best yields of Red River production to date have been from dolomite reservoirs. Understanding the processes of dolomitization is, therefore, crucial for the prediction of the connectivity, spatial distribution and heterogeneity of dolomite reservoirs.The Red River reservoirs in the Midale area consist of 3~4 thin dolomitized zones, with a total thickness of about 20 m, which occur at the top of the Yeoman Formation. Two types of replacement dolomite were recognized in the Red River reservoir: dolomitized burrow infills and dolomitized host matrix. The spatial distribution of dolomite suggests that burrowing organisms played an important role in facilitating the fluid flow in the backfilled sediments. This resulted in penecontemporaneous dolomitization of burrow infills by normal seawater. The dolomite in the host matrix is interpreted as having occurred at shallow burial by evaporitic seawater during precipitation of Lake Almar anhydrite that immediately overlies the Yeoman Formation. However, the low δ18O values of dolomited burrow infills (-5.9‰~ -7.8‰, PDB) and matrix dolomites (-6.6‰~ -8.1‰, avg. -7.4‰ PDB) compared to the estimated values for the late Ordovician marine dolomite could be attributed to modification and alteration of dolomite at higher temperatures during deeper burial, which could also be responsible for its 87Sr/86Sr ratios (0.7084~0.7088) that are higher than suggested for the late Ordovician seawaters (0.7078~0.7080). The trace amounts of saddle dolomite cement in the Red River carbonates are probably related to "cannibalization" of earlier replacement dolomite during the chemical compaction.  相似文献

本文叙述了对海南岛及其毗邻大陆边缘白垩纪到第四纪地层岩石进行古地磁研究的全部工作过程。通过分析岩石中剩余磁矢量的磁偏角及磁倾角的变化,提出海南岛白垩纪以来经历的构造演化模式如下:早期伴随顺时针旋转而向南迁移,后期伴随逆时针转动并向北运移。联系该地区及邻区的地质、地球物理资料,对海南岛上述的构造地体运动提出以下认识:北部湾内早期有一拉张作用,主要是该作用使湾内地壳显著伸长减薄,形成北部湾盆地。从而导致了海南岛的早期构造运动,而海南岛后期的构造运动则主要是受南海海底扩张的影响。海南地体运动规律的阐明对于了解北部湾油气盆地的形成演化有重要的理论和实际意义。  相似文献

The explosive growth of the Internet and database applications has driven database to be more scalable and available, and able to support on-line scaling without interrupting service. To support more client's queries without downtime and degrading the response time, more nodes have to be scaled up while the database is running. This paper presents the overview of scalable and available database that satisfies the above characteristics. And we propose a novel on-line scaling method. Our method improves the existing on-line scaling method for fast response time and higher throughputs. Our proposed method reduces unnecessary network use, i.e. , we decrease the number of data copy by reusing the backup data. Also, our on-line scaling operation can be processed parallel by selecting adequate nodes as new node. Our performance study shows that our method results in significant reduction in data copy time.  相似文献

R-Tree is a good structure for spatial searching. But in this indexing structure,either the sequence of nodes in the same level or sequence of traveling these nodes when queries are made is random. Since the possibility that the object appears in different MBR which have the same parents node is different, if we make the subnode who has the most possibility be traveled first, the time cost will be decreased in most of the cases. In some case, the possibility of a point belong to a rectangle will shows direct proportion with the size of the rectangle. But this conclusion is based on an assumption that the objects are symmetrically distributing in the area and this assumption is not always coming into existence. Now we found a more direct parameter to scale the possibility and made a little change on the structure of R-tree, to increase the possibility of founding the satisfying answer in the front sub trees. The geographic information service is enabled by the advancements in general Web service technology and the focused efforts of the OGC in defining XML-based Web GIS service. Based on these models, this paper addresses the issue of services chaining,the process of combining or pipelining results from several interoperable GIS Web Services to create a customized solution. This paper presents a mediated chaining architecture in which a specific service takes responsibility for performing the process that describes a service chain. We designed the Spatial Information Process Language (SIPL) for dynamic modeling and describing the service chain, also a prototype of the Spatial Information Process Execution Engine (SIPEE) is implemented for executing processes written in SIPL. Discussion of measures to improve the functionality and performance of such system will be included.  相似文献

Advances in wireless technologies and positioning technologies and spread of wireless devices, an interest in LBS (Location Based Service) is arising. To provide location based service, tracking data should have been stored in moving object database management system (called MODBMS) with proper policies and managed efficiently. So the methods which acquire the location information at regular time intervals then, store and manage have been studied. In this paper, we suggest tracking data management techniques using topology that is corresponding to the moving path of moving object. In our techniques, we update the MODBMS when moving object arrived at a street intersection or a curved road which is represented as the node in topology and predict the location at past and future with attribute of topology and linear function. In this technique, location data that are corresponding to the node in topology are stored, thus reduce the number of update and amount of data. Also in case predicting the location,because topology are used as well as existing location information, accuracy for prediction is increased than applying linear function or spline function.  相似文献
