犬静脉给予重组人组织型纤溶酶原激活剂缺失变体(K2tPA)1×106IU/kg、5×105IU/kg、2.5×105IU/kg对冠脉血栓产生显著的溶栓效果,栓塞冠脉血管很快出现再通,高、中、低剂量组残存血栓较溶剂对照组分别减少了72.7%、55.6%、42.5%;心肌梗死范围明显缩小.血纤维蛋白原明显降解,凝血酶原时间、凝血酶时间及陶土激活部分凝血酶时间明显延长,但抗凝血酶活性无显著改变.伤口出血量增加,出血时间延长.以上指标与等剂量(5×105IU/kg)的德国BoehringerMannheimGmbH产品Retavase作用相似.离体家兔血小板凝集实验结果表明:重组人K2tPA对家兔血小板聚集功能具有明显的抑制作用.  相似文献

重组人尿激酶原急性毒性和长期毒性的研究   总被引:1,自引:1,他引:0  
(1)重组人尿激酶原(prouk)单剂量分别经静脉和腹腔注射给药均未侧出小鼠的LD50,因此改作其最大给药量。结果表明,prouk单剂量经静脉及腹腔注射给药小鼠的最大给药量不低于9,000万IU/kg。(2) prouk 36.0万IU/kg、67.5万IU/kg、135.0万IU/kg和270.0万IU/kg四个剂量组连续给食蟹猴静脉滴注14d,每组4只猴,雌雄各半。实验过程中未见动物死亡。prouk36.0万IU/kg剂量组的食蟹猴给药期及恢复期,精神状态良好,一般行为活动正常,体重、分泌物、食欲及摄食量、心电图、血常规、血液生化、体表观察、尿常规和粪便隐血试验等基本正常,无明显变化。给药第14天和恢复期的脏器系数与病理组织学检查表明,动物的脏器、组织均未见由药物引起的异常病理改变,在生理盐水组或赋形剂组之间比较均无明显差异。而prouk67.5万IU/kg、135.0万IU/kg、270.0万IU/kg剂量组的食蟹猴出现皮下出血、充血、紫癜、尿、粪便隐血反应阳性、贫血以及心率加快等症状。给药期及恢复期,血清中检测出少量prouk抗体。结果表明,prouk的无毒安全剂量不低于36.0万IU/kg。  相似文献

试验结果显示 :尿激酶原 (prouk) 2 2 .5万IU/kg、4 5 .0万IU/kg和 90 .0万IU/kg3个组、赋形剂组和生理盐水组动物尾静脉注射给药 ,对小鼠一般行为活动表现和自发活动次数均无明显影响 .prouk 2 2 .5万IU/kg、4 5 .0万IU/kg和 90 .0万IU/kg三个组、赋形剂组和生理盐水组动物尾静脉注射给药 ,对大鼠一般行为活动表现均无明显影响 ,均能在旋转棒上协调平衡运动 .此外 ,给药后 2 0min ,三个剂量组大鼠凝血时间虽有延长 ,但与生理盐水组或赋形剂组比较无显著性差异 (P >0 .0 5 ) .prouk 11.2 5万IU/kg、2 2 .5 0万IU /kg、4 5 .0 0万IU/kg三组静脉滴注给药 ,麻醉犬的ECG、HR、呼吸均无明显改变 ,与赋形剂组比较无显著性差异 .但prouk高剂量组动物的SAP、DAP、MAP在给药 10~ 15min时短暂下降 ,这些反应在给药 30min后均恢复正常 .结果表明 ,prouk在受试剂量范围内对小鼠、大鼠精神神经活动和大鼠的凝血时间等均无明显影响 ,在 4 5 .0 0万IU/kg剂量时对麻醉犬有短暂的降低血压的作用  相似文献

第一部分 :采用交叉设计 ,自身比较 ,研究了猕猴静脉注射 0 .4 ,0 .8和 3.2mg·kg- 1 重组人尿激酶原 (rhprouk)或 1.2 8× 10 5IU·kg- 1 尿源性天然尿激酶 (un UK) (其中推注 15 %剂量 ,90min内恒速滴注 85 %剂量 )后血浆中单链尿激酶型纤溶酶原激活剂 (scu PA)和双链尿激酶型纤溶酶原激活剂 (tcu PA)的浓度随时间变化、代谢转化和各自的药代动力学 ,及代谢转化和药代动力学参数与剂量浓度的依赖关系 .结果表明给药后scu PA转化为tcu PA的速度与scu PA的剂量呈正相关性 .第二部分 :通过测定兔静脉推注加滴注1 2 5I rhprouk 0 .8mg·kg- 1 (其中 15 %剂量静脉推注 ,85 %剂量在 90min内滴注 ,比放射活性 2 .5 0 6MBq/mg)后不同时间体内各组织总放射性、TCA酸沉放射性 ,以及酸沉和总放射性的比值 ;胆汁引流物放射性累计排出量 ,尿和粪中累计排出量 ,及在体内外1 2 5I rhprouk与血浆蛋白的结合 .研究rhprouk在体内的分布、代谢及排泄情况 .结果表明rhprouk不与血浆中蛋白结合 ,代谢部位主要集中在胆、肾等部位 ,代谢产物主要随小便排出 .  相似文献

将含有 pET2 9a prouk重组质粒的人尿激酶原工程菌经 10L种子罐培养及 10 0L发酵 ,IPTG诱导表达 ,其表达量为占菌体总蛋白的 2 0 % ,表达产物经体外变复性 ,CM 纤维素离子交换层析 ,Superdex 75分子筛层析及Affi preppolymyxinsupport亲和层析去热源 ,每 10 0L发酵液得重组人尿激酶原纯品 6g ,纯度达 95 %以上 ,比活大于 1.2× 10 4 IU /mg ,双链含量低于0 .5 % ,内毒素及热源含量、宿主蛋白残留量、宿主DNA残留量等均达到临床使用标准 .其分子量 (质谱测定 )、氨基酸组成、N、C 端氨基酸序列分析等均与理论值相符 .进行了等电点及肽图等性质研究  相似文献

目的:分析急性脑梗死溶栓治疗后脑出血的相关影响因素.方法:53例急性脑梗死患者接受尿激酶(rt-PA)溶栓治疗,溶栓后24h复查头颅CT,观察溶栓后颅内出血情况,并对相关影响因素进行多元Logistic回归分析.结果:溶栓后各种出血12例(22.6%),脑出血10例(18.8%),症状性颅内出血4例(7.5%),多因素Logistic回归表明影响出血的因素为心房纤颤、糖尿病、尿激酶剂量与继发感染.结论:脑梗死溶栓治疗后脑出血的影响因素可能为心房纤颤、糖尿病、尿激酶剂量与继发感染.  相似文献

我们对48例AMI的病人进行溶栓治疗,用国产尿激酶(UK)2.2万IU/kg加入100ml葡萄糖液内静点,12h后皮下注射肝素7500u,每12h一次,用3~5日,滴尿激酶前口服肠溶阿斯匹林0.3,以后0.3/日,3~5日后改为50~150mg/日维持,至出院。对其疗效评价,结果显示,发病至溶栓时间6h内者再通率明显高于6h以上者(P<0.01)尿激酶剂量大,溶栓的效果好,不良反应轻,便于不能开展经皮冠脉内直接溶栓的基层医院的临床应用。  相似文献

利用在体和离体实验相结合 ,以动物接种肿瘤细胞后的生存时间、瘤重与体重比值、肝癌细胞生存率、超氧化物歧化酶 (SOD)活性及肝癌细胞凋亡为指标 ,研究了不同剂量榄仁树叶提取物 (LR 98)对肝癌细胞增殖的抑制性效应及可能机制 .在体实验结果表明 ,5× 10 - 3g/mLLR 98处理组动物的生存时间较对照组明显延长 (P <0 .0 1) ,不同剂量处理组 (5× 10 - 3g/mL ,2× 10 - 2 g/mL ,5× 10 - 2 g/mL)的瘤重与体重比值均较对照组明显减小 (P <0 .0 5) .体外培养的细胞经不同剂量LR 98(1× 10 - 5g/mL ,5× 10 - 5g/mL ,1× 10 - 4 g/mL)作用后 ,生存率及SOD活性均显著下降 (P <0 .0 1) .流式细胞仪分析表明 ,不同剂量LR 98作用于肝癌细胞 8h后 ,在G1 期前均出现一亚二倍体峰 ,表明均能诱导肝癌细胞发生凋亡 ,凋亡率分别达 2 9.4 4 % ,2 4 .4 5%和 2 5.2 2 % .以上结果说明LR 98可显著抑制肝癌细胞增殖 .降低SOD活性 ,诱导肝癌细胞凋亡可能是LR 98抑制肝癌细胞生长的机理之一 .  相似文献

目的:探讨血栓闭塞性脉管炎的表现及动脉内接触性溶栓治疗的临床疗效观察.方法:4例患者经健侧股动脉行seldinger敲术,从导管内缓慢注入罂粟碱、复方丹参注射液、尿激酶,然后分析DSAE图像及观察下肢缺血症状.结果:血栓闭塞性脉管炎主要病损累及中、小型动脉,DSA表现为闭塞端呈"怀口"状,狭窄段血管壁毛糙、不规则、粗细不均,分布失常.动脉内接触性溶栓治疗对疼痛及灼热感均有一定缓解,下肢缺血症状有一定减轻,但对已有坏疽者,难以起到挽救肢体作用.结论:发病时间较短的血栓闭塞性脉管炎及时进行接触性溶栓治疗是一种新的有效的治疗方法.  相似文献

The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.  相似文献

There are numerous geometric objects stored in the spatial databases. An importance function in a spatial database is that users can browse the geometric objects as a map efficiently. Thus the spatial database should display the geometric objects users concern about swiftly onto the display window. This process includes two operations:retrieve data from database and then draw them onto screen. Accordingly, to improve the efficiency, we should try to reduce time of both retrieving object and displaying them. The former can be achieved with the aid of spatial index such as R-tree, the latter require to simplify the objects. Simplification means that objects are shown with sufficient but not with unnecessary detail which depend on the scale of browse. So the major problem is how to retrieve data at different detail level efficiently. This paper introduces the implementation of a multi-scale index in the spatial database SISP (Spatial Information Shared Platform) which is generalized from R-tree. The difference between the generalization and the R-tree lies on two facets: One is that every node and geometric object in the generalization is assigned with a importance value which denote the importance of them, and every vertex in the objects are assigned with a importance value,too. The importance value can be use to decide which data should be retrieve from disk in a query. The other difference is that geometric objects in the generalization are divided into one or more sub-blocks, and vertexes are total ordered by their importance value. With the help of the generalized R-tree, one can easily retrieve data at different detail levels.Some experiments are performed on real-life data to evaluate the performance of solutions that separately use normal spatial index and multi-scale spatial index. The results show that the solution using multi-scale index in SISP is satisfying.  相似文献

The discovery of the prolific Ordovician Red River reservoirs in 1995 in southeastern Saskatchewan was the catalyst for extensive exploration activity which resulted in the discovery of more than 15 new Red River pools. The best yields of Red River production to date have been from dolomite reservoirs. Understanding the processes of dolomitization is, therefore, crucial for the prediction of the connectivity, spatial distribution and heterogeneity of dolomite reservoirs.The Red River reservoirs in the Midale area consist of 3~4 thin dolomitized zones, with a total thickness of about 20 m, which occur at the top of the Yeoman Formation. Two types of replacement dolomite were recognized in the Red River reservoir: dolomitized burrow infills and dolomitized host matrix. The spatial distribution of dolomite suggests that burrowing organisms played an important role in facilitating the fluid flow in the backfilled sediments. This resulted in penecontemporaneous dolomitization of burrow infills by normal seawater. The dolomite in the host matrix is interpreted as having occurred at shallow burial by evaporitic seawater during precipitation of Lake Almar anhydrite that immediately overlies the Yeoman Formation. However, the low δ18O values of dolomited burrow infills (-5.9‰~ -7.8‰, PDB) and matrix dolomites (-6.6‰~ -8.1‰, avg. -7.4‰ PDB) compared to the estimated values for the late Ordovician marine dolomite could be attributed to modification and alteration of dolomite at higher temperatures during deeper burial, which could also be responsible for its 87Sr/86Sr ratios (0.7084~0.7088) that are higher than suggested for the late Ordovician seawaters (0.7078~0.7080). The trace amounts of saddle dolomite cement in the Red River carbonates are probably related to "cannibalization" of earlier replacement dolomite during the chemical compaction.  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
